Çalışmamızda biyobenzetim modelleme ile farklı dozlarda uygulanan adiponektinin klinik yönden değerlendirildiklerinde ilerleyiş, yayılış ve tedavi açısından farklılıklar gösteren iki meme kanseri tipinde Toll Benzeri Reseptörler (TLR) üzerine/üzerinden yaptığı etkilerin histolojik olarak tanımlanması ve bu tanımlanan etkilerin genetik ve biyokimyasal olarak da araştırılması amaçlandı. Bu çalışmada hücre hatları American Type Culture Collection (ATCC)'dan temin edildi. MCF-7 (östrojen reseptörü pozitif) ve MDA-MB-231 (östrojen reseptörü negatif) meme kanseri hücre hatları kullanılarak; pasajlama ve çoğaltılma işlemleri gerçekleştirildi. Sayısı ve canlılık oranları belirlenen hücre hatları üzerinde doz belirleme deneyi yapıldı. Doz belirleme deneyi sonucuna göre belirlenen uygun adiponektin dozları MCF-7 ve MDA-MB-231 hücre hatlarına uygulandı. Daha sonra iki boyutlu MCF-7 ve MDA-MB-231 kültürlerinde bromodeoksiüridin, kaspaz-3, TLR3 ve TLR4 ile immünohistokimyasal işaretleme gerçekleştirildi. Histolojik farklılıkların gen ekspresyonu yöntemi ile reseptör bağlantısının araştırılması yapıldı ve metabolik yolaklarda artan/azalan proteinler kantitatif olarak belirlendi. Ayrıca histolojik ve gen ekspresyon yöntemi ile gösterilen yolak ve metabolitlere ait proteinlerin düzeyleri ELISA yöntemi ile kantitatif olarak ölçüldü. Deneyler sonunda tüm saatlerde kontrol gruplarındaki hücreler normal olarak çoğalmaya devam ederken, adiponektinin MCF-7 ve MDA-MB-231 meme kanseri hücrelerinin proliferasyonunu belirgin olarak azalttığı gözlendi. Kaspaz-3 ile yapılan immünohistokimyasal boyama deneylerinde; tüm saatlerde kontrol grubunda Kaspaz-3 ile işaretlenmenin deney grubuna göre daha az olduğu görüldü. TRL3 ve TRL4 ile yapılan immünohistokimyasal çalışma sonunda; tüm saatlerde kontrol grubunda deney grubuna göre daha az hücrenin TRL3 ve TRL4 ile işaretlendiği görüldü. Adiponektin ile farklı konsantrasyonlarda (100-500 ng/mL) muamele edilen MCF-7 ve MDA-MB 231 hücre kültürü hatlarında AdipoR1 gen ekspresyon seviyeleri RT-PCR ile analiz edilip, istatistiksel değerlendirmesi yapıldığında MCF-7 hücre hattında AdipoR1 gen ekspresyonu seviyesinin artan adiponektin konsantrasyonuna bağlı olarak artış gösterdiği belirlendi. MDA-MB 231 hücre hattında AdipoR1 gen ekspresyonu artışı sadece 48. saatteki 250 ng/mL adiponektin uygulamasında kontrol grubuna göre istatistiksel olarak anlamlı bulundu. MCF-7 hücre hattında CDK4 ölçümleri, 250 ve 500 ng/mL adiponektin uygulaması yapılanlarda ise kontrole göre düşük seviyede bulundu (p<0,05). MDA-MB 231 hücre hattında 250 ve 500 ng/mL adiponektin uygulaması yapılanlarda ise kontrole göre düşük seviyede bulunmasına rağmen kontrole göre CDK4 seviyesindeki azalış sadece 500 ng/mL uygulama yapılan kültürlerde istatistiksel olarak anlamlı bulundu (p<0,05). 250 ng/mL adiponektin ile muamele edilen MCF-7 hücre hattında CDK6 seviyeleri kontrole göre düşük bulundu ve bu azalma istatistiksel olarak da anlamlıydı (p<0,05). MDA-MB-231 hücre hattında tüm çalışılan saat ve adiponektin konsantrasyonlarında CDK6 seviyeleri kontrole göre istatistiksel olarak anlamlı düzeyde yüksek bulundu (p<0,05). Sonuç olarak bu çalışmadan elde edilen veriler, hücre döngüsü durumuyla ilgili olarak hem MCF-7 ve hem de MDA-MB-231 meydana gelen olayların olası sinyalleşme olay dizisinin çalışan bir modelini geliştirmek için yeterli bilgi sağlamıştır. Adiponektin tedavisi AdipoR1 seviyesini artırmak için AMPK'yi etkinleştirebilirsek, tümör için daha olumsuz büyüme mikro ortamı oluşturabiliriz. Adiponektin ve TLR'ler arasındaki ilişkinin tam olarak anlaşılması, obezite, meme kanseri ve hücre döngüsü düzenlemesi arasındaki fizyolojik/moleküler ilişki arasında köprü oluşturabilir. In our study, it was aimed to define histologically the effects of adiponectin applied at different doses on/by Toll-Like Receptors (TLR) in two breast cancer types that differ in terms of progression, spread and treatment when evaluated clinically, and to investigate these effects genetically and biochemically. In this study, cell lines were obtained from the American Type Culture Collection (ATCC). Using MCF-7 (estrogen receptor positive) and MDA-MB-231 (estrogen receptor negative) breast cancer cell lines; Passaging and duplication processes were carried out. Dose determination experiment was performed on cell lines whose number and viability were determined. Appropriate adiponectin doses determined according to the results of the dose determination experiment were applied to MCF-7 and MDA-MB-231 cell lines. Then, immunohistochemical marking was performed with bromodeoxyuridine, Caspase-3, TLR3 and TLR4 in two-dimensional cultures of MCF-7 and MDA-MB-231. The histological differences were investigated by gene expression method and the receptor linkage was investigated and the increased/decreased proteins in the metabolic pathways were determined quantitatively. In addition, the levels of proteins belonging to the pathway and metabolites shown by histological and gene expression methods were quantitatively measured by ELISA method. At the end of the experiments, while the cells in the control groups continued to proliferate normally at all hours, it was observed that adiponectin significantly reduced the proliferation of MCF-7 and MDA-MB-231 breast cancer cells. In immunohistochemical staining experiments with Caspase-3; Caspase-3 labeling was observed to be less in the control group at all hours compared to the experimental group. At the end of the immunohistochemical study with TRL3 and TRL4; It was observed that fewer cells were marked with TRL3 and TRL4 in the control group compared to the experimental group at all hours. AdipoR1 gene expression levels in MCF-7 and MDA-MB 231 cell culture lines treated with adiponectin at different concentrations (100-500 ng/mL) were analyzed by RT-PCR and statistically evaluated was found to increase with increasing concentration. The increase in AdipoR1 gene expression in MDA-MB 231 cell line was found to be statistically significant only in the 48th hour implementation of 250 ng/mL adiponectin compared to the control group. Statistically significant was found only in the 48th hour cultures of the cultures treated with 250 ng/mL in MDA-MB 231 cell line. CDK4 measurements in the MCF-7 cell lines were found to be lower in those treated with 250 and 500 ng/mL adiponectin compared to the control (p<0.05). Although it was found at lower levels in MDA-MB 231 cell lines in those treated with 250 and 500 ng/mL adiponectin compared to the control, the decrease in CDK-4 level was found to be statistically significant only in cultures treated with 500 ng/mL (p<0.05). CDK6 levels were found to be lower in MCF-7 cell line treated with 250 ng/mL adiponectin compared to control and this decrease was statistically significant (p<0.05). CDK6 levels in MDA-MB-231 cell line at all working hours and adiponectin concentrations were found to be statistically significantly higher than the control (p<0.05). In conclusion, the data from this study provided sufficient information regarding cell cycle status to develop a working model of the possible signaling cascade of events occurring in both MCF-7 and MDA-MB-231. Adiponectin treatment if we can activate AMPK to increase AdipoR1 level, we can create a more unfavorable growth microenvironment for the tumor. A thorough understanding of the relationship between adiponectin and TLRs can bridge the physiological/molecular relationship between obesity, breast cancer, and cell cycle regulation.
