Tüm dünyada probiyotik mikroorganizmaların sağlıklı etkileri belirlenmesiyle bu tür mikroorganizmaların alternatif kullanımları artmaktadır. Peynir çok yüksek besleyici -özellikle protein ve mineral- içeriğe sahip olması sebebiyle, çok geniş bir tüketici grubunun tercihi olan bir süt ürünüdür. Peynir yapımında kullanılan süt çeşitliliklerinin kullanımı, çiğ süt kalitesindeki farklılıklar, kullanılan kültür ve enzim çeşitliliği, üretim yönteminde ve olgunlaştırma koşullarındaki farklılıklardan dolayı dünyanın her yerinde üretilen binlerce peynir çeşidi vardır. Bu peynir çeşitlerinden biri de İtalya'da manda sütü kullanılarak üretilen pasta filata grubunda yer alan ve dolgulu bir peynir olan Burrata peyniridir. Bu araştırmanın amacı, Lacticaseibacillus casei ATCC 393'ün peynir ürününde kullanımıdır. Burrata peyniri taze ve dolgulu bir peynirdir ve bu sebep tercih edilmiştir. Burrata peynirin ısıl işlem uygulanmayan iç dolgusu Lacticaseibacillus casei ATCC 393 ile hazırlanarak (LC-BC) ve kontrol grubu (C-BC) peynirler üretilmiştir. Üretilen Burrata peynirlerinde 21 günlük depolama süresince fizikokimyasal (titrasyon asitliği analizi, pH, kuru madde tayini, yağ tayini, tuz tayini, toplam azot, suda çözünen azot, olgunlaşma indeksinin'nin belirlenmesi, su aktivitesi analizi), renk (CIE), mikrobiyolojik (laktik asit bakteri sayımı, M17 sayımı, toplam mezofil bakteri sayımı, maya ve küf sayımı, koliform grubu bakteri sayımı, toplam psikrofil bakteri sayımı, proteolitik bakteri sayımı ve lipolitik bakteri sayımı) analizler, aroma bileşenlerinin kantitatif olarak belirlenmesi (Gaz kromatografisi) ve duyusal analizlerin değerlendirilmesi ile ürünler arasında değişimler belirlenmiş ve yorumlanmıştır. Araştırmamızda Burrata peynirine Lacticaseibacillus casei ilavesinin 1. günde pH değerini azalttığı ancak raf ömrü süresince pH aralığını (5.80-5.22) koruduğu belirlenmiştir. Depolama süresince su aktivitesi, tuz, kurumadde, yağ, toplam azot, suda çözünen azot, olgunlaşma indeksi, protein, L*, a*,b* değerlerinde istatistiksel olarak anlamlı bir değişiklik gözlenmemiştir. LC-BC ve C-BC örneklerinin Lactobacillus spp. sayısı 1. gün değerleri sırasıyla 8.62 log kob/g ve 7.26 log kob/g olarak tespit edilmiş, 21 gün raf ömrü süresince laktik asit bakterilerinin anlamlı düzeyde arttığı belirlenmiştir (p>0.05). Burrata peynirlerinde her iki grupta araştırılan mezofilik bakteri, koliform, maya-küf, M17 bakterileri, psikrofilik bakteri, proteolitik bakteri, lipolitik bakteri sayısında de istatistiksel olarak anlamlı bir farklılık gözlenmemiştir. İki peynir arasındaki farklılığı esas belirleyen bileşenler LC-BC örneğinde diasetil, C-BC örneğinde ise butanoik asittir. Diasetil miktarının yüksek olması nedeniyle LC-BC örneğinde kremsi, hafif tatlı ve tereyağımsı aroma daha öne çıkmaktadır. Burrata peynirlerinin üretiminde Lacticaseibacillus casei kullanımı ürünün organoleptik özelliklerini olumlu etkilediği belirlenmiştir. Sonuç olarak Burrata peynirinin iç dolgusuna ilave edilen Lacticaseibacillus casei subsp. casei ATCC 393 peynir kalitesine, duyusal kabul edilebilirliğine ve raf ömrüne olumlu katkı sağlamıştır. Yapılan analizlerle Burrata peynirinin probiyotik kullanımına uygun bir peynir olduğu gösterilmiş ve literatüre katkı sağlanmıştır. Cheese is a dairy product preferred by a wide range of consumers, as it has a very high nutritive content especially protein and mineral. There are thousands of cheese varieties produced all over the world, such as the variety of milk, differences in raw milk quality, diversity of cultures and enzymes used, differences in production method and ripening conditions. One of these cheese types is Burrata cheese, which is a filled cheese in the pasta filata group produced using buffalo milk in Italy. The purpose of this research is the use of Lacticaseibacillus casei ATCC 393 in cheese product. Burrata cheese is fresh and filled cheese and it was chosen for this reason. The non-heat treated inner filling of Burrata cheese was prepared with Lacticaseibacillus casei ATCC 393 (LC-BC) and control group (C-BC) cheeses were produced. Physicochemical (titration acidity analysis, pH, dry matter determination, fat determination, salt determination, total nitrogen, water-soluble nitrogen, determination of ripening index, water activity analysis), color (CIE), microbiological (lactic acid bacteria count, M17 count, total mesophyl bacteria count, yeast and mold count, coliform group bacteria count, total psychrophilic bacteria count, proteolytic bacteria count and lipolytic bacteria count) analyzes, quantitative determination of aroma components (Gas Chromatography) and evaluation of sensory analyzes changes were identified and interpreted. In our study, it was determined that the addition of Lacticaseibacillus casei to Burrata cheese reduced the pH value at the 1st day, but maintained the pH range (5.80-5.22) during the shelf life. Water activity, salt, dry matter, oil, total nitrogen, water-soluble nitrogen, ripening index, protein, L*, a*, b* values were observed no statistically significant changes during storage. Lactobacillus spp. of LC-BC and C-BC samples values were determined as 8.62 log cfu/g and 7.26 log cfu/g, respectively at the 1st day and it was determined that lactic acid bacteria increased significantly during the shelf life (p>0.05). The number of mesophilic bacteria, coliform, yeast-mold, M17 bacteria, psychrophilic bacteria, proteolytic bacteria, and lipolytic bacteria wasn't observed statistically significant difference in both of Burrata cheeses groups. The main components that the difference between the two cheese group are diacetyl in the LC-BC sample and butanoic acid in the C-BC sample. Due to the high amount of diacetyl, creamy, slightly sweet and buttery aroma is more prominent in the LC-BC sample. It has been determined that the use of Lacticaseibacillus casei in the Burrata cheese production affects positively the organoleptic properties. As a result, Burrata cheese production with Lacticaseibacillus casei subsp. casei ATCC 393 was contributed positively to cheese quality, sensory acceptability and shelf life.
