Bu araştırmada, farklı oranlarda diyet lifi (inülin veya buğday lifi) ve probiyotik bakterilerin (Bifidobacterium animalis subsp. lactis BB-12 ve Lactobacillus acidophilus LA-5) eklenmesi ile az yağlı ve yağsız fonksiyonel Tulum peynirleri üretilmiştir. Olgunlaşma süresince (90 gün) diyet liflerin, probiyotik bakterilerin canlılığına ve Tulum peynirlerinin bazı özelliklerine etkisinin incelenmesi amaçlanmıştır. Farklı oranlarda (%1 ve %2) inülin ilavesi ve olgunlaşma zamanı, az yağlı ve yağsız probiyotik Tulum peynirlerinin her iki grubunda da yağ ve toplam azot oranları, randıman, olgunlaşma derecesi, eriyebilirlik, β-kazein, αs1-kazein ve diğer parçalanma ürünleri değerlerini önemli düzeyde etkilemiştir (p<0.05). İnülin ilavesiyle, az yağlı ve yağsız Tulum peynirlerinin özellikleri olumlu yönde gelişmiştir. Ayrıca olgunlaşma süresince β-kazein ve αs1-kazein konsantrasyonları azalırken, gama-kazein ve diğer parçalanma ürünlerinde artış meydana gelmiştir. Tekstür profil analizi sonucunda inülin ilavesinin ve olgunlaşma süresinin, az yağlı örneklerde dış yapışkanlık, iç yapışkanlık, çiğnenebilirlik, elastiklik ve yağsız örneklerde sertlik, iç yapışkanlık, çiğnenebilirlik, elastiklik değerleri üzerine istatiksel olarak etkisinin önemli olduğu tespit edilmiştir (p<0.05). Farklı oranlarda inülin ilavesi probiyotik bakterilerin gelişimini de teşvik etmiştir (p<0.05). Lactobacillus acidophilus LA-5'in olgunlaşma sonunda Bifidobacterium animalis subsp. lactis BB-12'den daha fazla canlı kaldığı bulunmuştur. İnülin içeren tüm peynirlerin mikroyapılarının daha düzgün ve pürüzsüz bir protein matrisine sahip olduğu saptanmıştır. İnülin içeren az yağlı Tulum peynirleri, tekstür ve organoleptik özellikleri ile daha kabul edilebilir bulunmuştur. Bu araştırma sonucunda inülinin, az yağlı ve yağsız probiyotik Tulum peynirlerinde yağ ikame maddesi olarak kullanılabileceği tespit edilmiştir. Her iki gruptan az yağlı probiyotik Tulum peynirinin üretici açısından istenen kalitede ürün elde edilmesini sağlayacağı saptanmıştır. Bununla birlikte %1 ve %2 inülin ilave edilen az yağlı peynirlerde genel kabul edilebilirlik puanları birbirine benzer bulunmuştur. İnülinin %1 oranında ilavesinin probiyotik bakteri gelişimini daha fazla teşvik ettiği görülmüştür. Bu nedenle sağlığa faydası ve istenen tekstür ve aromayı sağlaması bakımından tüketici talebini karşılayabilecek az yağlı probiyotik Tulum peyniri üretiminde %1 oranında inülin kullanılmasının daha uygun olacağı düşünülmektedir. Az yağlı ve yağsız probiyotik Tulum peynirlerine farklı oranlarda (%0.5 ve %2) buğday lifi ilave edilmesinin ve olgunlaşma süresinin etkisi, pH, titrasyon asitliği, yağ, kül, tuz, su aktivitesi, toplam azot oranları ile randıman, olgunlaşma derecesi, eriyebilirlik, β-kazein, αs1-kazein ve diğer parçalanma ürünleri değerleri üzerinde istatiksel olarak önemlidir (p<0.05). Buğday lifi, Tulum peynirlerinin bazı özelliklerini olumlu yönde etkilemiştir. Buğday lifi oranı arttıkça, tüm örneklerin eriyebilirlikleri belirgin bir şekilde azalmıştır. Her iki grupta da buğday lifi oranı arttıkça L* ve a* renk değerleri artarken, b* değeri azalmıştır (p<0.05). Buğday lifi, az yağlı ve yağsız peynirlerde probiyotik bakterilerin (Bifidobacterium animalis subsp. lactis BB-12 ve Lactobacillus acidophilus LA-5) gelişimini teşvik etmiştir (p<0.05). Olgunlaşma sonunda sağlığa fayda sağlamak ve terapötik faydalar elde etmek için yeterli canlı probiyotik sayısı, her iki buğday lifi oranında da (%0.5 ve %2) elde edilmiştir. Olgunlaşma döneminde az yağlı ve yağsız peynirlerin ana aroma grubunu hidrokarbonlar, ketonlar ve aldehitler oluşturmuştur. Peynirlerin mikroyapıları, buğday lifi ilavesiyle örneklerde sertliğin artmasını ve dış yapışkanlığın azalmasını desteklemiştir. Bununla birlikte %2 oranında buğday lifi ilavesi yapılan peynirlerde, lifler yüksek oranda şişerek gözle görülür şekilde peynirin yüzeyini kaplamıştır. Bu durum panelistler tarafından %2 buğday lifi içeren örneklerde belirlenen yapı puanlarının olumsuz etkilenmesine neden olmuştur. Bu araştırma ile buğday lifinin, az yağlı ve yağsız probiyotik Tulum peynirlerinin duyusal ve tekstürel özelliklerini olumlu yönde etkileyerek bir yağ ikame maddesi olarak kullanılabileceği saptanmıştır. Ancak %2 buğday lifi ilavesi, örneklerin eriyebilirlik, tekstür, mikroyapı ve duyusal özelliklerine olumsuz etki etmiştir. Araştırma sonucunda fonksiyonel özelliklere sahip Tulum peyniri üretimi için %0.5 oranında buğday lifi ilavesinin daha uygun olacağı belirlenmiştir. In this study, low-fat and non-fat functional Tulum cheeses were produced by adding different percentages of dietary fiber (inulin or wheat fiber) and probiotic bacteria (Bifidobacterium animalis subsp. lactis BB-12 and Lactobacillus acidophilus LA-5). It was aimed to investigate the effects of dietary fibers on the viability of probiotic bacteria and some properties of Tulum cheeses during the ripening period (90 days). The addition of inulin at different rates (1% and 2%) and ripening time significantly affected the fat and total nitrogen ratios, yield, ripening degree, meltability, β-casein, αs1-casein and other degradation products values in both groups of low-fat and non-fat probiotic Tulum cheeses (p<0.05). With the addition of inulin the properties of low-fat and non-fat Tulum cheeses were improved positively. In addition, β-casein and αs1-casein concentrations decreased during ripening, gamma-casein and other degradation products increased. As a result of the texture profile analysis, it was determined that the inulin addition and ripening time had statistically a significant effect on the adhesiveness, cohesiveness, chewiness, resilience in low-fat samples and on the hardness, cohesiveness, chewiness and resilience values in non-fat samples (p<0.05). The addition of inulin at different rates also enhanced the development of probiotic bacteria (p<0.05). Lactobacillus acidophilus LA-5 was found to be more viable than Bifidobacterium animalis subsp. lactis BB12 at the end of ripening. It was determined that the microstructure of all cheeses containing inulin had a more uniform and smooth protein matrix. Low-fat Tulum cheeses containing inulin were found to be more acceptable with their texture and organoleptic properties. As a result of this research, it was determined that inulin can be used as a fat replacer in low-fat and non-fat probiotic Tulum cheeses. It has been determined that low-fat probiotic Tulum cheese from both groups will provide the desired quality product for the producer. However, the general acceptability scores of low-fat cheeses with 1% and 2% inulin are found to be similar to each other. It was observed that the addition of 1% of inulin further encouraged the growth of probiotic bacteria. For this reason, it was thought that it would be more appropriate to use 1% inulin in the production of low-fat probiotic Tulum cheese, which could meet consumer demand in terms of health benefits and providing the desired texture and aroma. The effect of adding wheat fiber at different rates (0.5% and 2%) and ripening time to low-fat and non-fat probiotic Tulum cheeses, pH, titration acidity, fat, ash, salt, water activity, total nitrogen ratios and yield, ripening degree, meltability, β-casein, αs1-casein and other degradation products values were statistically significant (p<0.05). Wheat fiber affected some properties of Tulum cheeses positively. As the wheat fiber ratio increased, the meltability of all samples decreased significantly. As the wheat fiber content increased in both groups, L* and a* color values increased, while b* value decreased (p<0.05). Wheat fiber enhanced the growth of probiotic bacteria (Bifidobacterium animalis subsp. lactis BB-12 and Lactobacillus acidophilus LA-5) in low-fat and non-fat (p<0.05). At the end of ripening, sufficient viable probiotic numbers were obtained in both wheat fiber ratios (0.5% and 2%) to provide health benefits and therapeutic benefits. During the ripening period, hydrocarbons, ketones and aldehydes formed the main aroma group of low-fat and non-fat cheeses. The microstructures of the cheeses supported the increase of hardness and the decrease of adhesiveness in the samples with the addition of wheat fiber. However, in cheeses with 2% wheat fiber addition, the fibers swelled at a high rate and visibly covered the surface of the cheese. This situation caused the structure scores determined by the panelists in samples containing 2% wheat fiber to be negatively affected. With this research, it was determined that wheat fiber could be used as a fat replacer by positively affecting the sensory and textural properties of low-fat and non-fat probiotic Tulum cheeses. However, the addition of 2% wheat fiber adversely affected the meltability, texture, microstructure and sensory properties of samples. As a result of the research, it was determined that 0.5% wheat fiber addition would be more suitable for the production of Tulum cheese, which has functional properties.
