Mayalar, bazı bakteri ve maya türleri üzerinde inhibitör etki gösterebilen protein veya glikoprotein yapısında toksinler üretebilmektedir. Bu toksinlerin, gıdalarda bozulma etmeni ve patojen mikroorganizmaların biyokontrolünde kullanılma olanakları araştırılmaktadır. Bu amaçla, söz konusu maddeleri üretme yeteneğine sahip mayaların elde edilebilmesi için çok sayıda doğal materyal ve fermente gıdadan izolasyon yapılmıştır. Ön taramalar sonucunda farklı 110 maya suşu izole edilerek saflaştırılmıştır. Elde edilen izolatların Escherichia coli, Micrococcus luteus ve Candida albicans üzerindeki inhibitör etkileri test edilmiştir. İzolatlardan 8 adedi M. luteus, 2 adedi E. coli ve 4 adedi C. albicans üzerinde inhibitör etki göstermiştir. İnhibitör etki gösteren her maya suşu için optimum besiyeri bileşimi ve inkübasyon süresi tespit edilmiştir. Farklı oranlarda glikoz denemesinde M. luteus'a karşı en iyi inhibitör etkinin % 5 glikoz oranında 4 gün inkübe edildiğinde, C. albicans'a karşı ise % 3 glikoz oranında 3 gün inkübe edildiğinde olduğu belirlenmiştir. Farklı oranlarda gliserol denemesinde M. luteus ve C. albicans'a karşı en iyi sonucun % 4 gliserol oranında M. luteus'a karşı 3 gün ve C. albicans'a karşı 4 gün inkübe edildiğinde olduğu belirlenmiştir. Farklı oranlarda Na-asetat uygulamasında M. luteus ve C. albicans'a karşı en iyi sonucun % 2 Na-asetat oranında 3 gün inkübe edildiğinde olduğu belirlenmiştir. Farklı oranlarda etanol uygulamasında M. luteus ve C. albicans'a karşı en iyi sonucun % 3 etanol oranında 3 gün inkübe edildiğinde olduğu belirlenmiştir. İnhibitör madde ürettiği düşünülen suşlar farklı sıcaklıklarda (20-25-30 ve 35°C), farklı pH'larda (5, 6, 7 ve 8) ve farklı karbon kaynakları varlığında (glikoz, gliserol, etanol, asetat) üretilmiştir. C. albicans'a karşı denemesi yapılan 11, 17, 25 ve 27 numaralı mayalardan en büyük zon çapını 17 numaralı maya suşu (36.67±3.28 mm) glikozun karbon kaynağı olarak kullanıldığı 25°C'de pH 7'de; M. luteus'a karşı denemesi yapılan 1, 19, 26, 31, 51, 54, 94 ve 95 numaralı mayalardan en büyük zon çapını 19 numaralı maya suşu (24.67±0.33 mm) gliserolün karbon kaynağı olarak kullanıldığı 30°C'de pH 7'de elde edilmiştir. İnhibisyon testinde inhibitör etki gösteren suşların Staph. aureus, L. monocytogenes, Sacc. cerevisiae, Pseu. aeruginosa, E. coli O157 ve O157:H7, B. subtilis, S. Typhimurium üzerinde de inhibisyon testleri uygulanmıştır. İnhibitör etkinin farklı patojenler üzerinde farklı derecelerde olduğu gözlenmiştir. Patojenlere karşı en etkin katil özellik gösteren izolatların tanımlanması sonucunda Metschnikowia pulcherrima, Metschnikowia reukaufii ve Saccharomyces cerevisiae oldukları belirlenmiştir. Etanol çöktürmesi yöntemi ile saflaştırılan M. pulcherrima'dan elde edilen toksin M.luteus ve Staph. aureus'a karşı inhibisyon zonu oluştururken, E. coli'ye karşı zon oluşturmamıştır. Aynı yöntemlerin uygulanmasına karşın M. reukaufii ve Sacc. cerevisiae'dan elde edilen toksin veya toksin benzeri proteinler kuyucuk yöntemi ile zon oluşturmamıştır. M. pulcherrima'dan elde edilen toksinin Bradford yöntemi ile protein miktarı 61.23 mg/mL bulunmuştur. SDS-PAGE ile elde edilen protein profiline göre M. pulcherrima katil suşunun ürettiği inhibitör maddenin molekül ağırlığının yaklaşık 7.4 kDa olduğu belirlenmiştir. % 1 (0.61 mg/mL) ve % 2 (1.22 mg/mL) oranlarında farklı konsantrasyonlarda hazırlanan örneklerinin L929 fare fibroblast hücreleri üzerindeki 24 saatlik etkileşimleri sonunda toksik olmadığı tespit edilmiştir. Model gıda olarak hazır köfte seçilmiş ve hiçbir muamele yapılmayan kontrol (K), inhibitör maddenin % 1 (0.61 mg/mL, K1) ve % 2 (1.22 mg/mL, K2) oranında eklenen üç grup oluşturulmuştur. Köfte hamurunda kuru madde (%41.17), pH (5.67), titrasyon asitliği, su aktivitesi, TBA değeri, renk analizi ile L değeri, a değeri, b değeri ve mikrobiyolojik analizler yapılmıştır. TAMB sayısı (6.32±0.01 log10 kob/g), maya ve küf sayısı (5.22±0.07 log10 kob/g), LAB sayısı (4.66±0.05 log10 kob/g), mikrokok-stafilokok sayısı (5.17±0.01 log10 kob/g), koliform grup bakteri sayısı (3.64±0.05 log10 kob/g) ve TAPB sayısı (6.28±0.02 log10 kob/g) olarak belirlenmiştir. K, K1 ve K2 gruplarındaki +4°C' de 10 gün depolama süresince tüm analizler 2., 4., 6., 8. ve 10. gün tekrarlanarak günlere göre değişimleri izlenmiştir. Genel olarak K1 grubunda bozulma göstergesi olarak kabul edilen fiziksel, kimyasal ve mikrobiyolojik analiz sonuçları K2 grubuna göre daha yüksek bulunmuştur. K grubunda ise K1 ve K2 grubundan daha yüksek olduğu belirlenmiştir. Bu bulgular katil toksinin köfteye uygulama oranı arttıkça daha fazla etkili olduğunu göstermektedir. Yeasts may produce toxins in the structure of protein or glycoproteins which may all inhibitory effects on certain bacterial and yeast species. The possible use of these toxins on biocontrol of food spoilage and pathogenic microorganisms were investigated. In order to obtain yeasts isolates capable of producing such substances, various natural materials and fermented foods were used. After preliminary screening, 110 different yeast strains were isolated and purified. The inhibitory effects of the isolates on Escherichia coli, Micrococcus luteus and Candida albicans were determined. Eight of the isolates showed inhibitory effect on M. luteus, 2 of them on E. coli and 4 of the isolates on C. albicans. Optimum medium composition and incubation time were determined for each yeast strain that exhibited inhibitory effect. After trials at various glucose concentrations, the highest inhibitory effect against M. luteus was determined at 5% glucose concentration after 4 day incubation and at 3% glucose concentration after 3 day incubation against C. albicans. The best results at different glycerol concentrations were observed at 4 % glycerol for M. luteus and C. albicans after 3 day and 4 day incubation respectively. The highest inhibitory effects against M. luteus and C. albicans in different concentrations of Na-acetate assay were obtained after incubating at 2 % Na-acetate for 3 days. The best results against M. luteus and C. albicans were found when incubating at 3 % ethanol for 3 days. Strains thought to secrete inhibitory substances were produced at different temperatures (20-25-30 and 35°C), different pH (5, 6, 7 and 8) and in the presence of different carbon sources (glucose, glycerol, ethanol, acetate). Among the yeasts 11,17, 25 and 27 tested against C. albicans, the largest zone diameter was obtained from yeast strain 17 (36.67±3.28 mm) at pH 7 at 25°C where glucose was used as the carbon source; The largest inhibition zone was obtained from Strain 19 (24.67±0.33 mm) among the tested yeasts 1, 19, 26, 31, 51, 54, 94 and 95 against M. luteus when it was grown in glycerol in 30°C at pH 7. Inhibition tests were also performed on Staph. aureus, L. monocytogenes, Sacc. cerevisiae, Pseu. aeruginosa, E. coli O157 and O157: H7, B. subtilis, S. Typhimurium. It was observed that the inhibitory effect was variable on different pathogens. Metschnikowia pulcherrima, Metschnikowia reukaufii and Saccharomyces cerevisiae were identified as the most effective killer isolates against pathogens. The toxin produced by M. pulcherrima strain purified by using ethanol precipitation showed inhibition against M. luteus and Staph. aureus, but not against E. coli. Although the same purification methods were applied to toxin or toxin-like proteins produced by M. reukaufii and Sacc. cerevisiae did not generate any inhibitory effect by well diffusion test method. The concentration of toxin produced by M. pulcherrima was determined as 61.23 mg/mL by Bradford method. According to the results of SDS-PAGE method, the molecular weight of substance produced by the M. pulcherrima killer strain was determined as around 7.4 kDa. The samples prepared in two concentrations as 1 % (0.61 mg/mL) and 2 % (1.22 mg/mL) were nontoxic after 24 hours of interaction with L929 mouse fibroblast cells. Three test group of meatballs (K), 1 % (0.61 mg/mL, K1) and 2 % (1.22 mg/mL, K2) were formed and employed as the "model food". Dry matter (41.17 %), pH (5.67), titratable acidity, water activity, TBA value, L,a,b color values were also determined. TABM (6.32±0.01 log10 cfu/g) in meatball dough, the number of yeasts and molds (5.22±0.07 log10 cfu/g), LAB (4.66±0.05 log10 cfu/g), Micrococci-staphylococci (5.17±0.01 log10 cfu/g), Coliform bacteria (3.64±0.05 log10 cfu/g) and TAPB (6.28±0.02 log10 cfu/g) were determined. During the 10 day-storage at +4°C, all analyzes were repeated on the 2nd, 4th, 6th, 8th and 10th days changes of all values were observed for all 3 test groups. In general, physical, chemical and microbiological analysis results, which are considered as indicators of deterioration, were found to be higher in K1 group than K2 group. The findings indicated that killer toxin is more effective when its concentration in meatballs was increased.
