Bu çalışmada, MDA uygulamalar için, mevcut klasik çok yönlü anten sistemlerine göre daha performanslı, yönlülüğü daha yüksek, biyoimplant antenler tasarlanmıştır. Bu geniş band biyomedikal implant antenlerin ışıma, yansıma katsayısı, kazanç, empedans vb. parametreleri In Vivo, Ex Vivo uygulamalarında incelenmiştir. Tasarlanan biyoimplant anten sistemi ile klasik anten sistemlerinin avantaj ve dezavantajları karşılaştırılmıştır. Önerilen antenlerle, biyouyumluluk, güç, hüzme yönlendirme ve anten boyutlarından meydana gelen problemler optimize edilerek birçok probleme çözüm getirilmiştir. Süleyman Demirel Üniversitesi, İzmir Katip Çelebi Üniversitesi, Dokuz Eylül Üniversitesi ve Anadolu Üniversitesi Elektrik Elektronik Mühendisliği Bölümlerinin Laboratuvar imkanları ile Tubitak 3001 Ar-Ge Projeleri Destek Programı desteğiyle alımı sağlanan teçhizat ve sarf malzemelerinin kullanılmasıyla biyolojik anlamda uygulanacak deney sistemi kurulmuş, doku içi uygulama gerçekleştirilmiş, deney sisteminde biyoimplant antenlerle MDA uygulamaları gerçekleştirilmiştir. Elde edilen CGHV40100F-ND Güç Transistörü' nün Büyük Sinyal Modeli ile Lineer ve Lineer Olmayan Bölge Analiz Sonuçları AWR Design Enviroment programı kullanılarak elde edilmiştir. In Vivo sıçan uygulamaları için deneklerde 4T1 fare metastatik meme kanser tümörü oluşturulmuş ve bu uygulamalar CBU Tıp Fakültesi Histoloji ve Embriyoloji Anabilim Dalı Laboratuvarlarında gerçekleştirilmiştir. Gerçekleştirilen bu In Vivo ve Ex Vivo uygulamalarında sıcaklık ölçümü gömülü sistem platformu kullanılarak DS18B20 dijital sıcaklık sensörü ile gerçek zamanlı ve kablosuz olarak yapılmıştır. Deney sistemimizde kullanılan güç transistörünün (Cree CGHV40100F-ND) büyük sinyal modeli oluşturulmuş ve güç transistörü AWR platformunda nonlineer simulasyonlarla incelenmiştir. Bu tekniklerle sağlanan güç transistörünün hedef bölgedeki ısı artışının; uygulama süresi, uygulanan güç miktarı, frekans ve uygulama mesafesi ile olan değişimi incelenmiştir. Balb/c farelerde 4T1 fare metastatik meme kanser hücre dizini kullanılarak meme kanseri modeli oluşturulmuştur. Tüm deneklerde 1-1.5x1 cm boyutlarında meme tümörü elde edilirken hücre verilmeyen memelerde beklenildiği gibi tümör oluşumu izlenmemiştir. Radyatör tip anten sistemi olarak Dual band düzlemsel ters F anten (PIFA), Yüksek kazançlı kompakt anten-1 ve Yüksek kazançlı kompakt anten-2 tasarlanmış ve bu radyatör tip antenlerle non invasif uygulamalar gerçekleştirilmiştir. İntertisyel tip anten sistemi olarak Tek slotlu koaksiyel anten ve Çift slotlu koaksiyel anten tasarlanmış ve invasif uygulamalar gerçekleştirilmiştir. In Vivo uygulamalarda kullanılan 2.476 GHz' de -23.37 dB geri dönüş kaybına, 2.45 GHz' de 18641 (V/m) elektrik alan değerine ve 2.45 GHz' de 2.15 dBi yönlendiriciliğe sahip tek slotlu Sucoform_86 koaksiyel antenin üç katmanlı (deri, yağ, kas) tümörlü biyolojik doku modeline 2.45 GHz' de yaptığı ışıma sonucunda maksimum değer 32.21 W/kg SAR/1g olarak elde edildiği görülmüştür. Görüleceği üzere maksimum SAR değeri tümör dokusu üzerinde elde edilmiştir. Kas dokusu fantom sıvısı (2.45 GHz) oluşturulmuş ve doku simulasyonları yapılmıştır. Oluşturulan kas dokusu fantom sıvısı (2.45 GHz) ile kurulan MDA düzeneği içinde hipertermi uygulaması yapılmış, 20,48 W lık PA çıkış gücü ile hipertermi sıcaklığı olan 41℃ - 44 ℃ aralığına yaklaşık 42 s' de ulaşıldığı, PA çıkış gücü 25,6 W' a yükseltildiğinde ise hipertermi sıcaklığı olan 41℃ - 44 ℃ aralığına yaklaşık 23 – 24 s' de ulaştığı gözlenmiştir. Doku içi uygulama, fantom uygulaması ve In Vivo sıçan uygulamaları sonucunda elde edilen ölçüm ve simülasyon sonuçları karşılaştırılmış ve değerlendirilmiştir. T1, T2, T3 ve T4 anten uygulamaları yapılarak ImageJ yazılımı kullanılıp mikrodalga termal ablasyonu (MDA)' a etkisi incelenmiştir. Yeni kesilmiş sığır karaciğeri üzerinde NiTi malzemelerin temel fiziksel özellikleri, mekanik özellikleri ve elektriksel performansı incelendikten sonra besleme kısmı Sucoform – 86 koaksiyel kablo olan kablonun ucuna NiTi tel eklenilerek tasarlanmış olan ve 2.45 GHz' de 24463 (V/m) elektrik alan değerine, 2.45 GHz' de 3.07 dBi yönlendiriciliğe ve 2.44 GHz' de -10.46 dB geri dönüş kaybına sahip olan NiTi halka anten kullanılarak MDA uygulaması yapılmış, 47.92 dBm (yaklaşık 50 W) AM modülasyonlu sinüs dalgasının yaklaşık 5 dk. antenimize uygulanması sonucu 28,61 mm genişliğinde, 71,54 mm yüksekliğinde ablasyon bölgesi elde edilmiş ve termal kamera ile elde edilen sıcaklık – süre ilişkisine göre en yüksek sıcaklığın olduğu kırmızı bölgede 102 ℃ sıcaklığa ulaştığı görülmüştür. 4T1 hücre dizini ile yapılan tümör modelinde histolojik kesitlerde HE boyama, PCNA immune boyama, çoğalmanın daha özgün belirteci Ki67 immune boyama, oksidatif stress belirteci eNOS immune boyama, oksidatif stress belirteci iNOS immune boyama, apoptoz belirteci TUNEL immune boyama ve damarlanma belirteci VEGF immune boyama ile adenokarsinom hücrelerin küçük ve büyük büyütmelerdeki görünümleri elde edilmiş ve adenokarsinom hücrelerin mikroskobik görünümleri üzerinden yapılan hücre çoğalmasında MDA uygulamasının anlamlı bir etki gösterdiği saptanmıştır. Duyarlı hale getirilmiş dendritik hücrelerle tümör modelinde immuno-flap uygulaması yapılarak tümör hacmi karşılaştırılmış ve anlamlı bir küçülmenin olduğu görülmüştür.
In this study, bioimplant antennas have been designed for MDA applications, which have higher performance and higher directivity than conventional multi-directional antenna systems. This broadband biomedical implant antennas are designed to measure radiation, reflection coefficient, gain, parameters were examined in In Vivo, Ex Vivo applications. The advantages and disadvantages of the designed bioimplant antenna system and classical antenna systems were compared. With the proposed antennas, the problems resulting from biocompatibility, power, beam orientation and antenna dimensions have been optimized and many problems have been solved. For in vivo rat applications, 4T1 mouse metastatic breast cancer tumor was created in the subjects and these applications were performed in the Laboratories of Histology and Embryology Department of CBU Faculty of Medicine. In these In Vivo and Ex Vivo applications, temperature measurement is carried out in real time and wirelessly with the DS18B20 digital temperature sensor using the embedded system platform. The large signal model of the power transistor (Cree CGHV40100F-ND) used in our experiment system was created and the power transistor was examined by nonlinear simulations on AWR platform. The power transistor provided by these techniques is used to determine the temperature increase in the target region; application time, the amount of power applied, frequency and the distance with the application were examined. A breast cancer model was constructed using the 4T1 mouse metastatic breast cancer cell line in Balb / c mice. In all subjects, 1-1.5x1 cm in size breast tumor was obtained, but as expected, tumor formation was not observed in the breasts without cells. Dual band planar inverted F antenna (PIFA), High gain compact antenna - 1 and High gain compact antenna - 2 were designed as radiator type antenna system and non-invasive applications were realized with these radiator type antennas. Single slot coaxial antenna and double slot coaxial antenna have been designed as intertial type antenna system and invasive applications have been realized. The single-layer Sucoform_86 coaxial antenna with -23.37 dB return loss at 2.47 GHz, 18641 (V / m) electric field value at 2.45 GHz, and 2.15 dBi at 2.45 GHz used in In Vivo applications is a three-layer (skin, fat, muscle) ) 2.45 GHz radiation to the tumor tissue model as a result of the maximum value of 32.21 W / kg SAR / 1g was obtained. As can be seen, the maximum SAR value was obtained on tumor tissue. Muscle tissue phantom fluid (2.45 GHz) was generated and tissue simulations were performed. Hyperthermia was applied in the MDA mechanism established with the muscle tissue phantom fluid (2.45 GHz), PA output power of 20.48 W and the temperature range of 41 ℃ - 44 ℃ olan with hyperthermia temperature was reached at approximately 42 s, PA output power was 25.6 W. It was observed that the temperature of hyperthermia reached 41 ℃ - 44 ℃ in approximately 23 - 24 s. Measurement and simulation results obtained from intra-tissue application, phantom application and In Vivo rat applications were compared and evaluated. T1, T2, T3 and T4 antenna applications were performed and the effect on microwave thermal ablation (MDA) was examined by using ImageJ software. After examining the basic physical properties, mechanical properties and electrical performance of NiTi materials on freshly slaughtered bovine liver, feeding part was designed by adding NiTi wire to Sucoform - 86 coaxial cable end and at 24453 (V / m) electric field value at 2.45 GHz and at 2.45 GHz. MDA application was performed using NiTi ring antenna with 3.07 dBi orientation and -10.46 dB return loss at 2.44 GHz. The 47.92 dBm (about 50 W) AM modulated sine wave was used for approximately 5 min. 28,61 mm wide, 71,54 mm high ablation region was obtained and it was observed that the temperature reached 102 ℃ in the red region where the highest temperature was obtained according to the temperature - time relationship. In the tumor model with 4T1 cell line, HE staining, PCNA immune staining, Ki67 immune staining, oxidative stress marker eNOS immune staining, oxidative stress marker iNOS immune staining, apoptosis marker TUNEL immune staining and vascularization marker VEGF immune staining The appearance of adenocarcinoma cells in small and large growths was obtained and it was found that MDA application had a significant effect on cell proliferation performed on microscopic appearance of adenocarcinoma cells. Tumor volume was compared with sensitized dendritic cells by immuno-flap in the tumor model and a significant reduction was observed.
