Bu tez çalışmasında, koç testislerindeki mast hücre dağılımı ve yoğunluğunun farklı fiksatifler kullanılarak belirlenmesi amaçlanmıştır. Çalışmada altı adet yetişkin koçtan alınan testis kapsülü, epididimis, ductus deferens ve ductuli efferentes örnekleri materyal olarak kullanıldı. Fiksatif olarak, standart fiksatiflerden Bouin, % 10 Formaline, Schaudin, Orth, Alkolik Formalin ve Carnoy, boya olarak ise standart boyalardan Toluidin Blue (TB), Thionin, Alcian Blue (AB), Safranin (AB/SO) kullanıldı. Uygulanan tüm fiksatiflerde ve çalışılan bütün bölgelerden alınan kesitlere Thionin ve toluidine Blue uygulamaları sonucunda reaksiyona rastlandı. Alcian Blue, Safranin(+) ve Alcian Blue/Safranin(+) boyamaları sonucunda ise bölgelere göre anlamlı reaksiyon farklılıkları tespit edildi. Alkolik Formalin fiksasyonu sonucunda, ductus deferens ile ductuli efferentes bölgelerine AB/SO(+) boyası uygulaması sonucunda mukozal ve bağ dokularında bulunan mast hücrelerinde anlamli AB(+) reaksiyon gözlendi. Bouin fiksasyonunda ise ductus deferens bölgesi dışında ayni hücrelerde de benzer sonuçlar elde edildi. Ductus deferens bölgesinde, AB/SO(+) uygulaması ve Orth fiksasyonu sonucunda mukozal ve bağ dokularında bulunan mast hücreleri, Schaudin fiksatifte ise sadece mukozal mast hücrelerinin dışında SO(+) reaksiyon tespit edildi. Ductuli efferentes bölgesinde; %10 Formalin fiksasyonu, mukozal ve bağ dokularında lokalize olan mast hücrelerinde SO(+) reaksiyona rastlanmadı. Carnoy ve Schaudin fiksasyonlarinda ise bağ dokularında bulunan mast hücrelerinde reaksiyon gözlenmedi. Belirtilen bölgeler dışında ise değişen yoğunluklarda AB(+),SO(+) ve AB/SO(+) reaksiyon gösteren mast hücreleri saptandı. Sonuç olarak farklı fiksatiflerin ayni boya ve bölgede verdiği sonuçlarda anlamlı farklılıklar tespit edilmiştir. Anahtar Kelimeler: Koç, Testisler, Mast hücre, Heterojenite, Genital Sistem
In this thesis study, determination of distribution and density of mast cells in the testes of the ram by the use of different fixatives was aimed. Regions of testicular capsule, epididimis, ductus deferens and ductuli efferentes taken from six adult rams were used as materials. Bouin, 10 % Formaline, Schaudin, Orth, Alcoholic Formaline and Carnoy from standart fixatives and Toluidin Blue (TB), Thionin, Alcian Blue (AB), Safranin (AB/SO) from standart stains were used. For all the fixatives applied, in all sections there were positive reactions observed for Thionin and Toluidine Blue. There were significant differences determined according to regions by Alcian Blue, Safranin (+) and Alcian Blue/Safranin (+) staining. As the result of Alcoholic Formalin fixation with AB/SO (+) staining, significant AB (+) reaction was found in mucosal and connective tissue mast cells localized in ductus deferens and ductuli efferentes, as in Bouin fixation; similar results were obtained from same type of cells, except ductus deferens. In ductus deferens, with Orth fixation and AB/SO (+) staining, SO (+) reaction was observed in mucosal and connective tissue mast cells, also in Schaudin fixation, there was SO (+) reaction but only in connective tissue mast cells. In ductuli efferentes, with 10 % Formaline fixation, in mast cells localized in mucosal and connective tissue, there were no reactions observed. With Carnoy and Schaudin fixatives, in connective tissue mast cells there were no reactions determined. Mast cells reacts with AB (+), SO (+) and AB/SO (+) at varying densities were found except the indicated regions.
