İzmir kekiği (Origanum onites L.) tıbbi özellikleri olan ve Türkiye'den ihraç edilen başlıca aromatik bitki türlerinden biridir. Uçucu yağları ve terpenik bileşiklerinden dolayı antiseptik, antibakteriyel, antispazmodik, antifungal, antimikrobiyal, antiinflamatuar ve antikarsinojenik özelliklere sahip olduğu bilinmektedir. Doku kültürü tekniğini kullanarak bitki üretimi, birçok bitki türünde olduğu gibi tıbbi ve aromatik bitkilerin vejetatif olarak hızlı ve büyük miktarlarda çoğaltılmasını sağlayan bir üretim yöntemidir. Strigolaktonlar (SL), tohum çimlenmesi, kök ve sürgün yapılarının oluşumu, besin alımı, simbiyotik ilişkiler ve bitkilerin abiyotik ve biyotik sinyallere cevapları da dahil olmak üzere bitki büyümesi ve gelişimindeki düzenleyici rollerle ilişkilendirilen yeni sınıf fitohormonlardır. Bu çalışma, O. onites L. bitkisinin in vitro ortamda SL uygulamalarının kallus oluşumu ve uçucu yağ bileşenleri (karvakrol ve timol) üzerine etkisini belirlemek amacıyla yapılmıştır. Kallus kültürleri, benzilamino purin (BAP; 2.0 mg/l) ile naftalen asetik asit (NAA; 1.0 mg/l) ve çeşitli konsantrasyonlarda (0.0, 0.1, 0.5, 1.0, 2.0 ve 5.0 µM) SL içeren Murashige ve Skoog (MS) ortamında yaprak eksplantları ile başlatılarak elde edilmiştir. Deneyin her aşamasında, 2.0 mg/l BAP ve 1.0 mg/l NAA ile MS ortamına SL eklenmeden yerleştirilen eksplantlar kontrol grubunu oluşturmuştur. SL'ların farklı uygulamaları sonucu elde edilen kalluslar, kallus oluşum oranı, ağırlık, çapı, rengi ve yapısı, karvakrol ve timol gibi uçucu yağ bileşenleri ile bazı önemli fenolik bileşikler (rosmarinik asit, kuersetin, apigenin, kafeik asit, luteolin, p-kumarik asit ve viteksin) bakımından değerlendirilmiştir. Araştırma sonucunda, en fazla kallus oluşum oranı (%93.8), kallus ağırlığı (0.122 g) ve çapı (1.0 cm) 2.0 µM SL uygulamasında görülmüştür. 21 günlük kallus indüksiyonundan sonra, tüm uygulamalarda yaprak eksplantlarının sıkı bir kallus ürettiği bulunmuştur. Kallus rengi ise artan SL uygulamaları ile kahverenginden beyaza doğru açılma göstermiştir. O. onites'ten elde edilen uçucu yağ sonuçlarına göre GC-MS/SPME cihazı ile toplam 72 aromatik bileşik tespit edilmiştir. Aromatik bileşikler, çoğunlukla terpenoidler (monoterpen, seskiterpen) başta olmak üzere, hidrokarbon, aldehit, alkol, keton, karboksilik ve diğer türevleri olarak belirlenmiştir. Karvakrol (%33.02) ve timol (%49.30) en fazla bulunan terpenoidler olup, en yüksek miktarda 5.0 µM SL uygulamasında tespit edilmiştir. HPLC cihazı ile elde edilen sonuçlara göre en fazla tespit edilen fenolik bileşik, rosmarinik asit olup, toplamda 7 farklı fenolik bileşik saptanmıştır. SL uygulamaları rosmarinik asit, kafeik asit, p-kumarik asit ve viteksin fenolik bileşiklerinde artışa neden olurken, apigenin, kuersetin ve luteolin gibi bazı fenolik bileşik miktarlarında ise azalışa sebep olmuştur. Sonuç olarak, SL uygulamalarının İzmir kekiği gibi tıbbi ve aromatik bitkilerin in vitro çalışmalarında olumlu bir etkiye sahip olduğu ve kallus oluşumu, aromatik ve fenolik bileşiklerin üretimini artırmada başarılı bir şekilde kullanılabileceği görülmüştür. It is known to have antiseptic, antibacterial, antispasmodic, antifungal, antimicrobial, anti-inflammatory and anticarncinogenic properties due to its volatile oils and terpenic compounds. Plant production using the tissue culture technique is a method of production that enables medicinal and aromatic plants to be reproduced vegetatively rapidly and in large quantities, as in many plant species. Strigolactones (SL) are a new class of phytohormone that are associated with regulatory roles in plant growth and development, including seed germination, root and shoot architecture patterning, nutrient acquisition, symbiotic interactions, as well as mediation of plant responses to abiotic and biotic cues. This study was carried out to determine the effect of in vitro SL applications of O. onites L. plant on callus formation and essential oil components (carvacrol and thymol). Callus cultures were obtained by initiating leaf explant on Murashige and Skoog (MS) medium supplemented with benzylaminopurine (BAP; 2.0 mg/l) in combination with naphthalene acetic acid (NAA; 1.0 mg/l) and various concentrations (0.0, 0.1, 0.5, 1.0, 2.0 and 5.0 µM) of SLs. In each stage of the experiment, explants placed on MS media with 2.0 mg/l BAP and 1.0 mg/l NAA without the addition of SL were the control. Calluses obtained from different applications of SLs were evaluated in terms of callus formation rate, weight, diameter, color and structure, essential oil components such as carvacrol and thymol and some important phenolic compounds (rosmarinic acid, quercetin, apigenin, caffeic acid, luteolin, p-coumaric acid and vitexin). As a result of the study, the highest callus formation rate (93.8%), callus weight (0.122 g) and diameter (1.0 cm) were observed in 2.0 µM SL application. After 21 days of callus induction, it was found that all the explants produced a compact callus. Callus color, on the other hand, turned from brown to white with increasing SL applications. A total of 72 aromatic compounds were detected in the results of the essential oil obtained from O. onites according to the GC-MS / SPME device. Aromatic compounds have been identified as terpenoids (monoterpene, sesquiterpene), hydrocarbon, aldehyde, alcohol, ketone, carboxylic and other derivatives, with terpeniods being the most of all. Carvacrol (33.02%) and thymol (49.30%) were the most abundant terpenoids, and the highest amount was detected in 5.0 µM SL application. According to the results obtained with the HPLC device, the most detected phenolic compound was rosmarinic acid, and 7 different phenolic compounds were detected in total. SL applications caused an increase in rosmarinic acid, caffeic acid, p-coumaric acid and vitexin phenolic compounds, while it caused a decrease in the amount of some phenolic compounds such as apigenin, quercetin and luteolin. As a result, it has been observed that SL applications have a positive effect on in vitro studies of medicinal and aromatic plants such as Izmir Oregano, and can be used successfully to increase the production of callus formation, aromatic and phenolic compounds.
