Fermente süt ürünlerindeki besinsel, mikrobiyal, kimyasal ve duyusal içeriklerinde farklılıklar üretim yöntemleri ve kullanılan mikroorganizmalardan kaynaklanmaktadır. Bu tezin amacı, farklı kültürler kullanılarak çeşitli fermente süt ürünleri üretmek ve fermantasyon süresinde yağda çözünen vitamin (A, D, E, K) miktarlarını belirlemektir. Özellikle bu araştırmada, fermente ürünlerde fermantasyon süresince bakteriyel kaynaklı Vitamin K1 ve Vitamin K2 üretimindeki değişim değerlendirmiştir. Çalışmada altı farklı fermente süt ürünü üretilmiştir. İki farklı kültür kullanılarak üretilen kefir örnekleri, iki farklı kültür kullanılarak üretilen yoğurt örnekleri ve iki farklı probiyotik özellikli bakteri kültürü (Lactobacillus acidophilus ve Bifidobacterium bifidum) kullanılarak üretilen fermente ürün örnekleri kullanılmıştır. Her fermente ürün için fermantasyon süresince örnekler alınarak, yağ ekstraksiyonu sağlanmıştır ve yağda çözünen vitaminlerin (A, D, E ve K) standartları çalışılmıştır. Vitaminlerin belirlenmesinde uygun kolon ve koşullar sağlanarak Yüksek Performanslı Sıvı Kromotografisi (HPLC) cihazı kullanılmıştır. Farklı kefir kültür ile üretilen kefirlerde fermantasyon süresince (5., 10., 15. saatler) Vitamin A (retinol) ve Vitamin E (tokoferol) değerleri azaldığı, Vitamin D, Vitamin K1 ve Vitamin K2 değerinde arttığı tespit edilmiştir. Farklı yoğurt kültür ile üretilen yoğurtlarda fermantasyon süresince (1., 3., 4. saatler) Vitamin A (retinol) ve Vitamin E (tokoferol) değerleri azaldığı, Vitamin D değerinin 1. ve 3. saatlerde arttığı, Vitamin K1 ve Vitamin K2 değerinin 4. saatte arttığı tespit edilmiştir. Lactobacillus acidophilus kültürü ile üretilen fermente ürün fermantasyon süresince (5., 10., 15. saatler) Vitamin A (retinol) ve Vitamin E (tokoferol) değerleri azaldığı, Vitamin D, Vitamin K1 ve Vitamin K2 değerinde 10. saatte arttığı arttığı tespit edilmiştir. Bifidobacter bifidum kültürü ile üretilen fermente ürün fermantasyon süresince (5., 10., 15. saatler) Vitamin A (retinol) ve Vitamin E (tokoferol), Vitamin D, Vitamin K1 ve Vitamin K2 değerlerinin hepsinde azalma tespit edilmiştir. Fermantasyon süresince bakteriyel kaynaklı üretimin Vitamin K1 ve Vitamin K2 değerlerindeki değişim belirlenmiştir. Vitamin K2 üretimi bakteriyel kaynaklı olduğu belirtmektedir ve fermente ürünlerdeki Vitamin K2 miktarlarına yönelik çalışmalar bulunmaktadır. Bu çalışma ise çeşitli fermente ürünlerin fermantasyon süresince Vitamin K1 ve Vitamin K2 miktarlarındaki değişimin belirlenmesinde bir ilk olmuştur. The differences in nutritional, microbial, chemical, organoleptic properties in fermented milk products are stemmed from the production methods and microorganisms used. Objective of this paper is to produce fermented dairy products by using different cultures and determine the amounts of fat soluble vitamins (A, D, E, K) during the fermentation process. Particularly in this research, changes in the production of bacteria synthesized Vitamin K1 and Vitamin K2 during the fermentation process in fermented products is evaluated. In the study six different fermented dairy product is produced. Kefir samples produced with two different cultures, yoghurt samples produced with two different cultures and fermented product samples which were produced by using two different bacteria culture (Lactobacillus acidophilus and Bifidobacterium bifidum) with probiotic properties are used. Fat extraction is achieved by obtaining samples during the fermentation process for each fermented product and fat soluble vitamin (A, D, E, K) standards are studied. High-Performance Liquid Chromatography (HPLC) device is used to for the identification of Vitamins by ensuring suitable colon and conditions. In the kefir produced with different kefir culture (5., 10., 15. hours); decrease in Vitamin A (retinol) and Vitamin E (tocopherol) levels and increase in Vitamin D, Vitamin k1 and Vitamin K2 levels during the fermentation process is identified. In the yogurts produced with different yogurt culture; decrease in Vitamin A and Vitamin E levels and increase in Vitamin D level at 1st and 3rd hours, increase in Vitamin K1 and Vitamin K2 levels at 4th hour is recorded. During the fermentation process of product produced with Lactobacillus acidophilus culture, decrease in Vitamin A and Vitamin E levels, increase in Vitamin k1 and Vitamin K2 levels at 10th hour is recorded. During the fermentation process of the product produced with Bifidobacter bifidum culture an overall decrease in all levels of Vitamin A, Vitamin E, Vitamin D, Vitamin K1 and Vitamin K2 is identified. Changes in the production of bacteria synthesized Vitamin K1 and Vitamin K2 during the fermentation process is identified. It is indicated that Vitamin K2 production is of bacterial origin and there are studies available regarding Vitamin K2 levels in fermented products. This study on the other hand, has become a first in identifying the changes of Vitamin K1 and Vitamin K2 levels during the fermentation process in various fermented products.
