Günümüzde cilt yaralarının tedavisi, çoklu ilaç direncine ve biyofilm oluşturma kapasitesine sahip patojenler nedeniyle oldukça zordur. Bu yaraların etkili tedavisinin sağlanabilmesi için, bakteriyel enfeksiyonu engelleyerek yara iyileşme sürecini hızlandıracak yeni ürünlere ihtiyaç duyulmaktadır. Rosa damascena Mill.; antibakteriyel, anti-kanserojen, antioksidan etkinlik de dahil pek çok biyolojik özelliği sergileyen, Türkiye ve özellikle Gül Şehri olarak bilinen Isparta için ekonomik ve kültürel değeri olan bir gül türüdür. R. damascena Mill. ürünleri olan gül yağı (GY) ve gül absolüt (GA); , antibakteriyel ve antibiyofilm potansiyellerinin yara örtüsü içerisinde değerlendirilmesi amacıyla seçilmiştir. Bu amaçla, öncelikle GY ve GA'nın antibakteriyel, antibiyofilm, antioksidan, antihemoliz ve koagülan aktiviteleri değerlendirilmiştir. Sonrasında ise yara örtüsü olarak değerlendirmek amacıyla %4, %6 ve %8 oranlarında GA içeren polilaktik asit (PLA)-jelatin (JEL) (GAPJ) nanolifler elektroeğirme tekniği ile hazırlanmıştır. PLA, JEL ve GA Amerikan Gıda ve İlaç Dairesi (FDA) onaylı ürünlerdir. Taramalı Elektron Mikroskobu (SEM) görüntülerinde nanoliflerin tek tip ve çaplarının %0 GAPJ için 305.054 ± 100.121 nm, %4 GAPJ için 357.115 ± 74.054 nm, %6 GAPJ için, 363.353 ± 296.055 nm ve %8 GAPJ için 463.414 ± 156.145 nm olduğu belirlenmiştir. Yapılan termal analizde polimerik matrikse eklenen GA miktarı arttıkça matriksin termal kararlılığının arttığı gözlemlenmiştir. PLA hidrofobik bir polimer olmasına karşın GAPJ nanoliflerdeki GA miktarı arttıkça hidrofilite artmıştır. PLA'nın su temas açısı 1570 iken içine GA eklenmesi ile bu açı 1100'ye kadar düşmüştür. %4-8 GAPJ nanoliflerden GA'nın 15 gün boyunca sürekli ve yavaş salım profiliyle salındığı gözlenmiştir. Ayrıca, %4-8 GAPJ nanoliflerin Pseudomonas aeruginosa PAO1 (%48-60) ve Enterococcus faecalis (%96-100) için biyofilm oluşumunu önemli derecede inhibe ettikleri, yüksek antioksidan aktiviteye sahip oldukları (%90-91) ve antihemolitik etkilerinin oldukça düşük olduğu (%0.2-1.7) belirlenmiştir. Sonuç olarak; GA içeren PLA-JEL nanolifler, GA içermeyen PLA-JEL nanoliflere oranla daha yüksek hidrofilite ve termal dayanıklılık kazanmıştır. GAPJ nanoliflerin iyi antibiyofilm özelliğe sahip oldukları da gösterilmiştir. GA içeren nanolif ilk defa bu tez çalışması ile üretilmiştir. Yapılan fizikokimyasal ve biyolojik analizler, GAPJ nanoliflerin cilt yaraları için biyofilm oluşumunu engelleyecek potansiyel bir yara örtüsü materyali olduğunu göstermektedir. Today, the treatment of skin wounds is very difficult due to multi-drug resistant and biofilm-forming pathogens. In order to treat these wounds effectively, there is an urgent need for new products that will potentially promote the wound healing process by inhibiting bacterial infection. Rosa damascena Mill. is a rose species that is known to have many biological properties, such as antibacterial, anti-cancer, antioxidant, and has important economic and cultural value for Turkey and particularly the province of Isparta, also known as the City of Roses. For this study, rose essential oil(RO) and rose absolute (RA), which are produced from R. damascena Mill. were selected to investigate their potential antibacterial and antibiofilm activities in wound dressing. For this purpose, firstly, antibacterial, antibiofilm, antioxidant, antihemolytic, and coagulant properties of RO and RA were evaluated. Afterward, RAPG nanofibers, containing polylactic acid (PLA), gelatin (GEL), and 4%, 6%, and 8% RA, were prepared by electrospinning to use them as wound dressings. PLA, GEL, and RA are approved by the US Food and Drug Administration (FDA). It was understood from the Scanning Electron Microscopy (SEM) images that nanofibers were uniform and the diameters were 305.054 ± 100.121 nm for %0 RAPG, 357.115 ± 74.054 nm for %4 RAPG, 363.353 ± 296.055 nm for %6 RAPG, and 463.414 ± 156.145 nm for %8 RAPG. The thermal analysis revealed that the thermal stability of the matrix increased as the amount of RA added into the polymeric matrix was increased. Although PLA is a hydrophobic polymer, the hydrophilicity was observed to have increased as the amount of RA in the RAPG nanofibers was increased. While the water contact angle of PLA was measured to be 1570, this angle decreased to 1100 with the addition of RA. The RA was observed to have been slowly and sustainably released from 4-8% RAPG nanofibers for 15 days. In addition, 4-8% RAPG nanofibers were detected to have significantly inhibited biofilm formation for Pseudomonas aeruginosa PAO1 (%48-60) and Enterococcus faecalis (96-100%), exhibited high antioxidant activity (90-91%), and exerted very low antihemolytic effects (0.2-1.7%). As a result, the RA-containing PLA-GEL nanofibers attained higher hydrophilicity and thermal stability compared to the RA-free PLA-GEL nanofibers. The RAPG nanofibers were also shown to exhibit excellent antibiofilm properties. This dissertation is the first research in which RA-containing nanofibers were produced. The physicochemical and biological analyses showed that the RAPG nanofibers can be used as potential wound dressing materials to inhibit biofilm formation in skin wounds.
