Bu çalışmada farklı kaynaklardan sağlanan kefir daneleri (K1, K2, K3, K4 ve K5) geleneksel yöntem ile üretilerek, elde edilen kefir örneklerinin fizikokimyasal, duyusal ve mikrobiyel özellikleri belirlenmiştir. Özellikle fermantasyon süresince mikroorganizma sayılarındaki değişim kültürel sayım ve kültürden bağımsız yöntem ile (qPZR ile sayım) izlenerek mikrobiyotadaki farklılıkların kaliteye yansıması değerlendirilmiştir. Fermantasyon sonrası tüm kefirlerin; fizikokimyasal (kurumadde, yağ, titrasyon asitliği, pH, viskozite, renk analizi), duyusal ve mikrobiyolojik (Lactobacillus spp., Lactococcus spp., maya, asetik asit bakterileri (AAB) ve Bifidobacterium spp. sayımı) özelliklerinin yanı sıra uçucu aroma bileşenleri de belirlenerek en iyi özelliklere sahip kefir danesi seçilmiştir. Fizikokimyasal analiz sonuçlarına göre kefir gurupları arasında titrasyon asitliği (%LA) pH ve viskozite değerlerinde önemli (p<0.05) düzeyde farklılık saptanırken; kurumadde, yağ ve renk (L*, a*, b*) değerleri arasındaki farkın önemli olmadığı (p>0.05) tespit edilmiştir. Sayım sonuçlarına göre kefir örneklerinde en fazla bulunan Lactobacillus spp. ve Lactococcus spp.'yi maya, AAB ve Bifidobacterium spp. takip etmiştir. K5 kefir örneği duyusal analiz sonucunda diğer kefir örneklerine göre daha çok beğenilmiştir. Tüm analiz sonuçları bir arada değerlendirildiğinde diğerlerine kıyasla üstün özelliklere sahip K5 kefiri üretilerek fermantasyon süresince (0, 4, 8, 12, 14, 16, 18, 20, 22 ve 24. saat) yukarıda belirtilen analizler tekrarlanmıştır. Böylece fermantasyon sürecinde mikrobiyel gelişmeye bağlı olarak meydana gelen değişimler izlenmiştir. Fermantasyon ilerledikçe özellikle uçucu aroma bileşenlerinin farklılık gösterdiği ve en yüksek aroma bileşen profiline 12. ve 16. saatlerde ulaşıldığı saptanmıştır. Yapılan çalışma ile fermantasyon sırasında mikroorganizma sayılarında meydana gelen değişimin SPME ile belirlenebilen uçucu aroma bileşenlerine etkisi aydınlatılmıştır. Böylece mikroorganizma sayılarında ve buna bağlı olarak uçucu aroma bileşenlerindeki değişim esas alınarak tüketici talebine uygun ürünler için gerekli fermantasyon süreleri belirlenmiştir. K5 kefir örneğinin kültürel sayım ve qPZR ile yapılan sayım sonuçları arasında da istatistik açıdan önemli fark olduğu (p<0.05) görülmüştür. qPZR sayım sonuçları, kültürel sayım sonuçlarına göre yaklaşık 0.05-2.20 logaritmik birim yüksek bulunmuştur. In this study, kefir grains (K1, K2, K3, K4, and K5) which are obtaining from different sources were produced by the traditional method. The physicochemical, sensory, and microbial properties of these kefir samples were determined. The change in the number of microorganisms during the fermentation was monitored by cultural counting and culture-independent method (qPCR). The effect of differences in microbiota on quality was evaluated. Various analysis results obtained from all kefir groups at the end of fermentation were taken into account in selecting the kefir grains with the best characteristics. These analyzes are; physicochemical (dry matter, oil, titration acidity, pH, viscosity, colour analysis), sensory and microbiological (Lactobacillus spp., Lactococcus spp., yeast, acetic acid bacteria (AAB), and Bifidobacterium spp. count) properties as well as volatile aroma component results. According to the results of physicochemical analysis, a significant (p<0.05) difference was found between kefir groups in titration acidity (%LA), pH and viscosity values. It was determined that the difference between dry matter, oil and colour (L*, a*, b*) values was not significant (p>0.05). According to the microbiological colony count results, Lactobacillus spp., and Lactococcus spp., were the most abundant in kefir samples, followed by yeast, AAB, and Bifidobacterium spp. According to the sensory analysis results, the K5 kefir sample was found to be better than other kefir samples. When all the analysis results were evaluated together, K5 kefir with thoroughpaced properties was produced and the above-mentioned analyzes were repeated during the fermentation (0, 4, 8, 12, 14, 16, 18, 20, 22, and 24 hours). Thus, changes in the fermentation process due to microbial growth were observed. In the following days of fermentation, it was determined that especially the volatile aroma components differed, and the highest aroma component profile was reached at the 12th and 16th hours. Our study results have provided the clarification of the effect of the change in the number of microorganisms during fermentation on the volatile aroma components that can be determined by SPME. Thus, the necessary fermentation times for products suitable for consumer demand were determined based on the change in the number of microorganisms and, accordingly, the volatile aroma components. It was observed that there was a statistically significant difference (p<0.05) between the cultural and qPCR counting results of the K5 kefir sample. qPCR results were approximately 0.05-2.20 logarithm higher than the cultural results.
