Gilaburu (Viburnum opulus L.) meyvesi içerdiği besin öğeleri ve biyoaktif bileşenler açısından oldukça zengin bir meyvedir. İçeriğinde bulunan biyoaktif bileşenlerden klorojenik asit, L-malik asit, antosiyaninler ve karotenoidlerin sağlık üzerine birçok olumlu etkileri (antidiyabetik, antioksidan ve antimikrobiyal) bulunmaktadır. Bu çalışmada, yanıt yüzey metodu (RSM) ve Box-Behnken deney tasarımı ile Gilaburu meyvesinin biyoaktif bileşenlerinden fenolik maddelerin mikrodalga destekli ekstraksiyonunun (MDE) optimize edilmesi amaçlanmıştır. Gilaburu meyvesinin maksimum ekstraksiyon verimi (%) ve toplam fenolik madde içeriği (mg GAE/g ekstrakt) optimize edildiğinde, sıcaklık, süre ve katı/çözücü oranı optimum değerleri sırasıyla 60 (°C), 5 dakika, 4.18 g/100 mL olarak bulunmuştur. RSM tasarımında belirlenen optimum parametreler kullanılarak üretilen sulu ekstraktın polifenol kompozisyonu, serbest radikalleri yakalama ve antidiyabetik aktivitesi tespit edilmiştir. Ayrıca, MDE'den elde edilen fenolik bileşiklerce zengin ve suda çözünür ekstraktın enkapsülasyonunda, iyonik jelasyon ve kitosan sistemi kullanılmıştır. Elde edilen hidrojel boncukların nem içeriği ve enkapsülasyon etkinliği ise sırasıyla %53.52 ve %91.26 olarak bulunmuştur. Bunun yanında hidrojel boncuklara da ekstrakt için belirlenen analizler uygulanmıştır. Yapılan HPLC analizi sonucunda hem ekstraktın hem de hidrojel boncuğun yüksek miktarda klorojenik asit içerdiği tespit edilmiştir ve bu değerler sırasıyla 34.18 mg/g ekstrakt ve 0.51 mg/g ekstrakt (56.45 g hidrojel boncuk) olarak bulunmuştur. Optimum şartlarda elde edilen ekstraktın α-glikozidaz aktivitesini %54, hidrojel boncuğun ise %99.9 inhibe ettiği tespit edilmiştir. Elde edilen sonuçlara göre gilaburu meyve fenoliklerinin yeşil ektraksiyon tekniklerinden mikrodalga ile ekstraksiyonu sonucunda kısa sürede, düşük enerji ihtiyacı ve maksimum verimle elde edildiği görülmüştür. Ayrıca, gilaburu meyvesinin sulu ekstraktının ve hidrojel boncuğunun antioksidan ve antidiyabetik aktivitesinin olduğu tespit edilmiştir. Dolayısıyla hem gilaburu meyve ekstraktının hem de hidrojel boncuklarının fonksiyonel gıda üretiminde ya da gıda takviyesi olarak kullanılabilme potansiyeli mecuttur. Gilaburu (Viburnum opulus L.) is a fruit that contains different nutrients and bioactive components. Chlorogenic acid, L-malic acid, anthocyanins and carotenoids are the most abundant bioactive components of the fruit. Also, gilaburu fruit indicates several health-promoting activities such as antidiabetic, antioxidant and antimicrobial. In this study, it was aimed to optimize the microwave assisted extraction (MAE) of bioactive components like phenolic substances from Gilaburu fruit with the response surface method (RSM) and Box-Behnken experimental design. The optimum extraction factors for gilaburu fruit phenolic were determined as temperature 60 (°C), time (5 minutes) and solid/solvent ratio (4.18 g/100 mL) based on the maximum extraction yield (%) and total phenolic content (mg GAE/g extract). The water extract of the fruit that produced based on optimum parameters determined with RSM design used to anlaysed phenolic composition, free radical and antidiabetic activities. In addition, this phenolic-rich aqueous extract were encapsulated using ionic gelation and chitosan sytem. Also, the moisture content and encapsulation efficency of these hydrogel beads were determined as 53.52% and 91.26% respectively. Same experiments were analysed for both extract and hyrdrogel beads. It was obtained chlorogenic acid was major compound of both extract and hydrogel beads, 34.18 mg/g ekstrakt and 0.51 mg/g ekstrakt (56.45 g hydrogel beads), respectively with HPLC. The inhibition activity of α-glucosidase in the presence of extract was detected as 54.9% for glucose. On the other hand hydrogel beads inhibited α-glucosidase activity at a rate of 99.9%. It could be concluded that, gilaburu fruit phenolics can be extracted in shorter time with low energy needs and maximum efficiency using MAE. Furthermore, it was discovered that aqueous extract of gilaburu fruit and its hyrogel beads exhibited antioxidant and antidiabetic acitivities. Consequently, both gilaburu fruit and its hydrogel beads can be considered as potential sources of invaluable compounds for development of functional food industry or food supplement.
