<p class="MsoNormal" style="margin-bottom:0cm;margin-bottom:.0001pt;text-align:justify;text-justify:inter-ideograph;line-height:normal"><span style="font-size:12.0pt;font-family:&quot;Cambria&quot;,serif;mso-ascii-theme-font:major-latin;mso-hansi-theme-font:major-latin">Mikotoksinler; Aspergillus, Penicillium gibimantar cinslerinin sekonder metabolizması sonucu oluşan doğal toksinlerdir.İnsan sağlığı açısından en önemli mikotoksinlerden olan Okratoksin A (OTA)buğday, arpa, mısır gibi tahıl ürünlerinin yanısıra ekmek, bazlama, pide gibitahıl ürünlerinde de bulunabilir. Bu çalışmada, ekşi mayalı bazlama üretimindefarklı fermentasyon süresinin, pişirmenin ve farklı sürelerde depolamanın OTAvarlığına etkisinin belirlenmesi amaçlanmıştır. Bu kapsamda bazlama üretimindeOTA içermeyen un, 5 µg/kg ve 10 µg/kg OTA konsatrasyonunda kirletilmiş un veekşi maya kullanılmıştır. Bu unlardan hazırlanan bazlama hamurlarına uygulananfarklı fermentasyon süresi (1.5 ve 3 saat), 300±5⁰C’ de pişirme ve odaşartlarında (25±2⁰C’de) 0, 5 ve 10 gün depolama işlemleri sonrası OTAseviyesindeki değişim floresans dedektörlü yüksek performanslı sıvıkromatografisi (HPLC-FLD) yöntemi ile belirlenmiştir. Bazlama üretiminde 5µg/kg OTA ile kontamine edilmiş undan elde edilen hamurların OTA miktarı 3.691µg/kg, 1.5, ve 3 saat fermentasyon işlemleri sonrası bu değer sırasıyla 3.537µg/kg ve 3.363 µg/kg, pişirme sonrası ise 3.114 µg/kg ve 2.963 µg/kg’ olaraksaptanmıştır. 10 µg/kg OTA ile kontamine edilmiş undan elde edilen hamurların,1.5 saat ve 3 saat fermentasyona bırakılması sonrası hamurların ve buhamurlardan pişirilen bazlamaların OTA miktarları ise sırasıyla 6.444 µg/kg,5.784 µg/kg, 5.259 µg/kg, 5.376 µg/kg ve 4.595 µg/kg olarak ölçülmüştür. Farklıdepolama sürelerinin OTA varlığı üzerindeki etkisi önemsiz bulunmuştur. Elde edilenbulgular sonucunda OTA seviyesinde genel olarak bir azalma tespit edilse deözellikle yüksek konsantrasyonda (10 µg/kg) OTA bulaşısı olan unlarda uzunsüreli fermentasyonun (en az 3 saat) daha etkili olduğu tespit edilmiştir. Uzunsüreli fermentasyon uygulanmadığı takdirde bazlama ekmeği üretimi esnasında hem5 µg/kg hem de 10 µg/kg OTA ile kontamine edilmiş unlardan üretilenbazlamaların Türk Gıda Kodeksi ve Avrupa Komisyonu tarafından kabul edilebilirmaksimum limit olan 3 µg/kg’ ın üzerinde OTA bulundurduğu ve bu şekildetüketime uygun olmadığı belirlenmiştir.<o:p></o:p></span></p><p class="MsoNormal" style="margin-bottom:0cm;margin-bottom:.0001pt;text-align:justify;text-justify:inter-ideograph;line-height:normal"><span style="font-size:12.0pt;font-family:&quot;Cambria&quot;,serif">Mycotoxins are naturaltoxins formed as a result of secondary metabolism of fungal genera such asAspergillus and Penicillium. Ochratoxin A (OTA), which is one of the mostimportant mycotoxins in terms of human health, can be found in cereal productssuch as bread, “bazlama” (traditional flatbread), and pita bread as well as cerealssuch as wheat, barley, and corn. This study, it was aimed to determine theeffect of different fermentation times, baking, and storage for differentperiods on the presence of OTA in sourdough bazlama production. In thiscontext, OTA-free flour (control), flour contaminated with OTA concentrationsof 5 µg/kg and 10 µg/kg and sourdough have been used. After differentfermentation times (1.5 and 3 hours), baking at 300±5⁰C and storage at roomconditions (25±2⁰C) for 0, 5 and 10 days, the change in OTA level has beendetermined by high-performance liquid chromatography with fluorescence detector(HPLC-FLD) method. The OTA content of the dough obtained from flour contaminatedwith 5 µg/kg OTA in bazlama production was 3.691 µg/kg, 3.537 µg/kg, and 3.363µg/kg after fermentation for 1.5 and 3 hours, respectively, and 3.114 µg/kg and2.963 µg/kg after baking. The OTA contents of doughs obtained from flourcontaminated with 10 µg/kg OTA, doughs left for fermentation for 1.5 hours and3 hours and bazlama samples baked from these doughs were 6.444 µg/kg, 5.784µg/kg, 5.259 µg/kg, 5.376 µg/kg and 4.595 µg/kg, respectively. The effect ofdifferent storage periods on the presence of OTA has been found to beinsignificant. As a result of the findings, although a general decrease in OTAlevel has been determined, it has been found that long-term fermentation (atleast 3 hours) was more effective especially in flours with high concentration(10 µg/kg) of OTA contamination. As long as long-term fermentation has not beenapplied, it has been determined that the bazlama produced from flours to whichboth 5 µg/kg and 10 µg/kg OTA were added during the production of bazlamacontained OTA above 3 µg/kg, which is the maximum acceptable limit by theTurkish Food Codex and the European Commission and is not suitable forconsumption in this way.<o:p></o:p></span></p>
