Bu tez çalışmasında hem normal hem de %24 PEG 6000 konsantrasyonu ile oluşturulan kuraklık stresine maruz bırakılan H. vulgare L. Bülbül-89 bitkisindeki fizyolojik (çimlenme yüzdesi, radikula sayısı, radikula uzunluğu, koleoptil uzunluğu, koleoptil yüzdesi ve taze ağırlık) ve moleküler (hücre döngüsü, mitotik aktivite, kromozom davranışları, total protein miktarı) özelliklere dışsal poliaminlerin (Kad, Spd, Put, Spm) etkileri araştırılmıştır. Çimlenme deneyleri için tohumlar sterilizasyonuna tabi tutulmuşlardır. Sterlizasyonu sağlanan tohumlar 50 ml çözelti (distile su, 10 µM Kad, Spd, Put ve Spm) olan erlen kaplarında 24 saat bekletildikten sonra saf su ve PEG 6000 konsantrasyonlarının olduğu petrilere ekilerek etüve alınmıştır. Etüvde bekletilen tohumlardan 7. gün sonunda fizyolojik parametreler ve hücre döngüsü analizleri yapılmıştır. Mitotik indeks ve kromozom davranışlarının belirlenebilmesi için çimlendirilen tohumların kök uçları 0,5-1 cm uzunluğuna ulaştığında kesilerek kök uçlarından preparasyon işlemleri yapılmıştır. Total protein analizleri için çimlendirilen tohumlar viyollere alınarak torf toprağa ekimi yapılmıştır. Daha sonra iklim kabinine alınan bitkilerden kontrol grubu bitkileri üç günde bir 7 ml saf su ile sulanmıştır. Kuraklık uygulanan bitki grubu ise onuncu gün 7 ml %24'lük PEG 6000 ile sulanmıştır. Örnekler 21. gün sonunda iklim kabininden alınmış ve sıvı azot ile parçalanarak -80°C'de depolanmıştır. Tek başına poliaminlerin olduğu ortamda çimlendirilen arpa tohumlarının taze ağırlıkları saf su ortamında çimlendirilen kontrol grubuna kıyasla artmıştır, diğer fizyolojik parametreler kontrol grubu ile benzer etkiler göstermişlerdir. Diğer yandan kuraklık stresi, çimlenme yüzdesi, taze ağırlık, radikula uzunluğu, radikula sayısı, koleoptil yüzdesi ve uzamasını önemli ölçüde engellemiştir. Stresli ortam koşullarında poliaminlerin uygulanması sonucunda taze ağırlık ve radikula sayıları üzerinde engelleyici etkinin devam ettiği belirlenirken, çimlenme yüzdesi, radikula uzaması, koleoptil yüzdesi, koleoptil uzunluğu parametrelerinde Spm en fazla artış sağlayan poliamin olmuştur. Yapılan çalışmada kontrol gruplarına uygulanan poliaminlerin hepsinin mitotik indeksi azalttığı belirlenmiştir. Kuraklık stresi sonucunda mitotik indekste azalma gözlenirken, Kad ve Put bu parametre üzerinde olumlu bir etki gösterememişlerdir. Kuraklık stresi altında Spd ve Spm mitotik indeksi kendi kontrol grubuna göre sırasıyla %20 ve %40 oranında arttırmıştır. Ayrıca kontrol grubuna uygulanan tüm poliaminler, çeşitli tip ve yüzdelerde kromozom anormalliklerine neden olmuştur. Stresli koşullar altında uygulanan tüm poliaminler kromozom anormallikleri üzerinde hafifletici etkiler göstermişlerdir ve Kad'ın bu anormallikleri en aza indiren poliamin olduğu tespit edilmiştir. Hücre döngüsü analizi sonuçlarına göre, Spd uygulaması S fazında kontrol grubuna göre artmış olmasına rağmen, incelenen diğer poliaminlerin azaltıcı bir etkisi olmuştur. G2 fazında ise Kad ve Spm uygulaması artışa neden olmuştur. Tek başına %24 PEG 6000 uygulaması sonucunda G1 ve G2 safhalarında kontrol grubuna göre sayısal değer olarak artış olurken, PEG 6000+Poliamin uygulamaları sonrasında G1 safhasında kendi kontrolüne göre azalma olduğu belirlenmiştir. G2 aşamasında kuraklık stresi Spm ve Spd uygulamalarında istatistiksel olarak anlamlı bir artışa neden olmuştur. Normal şartlar altında tek başına poliaminlerin uygulanması sonucunda, incelenen tüm poliaminlerde kontrole kıyasla toplam protein miktarında belirli bir artış olurken, en yüksek artış %100 ile Kad uygulamasında kaydedilmiştir. Sadece PEG 6000 uygulamasında, kontrol grubuna (saf su) kıyasla toplam protein miktarında 2 kat artış gözlenirken, PEG 6000 + Poliamin uygulamaları kontrolüne kıyasla (%24 PEG 6000) dikkate değer bir artış göstermiştir. Kuraklık stres koşullarında, toplam protein miktarı 1.667 mg / ml seviyesi ile en fazla Spd uygulamasından sonra gerçekleşmiştir. Yapmış olduğumuz çalışmada kuraklık stresi altında tohum çimlenmesi, fide büyümesi gibi fizyolojik parametrelerle, mitotik indeks, hücre döngüsü ve toplam protein miktarları üzerinde poliaminlerin iyileştirici etki gösterdiği ve tüm parametreler ışığında Spm'in en başarılı poliamin olduğu sonucu ortaya çıkmaktadır.Bununla birlikte poliaminlerin, özellikle Spm'in uygun zaman ve dozla birlikte uygulanmasının kuraklık stresinin bitki büyümesi ve gelişmesi üzerindeki olumsuz etkileri büyük ölçüde hafifletebileceği ve kurak arazilerde yetişebilecek bitkiler için kullanılmasının ülkemizin ekonomisine çok yararlı sonuçlar sağlayabileceği düşünülmektedir. In this thesis the effects of exogenous polyamines (Cad, Spm, Put, Spd) on physiological (germination percentage, number of radicula, radicular length, coleoptile lenght and fresh weight) and molecular features (cell cycle, mitotic activity, chromosome behaviors, total protein content) parameters in H. vulgare L. Bülbül-89 plant exposed to both normal and drought stress (%24 PEG 6000) were investigated. Seeds were subjected to sterilization for germination experiments. After the sterilise seeds were kept in flask containers with 50 ml of solution (distilled water, 10 µM Cad, Spd, Put ve Spm) for 24 hours, they was planted in petri dishes containing distilled water and PEG 6000 concentrations. Physiological parameters and cell cycle analyzes were performed at the end of the 7th day from the seeds kept in the oven. In order to determine the mitotic index and chromosome behaviors, the root tips of the germinated seeds were cut when they reached 0.5-1 cm length and the preparation was done from the cut root tips. The seeds germinated for total protein analysis were taken to viols and planted in peat soil. Later, the control group plants, which were taken into the climate cabin, were watered every three days with 7 ml of distilled water. On the tenth day, drought treated plants were irrigated with 7 ml of 24% PEG 6000. Samples were taken from the climate cabin at the end of the 21st day and stored at -80° C by breaking down with liquid nitrogen. The fresh weight of seeds germinated in the alone polyamines conditions increased when compared with the control group germinated in the distilled water environment, other physiological parameteres showed similar effects with the control group. On the other hand, drought stress significantly has prevented germination percentage, fresh weight, radicle length, coleoptile percentage and coleoptile length. Result of the application of polyamines under stressful enviroment conditions, while determining that the inhibitory effect on fresh weight and radicula numbers continues, but at the parameters of the germination percentage, radicle elongation, coleoptile percentage, coleoptile length was Spm which provided the highest increase. In these study, it was determined that all of the polyamines applied to the control groups decreased the mitotic index. While a decrease in the mitotic index was observed as a result of drought stress, Cad and Put could not show a positive effect on this parameter. Under drought stress, the Spd and Spm mitotic index increased by 20% and 40%, respectively, compared to their control group. In addition, all polyamines applied to the control group (distilled water) caused chromosome abnormalities of various types and percentages. All polyamines applied under stressful conditions was mitigating effect on chromosome abnormalities and it is determined that Cad is polyamine that minimizes these abnormalities. According to the results of the cell cycle analysis, although the Spd application increased in the S phase compared to the control group, other polyamines studied had a reducing effect. In the G2 phase, Cad and Spm application caused an increase. As a result of 24% PEG 6000 application alone, the G1 and G2 stages increased numerically compared to the control group, while after the PEG 6000 + Polyamine applications, the G1 stage decreased compared to its control. In the G2 phase, drought stress caused a statistically significant increase in Spm and Spd applications. As a result of the application of polyamines alone under normal conditions, there was a certain increase in the total protein amount compared to control in all polyamines studied, while the highest increase was recorded in Cad application at 100%. In PEG 6000 application alone, 2-fold increase in total protein amount compared to the control group (distilled water) was observed, while PEG 6000 + Polyamine applications showed a remarkable increase compared to its control (%24 PEG 6000). In drought stress conditions, total protein amount was realized after the most Spd application with 1.667 mg / ml level. In our study, it is revealed that polyamines have a mitigating effect under the drought stress on physiological parameters such as seed germination, seedling growth, mitotic index, cell cycle and total protein amounts, and Spm is the most successful when all parameters are examined. However, it is thought that the application of polyamines, especially Spm, with appropriate time and dose may greatly alleviate the adverse effects of drought stress on plant growth and development. In our country, it is thought that using Spm for plants that grow in arid region can provide very useful results.
