Bu tez çalışmasında, ultrafiltrasyon (UF) yöntemi ile üretilmiş Beyaz peynirde meydana gelen acılaşma problemlerinin nedenleri araştırılmıştır. Acılaşmanın kullanılan enzimden mi, yoksa başlatıcı kültürden mi olduğunu belirlemek için 2 farklı pıhtılaştırıcı enzim ve 2 farklı başlatıcı kültür kullanılarak UF pastörize ve UF çiğ sütle Beyaz peynir üretimi gerçekleştirilmiştir. Yapılan peynir çalışma modeli aşağıda verilmiştir. 1- A peynir örneği: Kimozin enzimi ile proteolitik aktivitesi yüksek başlatıcı kültür kullanılarak ultrafiltre pastörize sütten Beyaz peynir üretimi, 2- B peynir örneği: Mikrobiyal enzim ile proteolitik aktivitesi yüksek başaltıcı kültür kullanılarak ultrafiltre pastörize sütten Beyaz peynir üretimi, 3- C peynir örneği: Kimozin enzimi ile proteolitik aktivitesi düşük başlatıcı kültür kullanılarak ultrafiltre pastörize sütten Beyaz peynir üretimi, 4- D peynir örneği: Mikrobiyal enzim ile proteolitik aktivitesi düşük başaltıcı kültür kullanılarak ultrafiltre pastörize sütten Beyaz peynir üretimi, 5- E peynir örneği: Kimozin enzimi ile ultrafiltre çiğ sütten Beyaz peynir üretimi, 6- F peynir örneği: Mikrobiyal enzim ile ultrafiltre çiğ sütten Beyaz peynir üretimi yapılmıştır. Depolama sıcaklığının etkisini belirlemek amacıyla peynirler 4°C ve 8 °C' de depolanmıştır. Olgunlaşmanın 1., 30., 60., 90. ve 120. günlerinde fizikokimyasal, kimyasal, mikrobiyolojik ve duyusal analizleri yapılmıştır. UF Beyaz peynirlerin mineral madde analizleri (Cu, Zn, Cr, P, Mg, Ca, Fe, Ka, Na) ICP-OES ile belirlenmiştir. Peynirlerin proteoliz düzeyleri SDS-PAGE elektroforez ve ters faz yüksek performanslı likit kromotografi (RP-HPLC) ile incelenmiştir. Üretim farklılıklarının kurumadde, yağ, tuz, pH, titrasyon asitliği ve kül değerlerinde fazla bir değişikliğe neden olmadığı, suda çözünen azot, %12 trikloroasetik asitte çözünen azot (TCA) ve % 5 fosfotungustik asitte çözünen azot (PTA) ve asit sayısı değerinde artışa neden olduğu protein oranında ise düşüşe neden olduğu tespit edilmiştir. UF pastörize sütle ve UF çiğ sütle yapılan peynir örnekleri arasındaki farklılık mikrobiyolojik açıdan önemli bulunmuştur (p<0.05). UF pastörize sütle yapılan peynir örneklerinde toplam mezofilik aerob bakteri, psikrofilik bakteri ve maya-küf sayıları arasında önemli bir farklılık bulunmamıştır (p>0.05). Olgunlaşma sürecinde peynir örneklerinde proteolize bağlı olarak acılaşmalar tespit edilmiştir. Kimozin enzimi ve proteolitik aktivitesi yüksek başlatıcı kültürle üretilen A peynir örneği duyusal olarak en fazla acılaşmanın algılandığı peynirdir. 30. ve 90. gün peynirlerin SDS-PAGE elektroforez analiz sonuçlarına göre A peynir örneğinde %72,26 oranında αs-kazeinde parçalanma meydana gelmiş, HPLC sonuçlarına göre 40-60 dakika arasında hidrofobik karakterdeki peptidlerin alanında %106'lık artış meydana gelerek yoğun bir hidrofobik peptid oluşumu görülmüştür. In this thesis, the reasons of the bitterness in white cheese produced by ultrafiltration method were investigated. In order to determine whether the bitterness is from the enzyme or the starter culture, UF pasteurized and UF raw white cheese production was performed using 2 different coagulant enzymes and 2 different starter cultures. Cheese working model is given below. 1- A cheese sample: Beyaz (White) cheese produced from ultrafilter pasteurized milk using starter culture with high proteolytic activity and chymosin enzyme 2- B cheese sample: Beyaz (White) cheese produced from ultrafilter pasteurized milk using starter culture with high proteolytic activity and microbial enzyme 3- C cheese sample: Beyaz (White) cheese produced from ultrafilter pasteurized milk using starter culture with low proteolytic activity and chymosin enzyme 4- D cheese sample: Beyaz (White) cheese produced from ultrafilter pasteurized milk using starter culture with low proteolytic activity and microbial enzyme 5- E cheese sample: Beyaz (White) cheese production from raw milk by ultrafiltration with chymosin enzyme 6- F cheese sample: Beyaz (White) cheese production from ultrafilter raw milk by microbial enzyme In order to determine the effect of storage temperature, the cheeses were stored at 4 ° C and 8 ° C. Physicochemical, chemical, microbiological and sensory analyzes were determined on the 1st, 30th, 60th, 90th and 120th days of ripening. Mineral matter analysis (Cu, Zn, Cr, P, Mg, Ca, Fe, Ka, Na) of UF white cheese was made by using ICP-OES. Proteolysis levels of cheeses were investigated by SDS-PAGE electrophoresis and reverse phase high performance liquid chromatography (RP-HPLC). In all samples, no significant changes were observed in dry matter, fat, salt, pH, titration acidity and ash. In addition, water soluble nitrogen, nitrogen soluble in 12% trichloroacetic acid (TCA), nitrogen soluble in 5% phosphotungstic acid (PTA) and acid degree increased while decreasing in protein ratio. There were a significant difference in microbiological values between UF pasteurized milk and UF raw milk samples (p <0.05). Depending on proteolysis, bitterness in cheese samples was determined in the ripening process. A cheese sample produced by chimosin enzyme and starter culture with high proteolytic activity showed the highest bitterness of sensory. According to SDS-PAGE electrophoresis analysis of cheeses on day 30th and 90th, αs-casein fragmentation occurred by 72.26% in the A cheese sample. HPLC results also showed an increase of 106% in the area of the hydrophobic peptides between 40 and 60 minutes, resulting in an intense hydrophobic peptide formation.
