Antihistaminikler ve glukokortikoidler, alerji semptomlarının ve enflamatuar durumların tedavisinde yaygın olarak kullanılmaktadırlar. Bu çalışmada, proteinlerin sindirimi için pankreastan ince bağırsağa salgılanan tripsin ile, glukokortikoidlerden biri olan deksametazon ve bir antihistaminik olan, feniraminin in vitro bağlanma etkileşimleri, floresans emisyon spektroskopisi (FES), UV-VIS spektroskopisi, senkron floresans spektroskopisi (SFS), Dairesel dikroizm (CD) spektroskopisi ve moleküler modelleme yöntemleri kullanılarak araştırıldı. Ayrıca deksametason ve feniraminin tripsininin katalitik aktivitesi üzerindeki etkileri belirlendi. Floresan söndürme deneyleri, deksametason ve feniraminin, artan konsantrasyonları ile tripsinin içsel floresansını söndürdüğünü gösterdi. FES ve UV-VIS spektrumlarından elde edilen sonuçlar, deksametazon ve feniraminin tripsin ile etkileşimini kanıtlamaktadır. CD spektrumları, deksametason ve feniraminin, tripsin enzimin ikincil yapısını değiştirdiğini ortaya koydu. Tüm bu spektroskopi sonuçları, moleküler kenetleme yöntemleri ile de doğrulanmıştır. Her iki ilaç da tripsinin katalitik aktivitesini inhibe ettiği ve bu inhibisyonun IC50 değerleri deksametazon ve feniramin için sırasıyla 0.0049 mM ve 0.0038 mM olarak bulundu. Bu çalışmanın sonuçları, farmakokinetik ve farmakodinamik alanında değerli bilgiler sağlayabilir. Anahtar Kelimeler: Tripsin, Etkileşim, spektroskopi, floresans, kenetleme.
Antihistamines and glucocorticoids are commonly used to treat allergy symptoms and the inflammatory conditions. In present study, the in-vitro binding interactions a glucocortikoid, dexamethasone/an antihistamine, pheniramine with Trypsin secreted from pancreas to small intestine for protein digestion were investigated by fluorescence emission spectroscopy (FES), UV-VIS spectroscopy, synchronous fluorescence spectroscopy (SFS), Circular dichroism (CD) spectroscopy and molecular modeling methods. Also, the effect of these drugs on the catalytic activity of trypsin was determined. The fluorescence quenching experiments indicated that each drug quenched the intrinsic fluorescence of trypsin with their increased concentrations. The results of SFS and UV-VIS spectra proved the interaction of dexamethasone and pheniramine with trypsin. CD spectra showed that the secondary structure of enzyme was altered in the presence of the drugs. All these spectroscopy results were validated and explained by molecular docking studies. Both drugs have inhibition effect on the catalytic activity of trypsin and the IC50 values were determined as 0.0049 mM and 0.0038 mM for dexamethasone and pheniramine, respectively. The results of this study can provide valuable information in the field of pharmacokinetics and pharmacodynamics. Keywords: Trypsin; interaction, spectroscopy; fluorescence, docking.
