Staphylococcus aureus (S. aureus) insan ve hayvanlarda gıda kaynaklı ciddi enfeksiyonlara neden olabilen önemli patojenlerden birisidir. Açık pazarlarda satışa sunulan süt ve süt ürünlerinin tüketimi S. aureus kontaminasyonu açısından önemli risklere sahiptir. Antibiyotik direncine sahip S. aureus suşları ise, insanları ve hayvanları kolonize etme ve enfekte etme kabiliyeti nedeniyle ciddi bir halk sağlığı sorunu teşkil eder. Bu çalışmada, Isparta ilinde açık pazarlarda satışa sunulan 100 adet süt ve süt ürünü örnekten, S. aureus izolasyonu gerçekleştirilmiştir. İzolatların fenotipik karakterizasyonu için Gram boyama, oksidaz, katalaz, mannitol, koagülaz, lateks aglütinasyon testi ve DNaz testleri yapılmıştır. S. aureus izolatlarının moleküler karakterizasyonu spa geni için Real time PCR ile gerçekleştirilmiş ve izolatların 17 antibitotiğe karşı dirençleri disk difüzyon yöntemi kullanılarak ve biyofilm oluşturma kapasiteleri spektrofotometrik olarak değerlendirilmiştir. Genel olarak 100 süt ve süt ürünü örneğinin 23 tanesinin spa pozitif olduğu belirlenmiştir. S. aureus, 12 adet çökelek örneği (%52.2), 5 adet çiğ süt örneği (%21.7), 2 adet kaşar peyniri örneği (%8.7) ve 4 adet meyveli dondurma örneğinden (%26.1) izole edilmiştir. İzolatların 17 antibiyotik için Okzasiline %100 oranında ve Fusidik asite %47,8 oranında dirençli olduğu, diğer 15 antibiyotiğe ise izolatların %95.65-100 oranında duyarlı olduğu belirlenmiştir. Bu çalışmada izolatlar arasında düşük bir antibiyotik direnci prevalansı belirlenmesine rağmen, bu çalışmanın sonuçları, çiğ sütün işlenmesi veya kontamine süt ürünlerinin tüketimi ile S. aureus bağlantılı enfeksiyon riskinin göz ardı edilmemesi gerektiğini açıkça göstermektedir. Staphylococcus aureus (S. aureus) is one of the most important pathogens that can cause serious foodborne infections in humans and animals. Consumption of milk and dairy products for sale in open markets carries significant risks of S. aureus contamination. Antibiotic-resistant S. aureus strains pose a serious public health problem due to their ability to colonize and infect humans and animals. In this study, S. aureus was isolated from 100 milk and dairy product samples sold in open markets in Isparta Province. For phenotypic characterization of the isolates, Gram stain, oxidase, catalase, mannitol, coagulase, latex agglutination test and DNase tests were carried out. Molecular characterization of S. aureus was carried out by Real time PCR for spa gene, resistance of isolates to 17 antibiotics was determined using disc diffusion method and their ability to form biofilms was evaluated using spectrophotometric method. Overall, 23 out of 100 milk and milk product samples (23%) were found to be positive for spa gene. S. aureus was isolated from 12 (52.2%) of cottage cheese, 5 (21.7%) of raw milk, 2 (8.7%)cheddar cheese and 4 (26.1 %) of fruit ice cream samples. The isolates were found to be 100% resistant to oxacillin and 47.8% to fusidic acid for 17 antibiotics, while the isolates were sensitive to 95.65-100% of the other 15 antibiotics. And most of the isolates (82.6 %) possessed moderate or weak and 4 isolates (17.4%) strong biofilm-formation ability. Although we found a low prevalence of antibiotic resistance in isolates in this study, the results of this study clearly indicate, that the risk of S. aureus-associated infection should not be ignored when processing raw milk or consuming contaminated dairy products.
