Bu tez çalışmasında, bıldırcınlarda solunum sisteminin bir parçası olan sirinksin yapısına katılan doku bölümlerinde mast hücrelerinin dağılımı ve heterojenitesinin belirlenmesi amaçlandı. Çalışmada 10 adet sağlıklı erişkin bıldırcına (Coturnix coturnix) ait sirinks örnekleri kullanıldı. Alınan sirinks dokuları Bouin, %10'luk formaldehit, Schaudin, Orth ve Alkolik Formalin fiksatif solüsyonlarında tespit edilerek, rutin histolojik doku takibi işlemlerinden geçirildikten sonra parafinde bloklandı. 5 mikrometre kalınlığında alınan seri kesitlere genel histolojik yapının belirlenmesi amacıyla Hematoksilen-Eozin boyama yöntemi, mast hücrelerinin dağılımlarının belirlenmesi için Standart Toluidine Blue (TB) boyama yöntemi ile mast hücrelerinin heterojenitelerinin belirlenmesi amacıyla Thionin ve Alcian Blue/Safranin O (AB/SO) boyama yöntemleri uygulandı. Uygulanan boyama yöntemlerinin sonucunda bıldırcın sirinksinin histolojik yapısı ile bağ dokusu mast hücreleri (BDMH) ve mukozal mast hücrelerinin (MMH) dağılımları ve heterojeniteleri değerlendirildi. Bıldırcın sirinksinin trakenin kaudal bölümündeki son bir trakeal halka ile primer bronşların başlangıçlarındaki ilk dört-beş halkanın modifikasyonu sonucu şekillendiği ve trakeya-bronşiyal tip olduğu ve pessulusun kemikleşmiş olduğu görüldü. MMH'nin kan damarları etrafında kümeler oluşturduğu ve kıkırdak halkalar arasında daha yoğun dağılım gösterdiği tespit edildi. Uygulanan Thionin boyama yönteminde trakeyal ve bronşiyal sirinksteki en yoğun BDMH ve MMH dağılımının Alkolik Formalin fiksasyonunda olduğu tespit edildi. TB boyama yönteminde trakeyal ve bronşiyal sirinksteki bazı mast hücrelerinin metakromazi gösterdiği görüldü. Ayrıca TB boyama yönteminde ideal ve en yoğun boyanmanın Schaudin fiksatifi ile olduğu, bununla birlikte %10'luk formaldehit ve Orth fiksatiflerinde daha az yoğunlukta mast hücresinin boyandığı saptandı. AB/SO boyama yönteminde ise en ideal fiksatiflerin Alkolik Formalin ve Schaudin olduğu belirlendi. Bununla birlikte uygulanan AB/SO boyama yönteminde Orth fiksatif solüsyonu ile tespit edilen dokulardan trakeyal sirinks hariç diğer tüm dokularda mast hücrelerinin sadece zayıf SO boyandığı belirlendi. Yapılan bu çalışma ile bıldırcınların sirinksinde bulunan mast hücrelerinin yerleşim yerleri, dağılımları ve heterojeniteleri de belirlendi. Sonuç olarak kullanılan tespit solüsyonlarına göre uygulanan üç boyama yönteminde de mast hücresi yoğunluklarında farklılıklar oluştuğu ortaya konulmuştur. In this thesis, it was aimed to determine the distribution and heterogeneity of mast cells in the tissue parts of the syrinx, which is a part of the respiratory system in quails. In the study, syrinx samples of 10 healthy adult quail (Coturnix coturnix) were used. The removed syrinx tissues were fixed in Bouin, 10% formaldehyde, Schaudin, Orth and Alcoholic Formalin fixative solutions, and after routine histological tissue follow-up procedures, they were embedded in paraffin. Serial sections taken at 5 micrometer thickness were applied Hematoxylin-Eosin staining method to determine the general histological structure, Standard Toluidine Blue (TB) staining method to determine the distribution of mast cells, and Thionin and Alcian Blue/Safranin O (AB/SO) staining methods to determine heterogeneity of mast cells. As a result of the staining methods applied, the histological structure of the quail syrinx, the distribution and heterogeneity of connective tissue mast cells (CTMC) and mucosal mast cells (MMC) were evaluated. It was observed that the quail syrinx was formed as a result of the modification of the last tracheal ring in the caudal part of the trachea and the first four-five rings at the beginning of the primary bronchi, and it was the trachea-bronchial type and the pessulus was ossified. It was determined that MMC formed clusters around the blood vessels and showed a denser distribution between the cartilage rings. In the applied Thionine staining method, it was determined that the most intense CTMC and MMC distribution in the tracheal and bronchial syrinx was in Alcoholic Formalin fixation. In the TB staining method, some mast cells in the tracheal and bronchial syrinx showed metachromasia. In addition, in the TB staining method, it was determined that the ideal and most intense staining was with Schaudin fixative, however, less intense mast cells were stained in 10% formaldehyde and Orth fixatives. In the AB/SO staining method, the most ideal fixatives were found to be Alcoholic Formalin and Schaudin. However, in the AB/SO staining method applied, it was determined that mast cells were only weakly stained with SO in all tissues, except for the tracheal syrinx, which were fixed with Orth fixative solution. With this study, the locations, distribution and heterogeneity of mast cells in the syrinx of quail were also determined. As a result, it was revealed that there were differences in mast cell densities in all three staining methods applied according to the fixation solutions used.
