Günümüzde artan sağlıklı beslenme bilinci tüketicilerin fonksiyonel gıdalara olan talebini artırmaktadır. Son yıllarda artan glüten intoleransı nedeniyle sağlıklı beslenme anlayışında ekmeğin beslenmeden çıkarılması bazı uzmanlar tarafından önerilmektedir. Aslen, uzun fermantasyon ile ekşi ekmek mayası kullanılarak üretilen geleneksel ekmek besin içeriği bakımından değerli bir gıdadır. Ekşi maya, çoğunlukla buğday, çavdar veya diğer tahıl unlarının su ile karıştırılması ve bunların fermantasyonuyla üretilmektedir. Bu tez çalışmasında doğal maya ve laktik asit bakterileri içeriğiyle zengin olan su kefiri kullanılarak ekşi maya üretiminin yapılması ve kurutulması, depolanması sürecinde özelliklerinin belirlenmesi ve ekmek üretiminde duyusal özelliklerinin belirlenmesi amaçlanmıştır. Su kefiri temelli ekşi maya üretimi başarıldıktan sonra iki farklı yöntemle (liyofilize ve termal kurutma) kurutularak, farklı sıcaklıklarda (4°C ve 25°C) depolanarak 45 gün süresince mikrobiyal, kimyasal ve duyusal özellikleri tespit edilmiştir. Bu çalışmada kullanılan su kefiri danelerinden üretilen su kefirinde RT-PCR ile yapılan tanımlamada Saccaharomyces spp., Zygotorulaspora florentina, Dekkera bruxellensis ve Lachancea fermentati tespit edilmiştir. Mikrobiyal bulgular ekşi maya içeriğinde laktik asit bakterileri (LAB) ve mayaların uygun fermantasyon koşullarında optimum düzeyde sağlandığını göstermiştir. 4 °C 'de depolanan LYO örneklerinde aktifleştirme sonrasında 1. gün Lactobacillus sp.: Maya oranında 9,42 log kob/g : 8,80 log kob/g oranı sağlanmış; depolamanın 45. gününde 8,39 log kob/g : 7,65 log kob/g olarak tespit edilmiştir (P<0,05). Depolama süresince aktifleştirilmiş ekşi mayaların pH değerleri 4,27 ile 4,10 aralığında değiştiği tespit edilmiştir (P>0,05). 4 °C'de depolanan LYO örneğinin laktik asit içeriği ilk depolama gününde 2045ppm iken, aynı sıcaklıkta depolanmış KK örneğinin laktik asit içeriği 1249ppm olarak tespit edilmiştir. 25 °C'de depolanan LYO örneğinin laktik asit içeriği ilk depolama gününde 2693 ppm iken, aynı sıcaklıkta depolanmış KK örneğinin laktik asit içeriği 1232ppm olarak tespit edilmiştir. Su kefiri mikrobiyotası ile üretilmiş ekşi maya örneklerinde esterler, alkoller ve karbonil bileşenleri bakımından yoğun aroma 9 profili tespit edilmiştir. Panelistler, su kefiri mikrobiyotasıyla geliştirilen ekşi mayadan üretilen ekmeklere yüksek skorlar vermiş, ekmekler beğenilmiştir. Sonuç olarak, zengin mikrobiyotaya sahip su kefiri ile özgün içeriğe sahip ekşi maya üretimi önemli bir potansiyele sahiptir. Nowadays, increasing awareness of healthy nutrition raises the demand of consumers for functional foods. Due to the increasing gluten intolerance in recent years, some experts recommend removing bread from the diet in the understanding of healthy eating. Sourdough is used in traditional sour dough bread production. Sourdough is mostly produced by mixing wheat, rye or other grain flours with water and fermentation thereof. In this thesis, it was aimed to produce sourdough by using water kefir, which is rich in natural yeast and lactic acid bacteria, to determine its properties in the storage process and to determine its sensory performance in bread production. After the production of water kefir-based sourdough was achieved, it was dried by two different methods (lyophilized and thermal drying), stored at different temperatures (4°C and 25°C), and its microbial, chemical and sensory properties were determined for 45 days. Saccaharomyces spp., Zygotorulaspora florentina, Dekkera bruxellensis and Lachancea fermentati were detected using RT-PCR in water kefir produced from water kefir grains used in this study. Microbial findings showed that lactic acid bacteria (LAB) and yeasts in the sourdough content were optimally provided under suitable fermentation conditions. In LYO samples stored at 4 °C, on the 1st day after activation, Lactobacillus sp. : yeast ratio 9.42 log cfu/g in: 8.80 log cfu/g; on the 45th day of storage it was 8.39 log cfu/g: 7.65 log cfu/g (P<0.05). It was determined that the pH values of the activated sourdoughs during storage ranged between 4.27 and 4.10 (P>0.05). While the lactic acid content of the LYO sample stored at 4 °C was 2045ppm on the first storage day, the lactic acid content of the KK sample stored at the same temperature was 1249ppm. While the lactic acid content of the LYO sample stored at 25 °C was 2693 ppm on the first storage day, the lactic acid content of the KK sample stored at the same temperature was 1232 ppm. Intense aroma profile was detected in terms of esters, alcohols and carbonyl compounds in sourdough samples produced with water kefir microbiota. The panelists gave high scores to the sourdough breads developed with the water kefir microbiota, and the breads were appreciated. 11 As a result, production of sourdough that has unique content due to rich microbiota of water kefir has an important potential.
