Bu tez çalışmasında, yanık yaralarının tedavisinde kullanım amaçlı nanolifli yara örtüsü geliştirilmiştir. Nanolifli malzemenin üretiminde elektro lif çekim yöntemi kullanılmış olup, iki katmandan oluşan nanolifli yara örtüsünün tasarımı, üretimi ve karakterizasyonu gerçekleştirilmiştir. Geliştirilen nanolifli yara örtüsünün ilk katmanı, yara eksüdasını emmesi ve yaralı bölgeyi nemli tutması beklenen, süper emici özelliğe sahip poliakrilik asit (PAA) esaslı nanoliflerden oluşmaktadır. Yaraya temas eden ikinci katmanın üretiminde ise, esnekliği, yüksek mukavemeti ve hidrofobik yapısından dolayı yaraya yapışmaması gibi özellikleri dikkate alınarak, polikaprolakton (PCL) polimeri kullanılmıştır. Bu katmanda yarayı iyileştirici etken madde olarak protein esaslı yumurta akı ve albümin tercih edilmiştir. Çalışmada öncelikle her iki katman için de iletkenlik, viskozite, yüzey gerilimi ve yoğunluk gibi polimer çözelti özellikleri belirlenmiştir. Daha sonra ise elektro lif çekim proses parametreleri (voltaj, mesafe, çözelti besleme vb.) optimize edilerek nanolifli yüzey üretimi gerçekleştirilmiştir. PAA polimer esaslı nanolifler üretildikten sonra suda çözünürlüğünü önlemek amacıyla ısı ile fiziksel çapraz bağlama işlemi uygulanmıştır. Daha sonra ise çapraz bağlı süper emici PAA esaslı nanolifli yüzey, farklı konsantrasyonlarda (% 0-1-3-5-7-9) yumurta akı veya albümin içeren PCL esaslı nanolifler ile kaplanarak iki katmanlı nanolifli yara örtüsünün üretimi tamamlanmıştır. Karakterizasyon işlemleri kapsamında, SEM, FT-IR, TGA, DSC ve XRD analizleri ile morfolojik, kimyasal ve termal açıdan nanolifler karakterize edilmiştir. Daha sonra, üretilen nanoliflerden etken maddenin salım performansı UV-vis yöntemi ile belirlenmiştir. Ayrıca iki katmanlı nanolifli yüzeylere su buharı geçirgenlik analizi yapılmıştır. Tez çalışmasının son aşamasında, iki katmanlı üretilen nanolifli yüzeylerin, Wistar Albino cinsi sıçanlar üzerinde in-vivo analizleri yapılarak, geliştirilen malzemenin ikinci derece yanık yarası üzerindeki iyileştirici performansı histopatolojik açıdan değerlendirilmiştir. Histopatolojik analizler sonucunda, nanolifli yara örtüsünün ikinci derece yanık yaralarının iyileşmesini hızlandırma potansiyeli olduğu belirlenmiştir. Bu tez kapsamında geliştirilen nanolifli yara örtüsünün, gerek kullanılan polimerler gerekse protein esaslı etken maddeler bakımından çevreye duyarlı, sürdürülebilirlik potansiyeli olan, katma değeri yüksek ve ekonomiye katkı sağlayacak bir biyomedikal malzeme olduğu düşünülmektedir. In this thesis, the nanofiber wound dressing was developed for use in the treatment of burn wounds. The electrospinning method was used in the production of nanofibrous material, and the designing, production and characterization of a two-layer nanofiber wound dressing was carried out. The first layer of the developed nanofiber wound dressing consists of polyacrylic acid (PAA) based nanofibers with super absorbent properties, which are expected to absorb wound exudate and keep the injured area moist. The polycaprolactone (PCL) polymer was used, for production of the second layer, that in contact with the wound, considering its flexibility, high strength and non-adhesion to the wound due to its hydrophobic structure. In this layer, protein-based egg white and albumin were preferred as wound-healing active ingredients. In the study, first of all, the polymer solution properties such as conductivity, viscosity, surface tension and density were determined for both layers. Then, the nanofiber production was carried out by optimizing the electrospinning process parameters (voltage, distance, solution feding, etc.). After the PAA-based nanofibers were produced, physical crosslinking with heat was applied to prevent water solubility. Then, the production of the two-layer nanofibrous dressing was completed by coating the cross-linked super-absorbent PAA-based nanofiber surface with PCL-based nanofibers containing egg whites or albumin at different concentrations (0-1-3-5-7-9%). The nanofibers were characterized morphologically, chemically and thermally by SEM, FT-IR, TGA, DSC and XRD analyzes within the scope of characterization processes. Then, the release performance of the active substance from the produced nanofibers was determined by the UV-vis method. In addition, water vapor permeability analysis was performed on two-layer nanofiber structures. In the last stage of the thesis, in-vivo analyzes of the bilayer produced nanofibers with Wistar Albino rats were made, and the curative performance of the developed material on the second degree burn wound was evaluated histopathologically. As a result of histopathological analysis, it was determined that nanofiber wound dressing has the potential to accelerate the healing of second-degree burn wounds. It is thought that the nanofiber wound dressing developed is a biomedical material that is environmentally friendly in terms of the polymers and protein-based active ingredients, has sustainability potential, has high added value and will contribute to the economy. In this thesis, the nanofiber wound dressing was developed for use in the treatment of burn wounds.
