Bu çalışmanın amacı tulum peynirinden izole edilen Enterococcus faecalis MBE1-9 suşu tarafından üretilen enterosinin karakterizasyonu, bu suşun teknolojik, probiyotik özellikleri ve suşun güvenilirliğinin araştırılmasıdır. Çalışma kapsamında öncelikle E. faecalis MBE1-9 suşunun farklı indikatör bakterilere karşı aktivite spektrumları kuyu difüzyon ve nokta ekim yöntemleri ile belirlenmiştir. Enterosinin aktivitesinin pH 4-8 aralığında stabil kaldığı, pH 2 ve 3'te 2 katına çıktığı, pH 9 ve 10'da %50 azaldığı gözlemlenmiştir. Enzim denemelerinde ise katalaz, lizozim, lipaz ve α- amilaz enzimleri varlığında enterosinin aktivitesinin stabil kaldığı, tripsin ve pepsin enzimleri varlığında aktivitesinin %50 oranında azaldığı, proteinaz K ve α-kemotripsin enzimleri ile de aktivitesinin kaybolduğu gözlemlenmiştir. Amonyum sülfat ile kısmi saflaştırılması yapılan enterosinin Trisin-SDS PAGE yöntemi ile aktif protein bandının moleküler büyüklüğü yaklaşık 5 kDa olarak belirlenmiştir. MBE1-9 suşunun yavaş asit üreticisi olduğu, hücre dışı proteolitik ve lipolitik aktivite göstermediği gözlemlenmiştir. API® ZYM testi ile MBE1-9 suşunun proteaz aktivitesi düşük, peptidaz aktivitesi ise yüksek bulunmuştur. MBE1-9 suşunun safra tuzuna dayanıklı olduğu tespit edilmiştir. E. faecalis MBE1-9 suşunun 4. ve 24. saat sonundaki otoagregasyon yüzdeleri sırasıyla 17.70±0.57 ve 41.05±0.14 olarak hesaplanmıştır. E. faecalis MBE1-9 suşunun patojen süspansiyonlarıyla 24 saat inkübasyonu sonucu hesaplanan %koagregasyon değerleri E. coli ATCC 25922 suşu ile 41.29±1.66, L. monocytogenes ATCC 19115 suşu ile 40.14±0.48, S. Typhimurium ATCC 14028 suşu ile 37.31±1.51 ve S. aureus ATCC 43300 suşu ile 33.22±0.66 olarak belirlenmiştir. E. faecalis MBE1-9 suşunun lizozim uygulaması sonrası lizozime dirençli olduğu gözlemlenmiştir. E. faecalis MBE1-9 suşu 3 saat ve 24 saat inkübasyon süresi sonunda %0.4 fenol varlığında canlılığını korumuştur. E. faecalis MBE1-9 suşu klinik olarak önemli birçok antibiyotiğe duyarlı bulunmuştur. E. faecalis MBE1-9 suşunda, aac(6')-Ie- aph(2'')-Ia (gentamisin), aph(3')-IIIa (streptomisin), ermB genleri tespit edilmiştir. MBE1-9 suşunun fenotipik olarak hemolitik aktivite ve jelatinaz aktivitesi göstermediği belirlenmiştir. E. faecalis MBE1-9 suşunda gelE, cpd, cob, ccf ve ace genleri belirlenmiştir. MBE1-9 suşunun efaAfm, efaAfs, espfm, espfs, cad, acm, agg ve hyl genlerini ise içermediği belirlenmiştir. MBE1-9 suşunun fenotipik olarak biyojen amin üretmediği belirlenmiş dekarboksilaz genleri varlığı tespit edilmemiştir. Kısmi saflaştırılması yapılmış bakteriyosinin ve E. faecalis MBE1-9 canlı kültürünün UHT süt ortamında L. monocytogenes'in gelişimi üzerine etkisinin 4°C'de depolama koşullarında test edildiği denemeler sonucu depolamanın 7. günü sonunda kontrol grubuna kıyasla saflaştırılmış bakteriyosin ve canlı kültür içeren deneme gruplarında L. monocytogenes hücre sayısında sırasıyla 0.41 log ve 1.13 log inhibisyon olduğu tespit edilmiştir. L. monocytogenes hücre sayısının 20°C depolama koşullarında saflaştırılmış bakteriyosin içeren deneme grubunda kontrol grubuna kıyasla depolamanın 7. günü sonunda 2.1 log inhibisyona uğradığı belirlenmiştir. Saflaştırılmış bakteriyosin içeren süt örneklerinin 37°C'de inkübasyonunda edilen L. monocytogenes sayısı 1. günde 4.44 log, 7. günde ise 0.59 log inhibisyona uğradığı tespit edilmiştir. E. faecalis MBE1-9 canlı kültürü içeren süt örneklerinde ise 1. günden itibaren inkübasyonun devam ettiği 7 gün boyunca L. monocytogenes hücre sayısının tespit edilemeyecek (<1 log kob/mL) sayıya düştüğü belirlenmiştir. E. faecalis MBE1-9 suşunun güvenlik değerlendirmesi sonucu canlı kültür olarak gıda üretim süreçlerinde kullanılmasının tüketici sağlığı açısından risk oluşturabileceği belirlenmiştir. Ancak bu suş tarafından üretilen enterosinin saflaştırılmış preparatının gıda muhafazası amacıyla kullanım potansiyelinin olabileceği düşünülmektedir. The aim of this study was to characterize enterocin produced by Enterococcus faecalis MBE1-9 strain isolated from Tulum cheese, to investigate technological and probiotic properties of this strain and safety of the strain. Within the scope of the study, activity spectrum of E. faecalis MBE1-9 strain against different indicator bacteria were determined by well diffusion and spot agar methods. It was observed that activity of enterocin remained stable at pH 4-8, doubled at pH 2 and 3, and decreased by 50% at pH 9 and 10. In enzyme tests, it was observed that activity of enterocin remained stable in presence of catalase, lysozyme, lipase and α-amylase enzymes, its activity decreased by 50% in presence of trypsin and pepsin enzymes, and its activity was lost with proteinase K and α-chemotrypsin enzymes. The molecular size of the active protein band was determined to be approximately 5 kDa by Trisin-SDS PAGE method of enterocin, which was partially purified with ammonium sulfate. It was observed that MBE1-9 strain was a slow acid producer and did not show extracellular proteolytic and lipolytic activity. With API® ZYM test was found, the protease activity of MBE1-9 strain was found low and the peptidase activity high. MBE1-9 strain was found resistant to bile salt. Autoaggregation percentages of E. faecalis MBE1-9 at the end of the 4th and 24th hours were calculated as 17.70±0.57 and 41.05±0.14, respectively. Coaggregation values (%) calculated as a result of incubation of E. faecalis MBE1-9 strain with pathogen suspensions for 24 hours, with E. coli ATCC 25922 41.29±1.66, L. monocytogenes ATCC 19115 40.14±0.48, S. Typhimurium ATCC 14028 37.31± 1.51 and 33.22±0.66 with S. aureus ATCC 43300. It was observed that E. faecalis MBE1-9 strain was resistant to lysozyme. E. faecalis MBE1-9 strain remained viable in the presence of 0.4% phenol at the end of 3 hour and 24 hour incubation periods. E. faecalis MBE1-9 strain was found sensitive to many clinically important antibiotics. aac(6')-Ie-aph(2'')-Ia (gentamycin), aph(3')-IIIa (streptomycin), ermB genes were detected in E. faecalis MBE1-9 strain. It was determined that MBE1-9 strain did not show phenotypically hemolytic activity and gelatinase activity. gelE, cpd, cob, ccf and ace genes were detected in E. faecalis MBE1-9 strain. Decarboxylase genes were not detected in MBE1-9 strain, which were determined not to produce biogenic amines phenotypically. As a result of the trials in which the effect of partially purified bacteriocin and E. faecalis MBE1-9 live culture on growth of L. monocytogenes in UHT milk medium was tested at 4°C storage conditions, the trial containing purified bacteriocin and live culture at the end of the 7 day of storage compared to the control group. It was determined that there was 0.41 log and 1.13 log inhibition in L. monocytogenes cell number in the groups, respectively. It was determined that number of L. monocytogenes cells in the experimental group containing purified bacteriocin at 20°C storage conditions was 2.1 log inhibited at the end of the 7 day of storage compared to the control group. It was detected that the number of L. monocytogenes was inhibited 4.44 log on the 1 day and 0.59 log on the 7 day in the milk samples containing purified bacteriocin incubated at 37°C. In milk samples containing E. faecalis MBE1-9 live culture, it was determined that the number of L. monocytogenes cells decreased to an undetectable (<1 log cfu/mL) number during the 7 days of incubation from 1 day. As a result of safety evaluation of the and E. faecalis MBE1-9 strain, it was determined that use of it as a live culture in food production processes may pose a risk in terms of consumer health. However, it is thought that purified preparation of enterocin produced by this strain may have potential of food preservation.
