Yüksük otu (Digitalis purpurea L.) zengin sekonder metabolit ve özellikle kalp ilaçlarının hammaddesini oluşturan kardenolit glikozitleri içermesinden dolayı ülke ekonomisine ve tıp sektörüne önemli katkıları olan Scrophulariaceae familyasına ait otsu bir bitkidir. Bu çalışmada yüksük otu bitkisinin in vitro kültürdeki başarısı ile stres koşullarında sekonder metabolit içeriği arasındaki ilişkinin belirlenmesi ve yüksük otunun fenolik ve aromatik madde miktarının maksimum olduğu büyüme ortamının tespit edilmesi amaçlanmıştır. Yüksükotu bitkileri, tohumların ekiminden itibaren 16 saat/gündüz 8 saat/gece fotoperiyodu düzeninde, 135 μmol m-2 s-1 PAR ışık yoğunluğu, 22-24 °C sıcaklık ve % 51-54 nem oranı koşulları altında ortalama 1-2 ay boyunca yetiştirilmiştir. Bitki büyüme odasında yetiştirilen bitkilerin yaprakları eksplant olarak kulanılmış ve eksplantlar yüzey sterilizasyonu yapıldıktan sonra farklı NaCl konsantrasyonlarını (0, 75, 100, 125, 150 ve 200 mM NaCl) içeren Murashige ve Skoog (MS) besin ortamlarında kültüre alınmıştır. Her NaCl uygulaması için kallus oluşum oranları belirlenmiş ve oluşan kallusların taze ağırlıkları ölçüldükten sonra eppendorf tüperine alınarak fenolik bileşiklerin miktarının belirlenmesi amacı ile -18°C'de saklanmıştır. Daha sonra kallusların fenolik madde miktarları HPLC, aromatik miktarları ise GC-MS ile her uygulama için ayrı ayrı belirlenmiştir. Çalışmamızda toplamda 976 yüksük otu yaprak eksplantı kültüre alınmış bu eksplantlardan 618 kallus elde edilmiştir (%63,31). En yüksek kallus oluşum oranının kontrol grubunda (NaCl içermeyen) (%81,17), en düşük oranın ise 100 mM NaCl uygulamasında (%44,37) olduğu bulunmuştur (P<0.01). 150 mM NaCl ortamında kontrol grubundan sonra en yüksek değer gözlenmiştir. 200mM'lık NaCl ortamında ise kallus oluşumu meydana gelmemiştir. Kallus taze ağırlıklarında NaCl'nın artan konsatrasyonları ile birlikte genel anlamda bir düşüş gerçekleşmiştir. Kontrol grubunda bu oran 0,1487 g iken 150 mM NaCl ortamında 0,0091 g'a düşmüştür (P<0.01). Yüksük otu yaprak analizinde kateşin tespit edilemediği halde toplam fenolik madde miktarı diğer çeşitli NaCl uygulamalarında oluşan kalluslardan çok daha fazla ve 157,6 μg/g olarak bulunmuştur (P<0.01). Kontrol grubu kalluslarında ise genel olarak NaCl uygulamalarından elde edilen kalluslara nazaran fenolik madde miktarının daha düşük olduğu bulunmuştur (P<0.01). GC-MS cihazıyla ölçülen aromatiklerin en yüksek madde miktarı 150 mM NaCl uygulamasında bulunmuştur. Yüksek konsantrasyonda NaCl bu grup kalluslarda fenolik madde miktarını düşürürken aromatik madde miktarını artırmıştır. Ayrıca kontrol grubu kalluslarında bulunmayan ve tuz stresine karşı savunma olarak sentezlendiği düşünülen farklı aromatikler tespit edilmiştir. Foxglove (Digitalis purpurea L.) is a herbaceous plant belonging to the Scrophulariaceae family, which has significant contributions to the national economy and the medical sector since it contains rich secondary metabolites and cardenolite glycosides which are the raw materials of heart medicines. The aim of this study was to determine the relationship between the success of in vitro callus culture of foxglove plant and secondary metabolite content under stress conditions and to determine the growth medium with maximum phenolic and aromatic substances of foxglove. Foxglove plants were grown for 16 hours/ ay 8 hours/night under the conditions of 135 μmol m−2 s−1 PAR light intensity, 22-24 ° C temperature and 51-54% humidity for an average of 1-2 months. The leaves of the plants grown in the plant growth chamber were used as explants and after surface sterilization, the explants were cultured in Murashige and Skoog (MS) containing different NaCl concentrations (0, 75, 100, 125, 150 and 200 mM NaCl). Callus formation rates were determined for each NaCl application and then the fresh weights of the callus were measured and taken into the eppendorf tube and stored at -18 °C to determine the amount of phenolic compounds. Then the phenolic contents of callus were determined by HPLC and the aromatic contents by GC-MS for each application. In our study, a total of 976 foxglove leaf explants were cultured and 618 callus was obtained from these explants (63.31%). The highest rate of callus formation was found in the control group (without NaCl) (81.17%) and the lowest rate was found in 100 mM NaCl (44.37%) (P <0.01). The highest value was observed in 150 mM NaCl medium after the control group. Callus formation didn't occur in 200 mM NaCl medium. There was a general decrease in callus fresh weights with increasing concentrations of NaCl. In the control group, this ratio decreased from 0.1487 g to 0.0091 g in 150 mM NaCl (P<0.01). Although catechin could not be detected in the foxglove leaf analysis, total phenolic content was found to be much higher and 157.6 μg/g compared to the calluses formed in various NaCl applications (P <0.01). In the control group callus, it was found that the amount of phenolic substances was lower than the callus obtained from NaCl applications in general (P<0.01). The highest amount of aromatics measured by GC-MS was found in 150 mM NaCl. High concentrations of NaCl decreased the amount of phenolic substances in this group callus while increasing the amount of aromatic substances. In addition, different aromatics which were not found in the callus of the control group and thought to be synthesized as defense against salt stress were determined.
