Bu çalışmada Isparta ve Antalya illerinde perakende satışı yapılan kanatlı etlerinde (tavuk, hindi, bıldırcın) Staphylococcus aureus yaygınlığının araştırılması, antibiyotik direnç profilleri ve direnç genlerinin yanı sıra enterotoksin genleri varlığının belirlenmesi amaçlanmıştır. Çalışma kapsamında, 32 tavuk, 9 hindi ve 9 bıldırcın olmak üzere toplam 50 kanatlı eti örneği incelenmiştir. Kanatlı etlerinden muhtemel S. aureus izolasyonunda Baird Parker agar ve mannitol salt agar besiyerleri kullanılmıştır. Baird Parker agar besiyerinde siyah merkezli, etrafında zon oluşumu görülen, mannitol salt agar besiyerinde ise sarı renkte olan toplam 130 koloni muhtemel S. aureus olduğu düşünülerek çalışma materyali olarak seçilmiştir. İzolatlara Gram boyama ve katalaz testi uygulanmış ve izolatların tamamının Gram pozitif, kok morfolojisinde ve katalaz pozitif özellikte olduğu belirlenmiştir. İzolatların tür düzeyinde tanısı S. aureus'da termostabilnükleaz genine (nuc) özgü primer çiftleri kullanılarak polimeraz zincir reaksiyonu (PZR) ile yapılmıştır. İzolatların 16 adedinde nuc genine özgü 458 bç büyüklüğünde amplikonlar elde edilmiştir. Bu bulgular ışığında 16 adet izolat S. aureus olarak tanımlanmıştır. S. aureus izolatlarının antibiyotik direnç profilleri disk difüzyon yöntemi ile belirlenmiştir. Disk difüzyon testi sonucu, izolatların tamamının (% 100) kloramfenikol ve teikoplanine duyarlı, penisilin G'ye ise dirençli olduğu belirlenmiştir. Ayrıca izolatların % 81.25'inin (13/16) metisilin dirençli S. aureus (MRSA) olduğu tespit edilmiştir. S. aureus izolatlarında antibiyotik direnç geni ve enterotoksin geni varlığı PZR ile araştırılmıştır. PZR işlemi sonucu S. aureus suşlarında en sık rastlanan antibiyotik direnç genin blaZ (% 62.5, 10/16) olduğu tespit edilmiştir. S. aureus suşlarının hiçbirinin sea, seb, sec, sed ve see enterotoksin geni içermediği tespit edilmiştir. In this study, it was aimed to investigate the prevalence of Staphylococcus aureus in poultry meat (chicken, turkey, quail) sold in the provinces of Isparta and Antalya, and to determine the presence of enterotoxin genes as well as antibiotic resistance profiles and resistance genes. Within the scope of the study, a total of 50 poultry meat samples, 32 chicken, 9 turkey and 9 quail, were examined. Baird Parker agar and mannitol salt agar media were used in the possible isolation of S. aureus from poultry meat. A total of 130 colonies with a black center, zoning formation around them on Baird Parker agar medium and yellow color on mannitol-only agar medium were selected as the study material considering that they were possible S. aureus. Isolates of Gram staining and catalase test were applied all it was determined that all isolates were Gram positive, cocci morphology and catalase positive. Isolates of S. aureus species level in the diagnosis of the thermostable nuclease gene (nuc) polymerase chain reaction using specific primer pairs (PCR) was performed. In 16 of the isolates, 458 bp amplicons specific to the nuc gene were obtained. In the light of these findings, 16 isolates were defined as S. aureus. Antibiotic resistance profiles of S. aureus isolates were determined by disk diffusion method. As a result of disk diffusion test, it was determined that all of the isolates (% 100 ) were susceptible to chloramphenicol and teicoplanin and resistant to penicillin G. In addition, it was determined that % 81.25 (13/16) of the isolates were methicillin resistant S. aureus (MRSA). Antibiotic resistance gene and enterotoxin gene in the presence of S. aureus isolates were tested by PCR. The most common antibiotic resistance gene in S. aureus strains was found to be blaZ (%62.5 , 10/16) as a result of PCR. It was determined that none of the S. aureus strains contained sea, seb, sec, sed and see enterotoxin genes.
