Bu tez çalışmasında, hem normal koşullar hem de tuz stresi altında çimlendirilen Allium cepa L.'nın tohum çimlenmesi, fide büyümesi (taze ağırlık, radikula sayısı ve radikula uzunluğu), kromozom anormallikleri ve mitotik aktivite üzerine aspartik asitin (C4H7NO4) etkileri araştırılmıştır. Tek başına aspartik asitli ortamda çimlendirilen tohumların nihai çimlenme yüzdesi ile radikula uzunluğu istatiksel açıdan saf su ortamında çimlendirilen kontrol tohumlarınınkilerle aynı değerleri gösterirken, taze ağırlığı ve radikula sayısı saf su ortamında çimlendirilen kontrol tohumlarınınkilere göre kısmen artmıştır. Ayrıca, tek başına aspartik asitli ortamda çimlendirilen Allium cepa L. tohumlarının kök ucu meristemlerinde mitotik indeks saf su ortamında çimlendirilen kontrol tohumlarınınkilere göre kısmen bir azalma gösterirken, kromozom anormalliklerinin sıklığı kontrolünkilere göre aşırı derecede artış göstermiştir. Diğer taraftan, tuz stresi Allium cepa'nın fide büyümesi ve tohum çimlenmesini önemli ölçüde engellemiştir. Dahası, tuz stresi tohumların kök ucu meristematik hücrelerinde mitotik indeksi önemli ölçüde azaltmış, kromozom anormalliklerinin sayısını ise artırmıştır. Halbuki tuz stresinin kromozom anormallikleri, fide büyümesi ve tohum çimlenmesi üzerindeki engelleyici etkisi aspartik asit uygulaması ile önemli ölçüde hafifletilmiştir. Ancak, bu amino asit mitotik index üzerindeki tuz hasarının azaltılmasında etkisiz kalmıştır. In this thesis work, the effects of aspartic acid (C4H7NO4) on the seed germination, seedling growth (fresh weight, radicle number and radicle length), chromosomal aberrations and mitotic activity of Allium cepa L. germinated under both normal conditions and salt stress were investigated. The final germination percentages and radicle lengths of seeds germinated in the medium with aspartic acid alone statistically showed the same values as ones of the control seeds germinated in pure water medium, while their fresh weights and radicle numbers partly increased according to ones of the control seeds germinated in pure water medium. Furthermore, the mitotic index in root tip meristems of Allium cepa L. seeds germinated in the medium with aspartic acid alone partially demonstrated a decrease according to ones of the control seeds germinated in pure water medium while their frequency of chromosomal aberrations excessively increased according to ones of the control. Barely, salt stress significantly inhibited seedling growth and seed germination of Allium cepa. Moreover, it significantly decreased the mitotic index in the root tip meristematic cells of the seeds and increased the number of chromosomal abnormalities. However, inhibitive effect of salt stress on the chromosomal abnormalities, seedling growth and seed germination was significantly decreased with aspartic acid application. But, this amino acid was ineffective in reducing of salt damage on the mitotic index.
