Bu tez çalışmasında, Yapay Sinir Ağları (ANN) kalibrasyon yöntemi iki farklı ilaç aktif maddesi olan Levadopa (LEV) ve Benserazid (BEN)'in aynı anda miktar tayinleride hiçbir ayırma işlemi yapılmadan başarıyla uygulanmıştır. UV/Görünür Bölge Spektroskopisi yönteminden elde edilen veriler ise kemometrik olarak değerlendirilerek yorumlanmıştır. Spektrofotometrik yöntemde, levadopa ve benserazid' in stok çözeltisi hazırlamak için 10 mg tartılarak 0,1M HCl çözücüsünde çözüldükten sonra son hacim 100 mL' ye tamamlanmıştır. Hazırlanan bu çözeltilerden levadopa için 2-10 ppm ve benserazid için 0,5-2,5 ppm arasındaki derişimlerde aktif maddeler stok çözeltilerden alınarak toplam hacim 25 mL'ye tamamlanmıştır. Çözücüden oluşan köre karşı absorbanslar 0,1 nm aralıklardaki duyarlılıkta okunmuştur. Ticari olarak alınan tablet (ilaç) numunesi Madopar ilacından 20 tablet havanda iyice ezilip ilacın bir tabletinin toplam ağırlığına karşılık gelen miktar ilaç tartılmıştır. Tartılan kısma 0,1M HCl eklenip iki saat manyetik karıştırıcıda karıştırılmış ve son hacimleri 100 ml'ye tamamlanmıştır. Bu çözeltinin absorbans değerleri kaydedilerek elde edilen değerlere yapay sinir ağları yöntemi uygulanmıştır. İlaç aktif madde karışımlarının yapay sinir ağları metoduyla elde edilen ortalama geri kazanım değerleri LEV için % 99,98, SS 0,18 BEN için % 99,89 SS 0,45 olarak elde edilmiştir. Çalışmada uygulanan kemometrik yöntem yardımıyla elde edilen sonuçlar son derece hızlı, basit ve güvenilir sonuçlarlardır. In this thesis, Artificial Neural Networks (ANN) calibration method has been successfully applied in the simultaneous quantification of two different drug active substances, Levadopa (LEV) and Benserazide (BEN), without any separation. The data obtained from UV / Visible Spectroscopy method were evaluated and interpreted chemometrically. In the spectrophotometric method, to prepare the stock solution of levadopa and benserazide, the final volume was completed to 100 ml after 10 mg was weighed and dissolved in 0.1M HCl solvent. From these prepared solutions, active substances were taken from the stock solutions at concentrations between 2-10 ppm for levadopa and 0.5-2.5 ppm for benserazide, and the total volume was completed to 25 mL. Absorbances against the solvent-based blank were read at sensitivity ranges of 0.1 nm. Commercially taken sample of tablets (drug) 20 tablets of Madopar drug were crushed thoroughly in a mortar and the amount of drug corresponding to the total weight of one tablet of the drug was weighed. 0.1M HCl was added to the weighed part and mixed in a magnetic stirrer for two hours and the final volume was completed to 100 ml. Artificial neural networks method was applied to the values obtained by recording the absorbance values of this solution. The average recovery values of the drug active ingredient mixtures obtained by artificial neural network method were 99.98% for LEV and 99.89% SS 0.45 for SS 0.18 BEN. The results obtained using the chemometric method applied in the study are extremely fast, simple and reliable results.
