Bu tez çalışmasında; yüksek verim elde edilmesine rağmen kesikli olması nedeniyle endüstriyel ortamda kullanılamayan Soxhlet yöntemi yeniden tasarlanarak, aynı anda ekstraksiyon-distilasyon işlemlerini gerçekleştirebilen tek kolonlu sistem oluşturulmuştur. Tasarlanan düzenekte atkestanesinden polifenollerin ekstraksiyonu için farklı çözücüler ve kombinasyonları kullanılmıştır. Elde edilen polifenol miktarları hem Folin-Ciocalteu hem de HPLC yöntemleri ile analiz edilmiştir. Alan fraksiyonu yaklaşımıyla bu iki yöntem sonuçlarını birbiriyle ilişkilendiren bir denklem türetilmiştir. Tasarlanan düzenekteki enerji kayıpları ve elde edilen polifenol verimleri incelenerek endüstriyel üretim için farklı alternatifler oluşturulmuştur. Atkestanesi kabuklarının bitkiden ayrıldıktan sonra boyutlarının küçültülüp kurutulması ve öğütüldükten sonra kolona beslenmesinin mevcut polifenol miktarındaki kayıpları en aza indireceği tespit edilmiştir. Ekstraksiyona etki eden parametreler tekli optimizasyon yöntemiyle çalışılmış ve süre, çözücü türü ve katı/sıvı oranı için sırasıyla 200 dakika, metanol ve 8/100 (g/ml) değerleri optimum bulunmuştur. Tasarlanan düzenekte, katı materyal çözücüyle balon içerisinde ekstrakte olurken aynı zamanda karışım alttan ısıtıldığı için de çözücü distilasyona uğramakta, dolayısıyla da Soxhlet yönteminde olduğu gibi, katı belli aralıklarla taze çözücüyle karşılaşmaktadır. Endüstriyel üretime yönelik olan sürekli sistemde su-metanol çözücü kombinasyonuyla üç kolonda tek döngüyle tamamlanan ekstraksiyon işleminin polifenol miktarı, üç döngülü kontrol grubuyla karşılaştırıldığında %80-86-72 (1.kolon-2.kolon-3.kolon) oranında artış sağlamıştır. Çözücünün ekstrakte edilecek materyalle daha uzun süre maruz kalması, dolayısıyla uzun sürede çözünen bileşenlerin de çözünmesi, sürekli sistemde Soxhlet aparatındaki karışımın kontrol grubundaki gibi balona değil bir sonraki kolona beslenmesi nedeniyle ekstraktın alındığı ortamın kontrole göre daha konsantre olması bu durumun nedenlerindendir. Sürekli sistemde üretilmek istenen ürün kuersetin ise çözücülüğü yüksek olan metanol, toplam polifenol miktarı yüksek ürün üretilmek isteniyorsa su-metanol ikili kombinasyonunun kullanılmasının gerektiği sonucuna varılmıştır. Kesikli sistemde değerler karşılaştırıldığında ilk döngünün ikinci ve üçüncü döngüye göre, sürekli sistemde de birinci kolonun ikinci ve üçüncü kolona göre daha az ısı akısına sahip olduğu belirlenmiştir. Sistemlerdeki toplam ısı akıları bütünsel olarak karşılaştırıldığında kesikli sistem sürekliye göre maksimum %6,22 ve minimum %2,71'lik bir artış gerçekleştirmiştir. Bu durumda kesikli sistemin sürekliye oranla daha fazla ısı kaybına sahip olduğu göstermektedir. Sürekli ekstraksiyon-distilasyon kolonunun Soxhlet aparatı kısmının (sadece distilasyonun gerçekleştiği kısım) yalıtılmasının %46 oranında enerji tasarrufu sağlayacağı belirlenmiştir. En çok polifenol bileşeni metanol çözücülüğünde elde edilirken, en az sürekli sistem için üçlü çözücü karışımında, kesikli sistem için su-etanol ikili karışımında tespit edilmiştir. Tasarlanan sistemler arasında en çok bileşene sahip çözücü ekstraktlarının polifenol miktarının az bileşen sayısına sahip olanlardan daha düşük olduğu belirlenmiştir Belirlenen en yüksek verime sahip özütlerin HPLC analizleri sonucunda, numunelerden sadece kontrol grubunda gallik asit olduğu belirlenmiş ve numunelerin polifenol bileşimi içerisindeki kuersetin miktarları net olarak analizlenmiştir. Kuersetin miktarları, Folin-Ciocalteu sonuçları ve her bir özüte ait HPLC kromatogramları kullanılarak geliştirilen denklemle her bir polifenol bileşeninin miktarı belirlenmiştir. Kuersetin bazlı geliştirilen bu denklemin korelasyon katsayısının yaklaşık 9.10-6 olduğu tespit edilmiştir. Çalışmada üretilmiş atkestanesi ekstraktlarının gıda endüstrisinde örnek bir uygulama olarak zeytinin olgunlaştırma prosesinde kullanılabilirliği incelenmiş, mikroorganizma oluşumu, tat ve doku bakımından tuz kullanımına alternatif oluşturabileceği görülmüştür. Gerçekleştirilen bu çalışma, endüstriye hem ekstraksiyon hem de distilasyon işlemlerini aynı kolonda gerçekleştirebilen bir proses tasarımı sunmakta, bu sayede Soxhlet ekstraksiyonundaki taze çözücüyle karşılaşma avantajını sürekli sisteme taşıyan bir çalışma sistemi ortaya koymaktadır. Aynı zamanda bu proses Soxhlet'in aksine sürekli sistemde çalıştığından, sürekli sistemin avantajları olan hep aynı kalitede üretim, düşük kontrol maliyeti, kolay kontrol ve müdahale gibi birçok avantajı da bünyesinde barındırmaktadır. Enerji kayıplarının yüksek olması nedeniyle prosesin metanol çözücülüğünde, ısı ceketiyle birlikte kullanılmasının verimi çok daha yüksek seviyelere taşıyacağı öngörülmektedir. Çalışmada proses tasarımına ek olarak HPLC ve Folin-Ciocalteu yöntem sonuçları arasındaki bağıntıyı ilişkilendiren bir denklik de türetilmiştir. Bu denklem sayesinde, çözücüler değişmediği sürece tek bir materyalin HPLC analizi gerçekleştirilerek özütlerdeki bütün polifenol bileşiminin hesaplanabilmesi mümkün olabilecektir. Bu da çok daha kolay bir yöntem olan Folin-Ciocalteu yöntemiyle, hem istenilen polifenol bileşimine bağlı proses geliştirilmesini hem de HPLC analizlerine harcanacak maliyetin düşmesini sağlayacaktır. In this thesis, the Soxhlet method, which can produce the highest yield but that cannot be used in industry due to batch-wise operation, had been redesigned and a single column system capable of realizing the extraction-distillation processes simultaneously had been formed. Within this design, different solvents and their combinations were used for the extraction of polyphenols from horse chestnut. The amounts of polyphenols obtained were analyzed by both Folin-Ciocalteu and HPLC methods. With the area fraction approach, an equation that correlates the results of these two methods was derived. The energy losses and obtained polyphenol yields in the designed system were examined, and thus different alternatives were created for industrial production of different polyphenols. It has been determined that polyphenol losses during the whole process can be minimized by pre-treatment procedure including peeling, dicing, drying and grinding of the chestnut crusts. The parameters affecting the extraction were studied with single optimization method and the time, solvent type and solid/liquid ratio were found to be optimum for 200 minutes, methanol and 8/100 (g/ml) respectively. In the designed system, the solid material had been faced with the fresh solvent at certain intervals, so as in the Soxhlet method; because while the solid material had been extracted with the solvent in the flask, at the same time the solvent had been distilled off due to heating from the bottom. In the continuous system for industrial production, the polyphenol content of the extraction process completed by a single cycle in three columns with the combination of water-methanol solvent resulted in an increase of 80-86-72% (1.colon-2.colon-3.colon) compared to the three-cycle control group. The reason for this is that the solvent is exposed to the material to be extracted for a longer period of time, thus dissolving the long-dissolving components, since the mixture in the Soxhlet apparatus in the continuous system is fed to the next column rather than to the balloon as in the control group. It is concluded that quercetin should be used in the continuous system and methanol with high solubility and water-methanol binary combination should be used if the total amount of polyphenol is to be produced. When the values in the batch system were compared, it was determined that the first cycle had less heat flux than the second and third columns, and in the continuous system the first column had less heat flux than according to the second and third cycles. When the total heat fluxes in the systems are compared in total, the batch system had by a maximum of 6.22% and a minimum of 2.71% increase compared to the continuous system, which shows more heat loss occurring in the batch system than continuous one. It has been determined that the isolation of the Soxhlet apparatus portion of the continuous extraction-distillation column (only the part where the distillation takes place) would provide energy savings of 46%. Most polyphenol component was obtained in methanol solvent, while the least was obtained with the triple solvent mixture in continuous system, and with the binary mixture of water-ethanol in the batch system. Among the designed systems, it was determined that the highest amount of polyphenols containing extracts had the least number of components, and vice versa. As a result of the HPLC analysis of the extracts with the highest yields determined, gallic acid was found only in the control group and the quercetin content of the samples in the polyphenol composition was clearly analyzed. Quantities of each polyphenol component were determined based on the quercetin amounts, Folin-Ciocalteu results and the HPLC chromatograms of each extract. The correlation coefficient of this equation developed based on quercetin was found to be 9.10-6. In the food industry, the use of horse chestnut extracts as an exemplary application in the olive processing process had been investigated and it had been observed that it can be an alternative to salt usage in terms of microorganism formation, taste and texture. This study presents a process design that enables the industry to perform both extraction and distillation processes in the same column, thus revealing a working system that carries the advantage of encountering fresh solvent present in the Soxhlet extraction to the continuous system. In addition, because this process works continuously, as opposed to the Soxhlet, it has many advantages belonging to the continuous system; i.e. the same quality of production, low control costs, easy control and easy intervention. Due to the high energy losses, it is predicted that the use of this process with methanol as solvent, and in combination with the heating jacket will increase the yield to much higher levels. In addition to process design, an equation which correlates the relationship between HPLC and Folin-Ciocalteu method results was derived. With the aid of this equation, it is possible to calculate the entire polyphenol composition in the extracts just by performing HPLC analysis of a single polyphenol type, unless the solvents are changed. As a result, by applying much easier method, namely Folin-Ciocalteu method, it will be possible to develop a process for production of a specific polyphenol type, and it will reduce the cost of the HPLC analysis.
