Kefir, kefir danesi kullanılarak uygun sıcaklıkta sütün fermente edilmesiyle oluşan fermente süt ürünüdür. Kefir danesi içinde simbiyotik ve dengeli bir yaşam sürdüren bakteri ve mayalar farklı fermentasyonlar ile sütte bulunan tüm bileşenleri metabolize ederler ve kefir ürününü oluştururlar. Oluşan organik asitler, metabolize olan şeker, aminoasit kompozisyonları ve aroma bileşenleri farklılıkları ile kefirin organoleptik özelliklerine katkı sağlanır. Araştırmamızın amacı; kefir danesinde bulunan Lactobacillus kefiranofaciens (LKF), Lactobacillus parakefiri (LP), Lactobacillus kefiri (LK), Lactobacillus acidophilus (LA), Lactobacillus bulgaricus (LB), Lactobacillus casei (LC), Lactobacillus helveticus (LH), Bifidobacterium bifidum (BB), Streptococcus thermophilus (ST), Kluyveromyces marxianus (KM) standar suşları ve kefir danesi (DK), kefir starter kültürü (SK) süt ortamında geliştirerek, özellikle aroma bileşenlerini karşılaştırmaktır. Ayrıca tüm örneklerin belli bir pH'da fermentasyon sürelerini ve mikrobiyolojik içeriklerini takip ederek; kimyasal analizler (pH, titrasyon asitliği, kuru madde), Brookfieldviskozimetre ile reolojik özelikler (viskozite, kayma gerilmesi), Yüksek Performanslı Sıvı Kromatografisi (HPLC) cihazı ile organik asitler (laktik asit, asetik asit, sitrik asit ve formik asit), şeker kompozisyonları (glukoz, laktoz ve galaktoz) ve Gaz Kromotografisi-Kütle spektroskopisiyle (GC-MS) aroma bileşenleri ve miktarlarının belirlenmesi ve tanımlayıcı analizle duyusal değerlendirmesi yapılmıştır. Bu mikroorganizmaların kefir danesinden üretilen kefire sağladığı organoleptik özelliklere katkısı yorumlanmıştır. Ürünlerin mikrobiyolojik içerik aralığı 8-9 log kob/mL korunarak ve pH 4.7'de fermentasyon sonlandırılmıştır. Kuru madde içeriği değişmemiş, laktik asitte farklılıklar belirlenmiştir. Tüm ürünlerimiz newtoniyen olmayan pseudoplastik akışkan davranış ve tiksotropi akış tipi göstermiştir. Laktik asit içeriği en yüksek 0.801±0.028 g/100mL ile LKF örneğinde, en düşük 0.574±0.044 g/100mL ile ST örneğinde bulunmuştur. Asetik asit, sitrik asit ve formik asit değerleri sırasıyla 0.425-0.728 g/100mL, 0.046-0.176 g/100mL,0.116-0.164 g/100mL arasında değişmektedir. Tüm ürünlerde laktoz ve glukoz şeker miktarları değişmekte ancak istatistiksel olarak anlamlı bir farklılık bulunmamaktadır. Kefir danesi kullanılarak kefirin galaktoz miktarı 0.106±0.004 g/100mL olarak belirlenirken en düşük galaktoz içeriğinin LC örneğine, en yüksek galaktoz içeriğinin ise LB örneğine ait olduğu gözlemlenmiştir. BB örneği 191.49 μg/L ile en yüksek alkol içeriğine sahiptir, sıralamayı 46.56μg/L ile LKF örneği takip etmektedir. Aldehit grubu içerisinde en fazla 2-methylbutanal miktarı LKF örneğinde 10.86 μg/L olarak tespit edilmiştir. 2-bütanon, asetoin miktarı en yüksek daneden üretilen kefirde belirlenirken LKF ve LP örneğinde bulunmadığı gözlemlenmiştir. Hekzanoik asit en yüksek LKF ve LH örneklerinde, en düşük BB örneğinde tespit edilmiştir. Kefirde bulunmayan etil bütirat asetik asit etil ester, en yüksek BB ve KM örneklerinde belirlenmiştir. Starter kültür ile üretilen kefirde terpenler bulunmazken daneden üretilen kefirde 2.50 μg/L olarak bulumuştur. Toplam uçucu bileşen miktarının en fazla BB örneğinde, en az LA örneğinde bulunduğu gözlemlenmiştir. Duyusal analiz sonucunda en yüksek beğeniyi daneden üretilen kefir almıştır. Tek suşlu mikroorganizmalardan üretilen ürünler arasında en yüksek beğeni LA örneğine aittir. En az beğenilen ürün ise LB olmuştur. Sonuç olarak kefire özgü mikroorganizmalardan elde edilen ürünlerdeki bileşenlerin farklı miktarlarda olduğu gözlemlenmiş ve kefir aromasını etkilediği bu çalışma ile desteklenmiştir. Standart (tek suş) mikroorganizmalardan üretilen ürünler ile kefirler arasındaki aroma farklılıkları, mikroorganizmaların sinerjik etkilerinden kaynaklandığını düşündürmektedir. Kefir danesine özgü mikroorganizmaların ayrı ayrı bileşenlerinin hesaplanması ve kefir danesi kullanılarak üretilen kefir ile karşılaştırılarak yorumlanması literatüre katkı sağlamaktadır. Kefir is a fermented dairy beverage produced by fermentation of milk with kefir grains at optimal temperature. Kefir grains contain a microbial symbiotic mixture of bacteria and yeasts that metabolize the components of milk by different fermentation methods and produce kefir. The organic acids, metabolized sugar, amino acids and aroma forming compounds improve kefir's organoleptic properties. The aim of this study is to grow strains of Lactobacillus kefiranofaciens (LKF), Lactobacillus parakefiri (LP), Lactobacillus kefiri (LK), Lactobacillus acidophilus (LA), Lactobacillus bulgaricus (LB), Lactobacillus casei (LC), Lactobacillus helveticus (LH), Bifidobacterium bifidum (BB), Streptococcus thermophilus (ST), Kluyveromyces marxianus (KM) on different types of milk and kefir grains or starter cultures and to compare the aroma forming compounds. Moreover, by observing the duration of fermentation at a certain pH value and microbial components of the sample; chemical analyses (pH, titratable acidity and dry matter) were performed; rheological properties (viscosity, shear stress) were determined by Brookfield Viscometer; organic acids (lactic acid, acetic acid, citric acid and formic acid) and glucose composition (glucose, lactose and galactose) were analyzed by High-Performance Liquid Chromatography (HPLC) and Gas chromatography mass spectrometry (GC/MS) was used for determination of the aroma forming compounds and their amount and descriptive sensory evaluation was performed. We analyzed the organoleptic properties of kefir produced from kefir grains of these microorganisms. The microbiological content of the kefir was maintained at 8-9 log CFU/mL, and the fermentation was terminated when the pH value of the beverage reached 4.7. No difference was detected for the dry matter content; however, there was a difference in lactic acid. All the products were non-newtonian pseudoplastic fluids and showed thixotropic behavior. The highest lactic acid content was determined as 0.801±0.028 g/100mL in the LCF sample and the lowest lactic acid content was determined as 0.574±0.044 g/100mL in ST sample. The concentrations of acetic acid, citric acid and formic acid ranged between 0.425-0.728 g/100mL, 0.046-0.176 g/100mL, 0.116-0.164 g/100mL, respectively. The lactose and glucose content of products were different; however, the difference is not statistically significant. The galactose content of kefir produced from kefir grains was determined as 0.106±0.004 g/100mL. The lowest galactose content was determined as 0.106±0.004 g/100mL in the LC sample and the highest galactose content was detected in the LB sample. The BB sample had the highest alcohol content with 191.49 μg/L, and it is followed by the LFF sample with the alcohol content of 46.56 μg/L. Among aldehydes, the maximum amount of 2-methylbutanal was detected as 10.86 μg/L in the LKF sample. The highest amount of 3-hydroxy-2-butanone (acetoin) was detected in the kefir produced from the kefir grain, and it was detected in the LKF and the LP samples. The highest hexanoic acid content was observed in the LKF and the LH samples and the lowest content was detected in the BB sample. Ethyl butyrate, acetic acid and ethyl ester, which are not found in kefir, were detected in BB and KM samples. Kefir produced from starter culture did not contain terpenes; whereas kefir produced from kefir grains had 2.50 μg/L of terpenes. The total amount of volatile compounds was detected as highest in the BB sample and as the lowest in the LA sample. Pursuant to the sensory analysis, kefir produced from kefir grains was liked the most. Among the products produced from single-strain microorganisms, the LA sample was the most favored one, whereas the least liked product was the LB sample. It was observed that the components in the kefirs produced by specific microorganisms were in varying amounts, and this study showed that the components of kefir have an effect on its aroma. The aromatic differences between the products produced from single-strain microorganisms and kefir grains are considered to have stemmed from the synergistic effects of the microorganisms. The calculation of the individual components formed by the microorganisms that are specific to kefir grains separately and comparing the products with kefir produced from kefir grains can contribute to the literature.
