Bu tez çalışmasında gıda sektöründe ambalaj materyali olarak kullanılan yenilebilir filmlerin, aktinomisetlerden elde edilen transglutaminaz enzimi ile özelliklerinin iyileştirilmesi hedeflenmiştir. Bu amaçla Isparta'nın farklı bölgelerinden, farklı mevsimlerde ve farklı bitki türü yetiştirilen toprakların 5-15 cm derinliklerinden, sonbaharda 30, kış aylarında 10, ilkbaharda 20 ve yaz aylarında 15 olmak üzere toplam 75 farklı toprak örneği alınmıştır. Örneklerden SCA besiyerine ekim yapılarak toplam 265 adet aktinomiset izolatı edilmiştir. Elde edilen 265 adet aktinomiset izolatlatının transglutaminaz enzim aktivitelerinin tespiti amacıyla hazırlanan örnekler, ilk etapta UV Spektrofotometrede, daha sonra floresan spoktrofotometrede analiz edilerek enzim aktivitesi tespit edilmiş; daha sonra yapılan bu testlerden enzim aktivitesi yüksek olma ihtimali olan örnekler Q substratla HPLC cihazında incelenmiştir. İzolatların hiçbirinde transglutaminaz enzim aktivitesine rastlanamamıştır. Tezin ilerleyen aşamasında 2U, 4U ve 6U gibi farklı oranlarda ticari transglutaminaz enzimi kullanılarak sodyum kazeinat bazlı yenilebilir filmler elde edilmiştir. Bu örneklerin polimerizasyon ölçümü yapılmıştır. Yenilebilir filmlerin jel görüntülerinde kazein bantlarında transglutaminaz enzim miktarı artışı ile ters orantılı olarak azalma görülmüş ve transglutaminaz enzimi kullanılan filmlerde molekül ağırlığı daha büyük olan yeni bantlar oluşmuştur. Sodyum kazeinat ve transglutaminaz enzimi kullanılarak üretilen yenilebilir filmlerin gerilme kuvvetleri ölçülmüştür. Transglutaminaz enzimi kullanılmayan filmlerin gerilme kuvveti 8.04 MPa, enzim kullanılan filmlerin gerilme kuvvetleri de enzim miktarı artışı ile orantılı olarak 10.13 MPa (2U), 12.63 MPa (4U) ve 14.81 MPa (6U) olarak tespit edilmiştir. Kopma direnci ölçümlerinde ise kopmada uzama değerleri gerilme kuvveti ile ters orantılı olarak değişiklik göstermiş ve transglutaminaz enzimi kullanılmayan yenilebilir filmlerde %15.09 iken enzim kullanılanlarda sırası ile %13.29 (2U), %12.07 (4U) ve %9.30 (6U) olarak tespit edilmiştir. Hazırlanan örneklerin 14 günlük su buharı geçirgenliği ölçümlerinde transglutaminaz enzimi ilave edilmeden üretilen filmlerde %13.10 kayıp görülürken transglutaminaz enzimi ilave edilmiş örneklerde kayıp %12.78 (2U), %12.11 (4U) ve %11.53 (6U) olarak tespit edilmiştir. Hazırlanan yenilebilir filmlerin sıvı çözeltilerinde viskozite ölçümleri yapılmış ve transglutaminaz enzim miktarı artışı ile çözeltilerin kıvamında artış tespit edilmiştir. Transglutaminaz enzimi kullanılmayan filmlerde viskozite 74.45 cP, enzim kullanılan filmlerde 100.80 cP (2U), 128.40 cP (4U) ve 145.05 cP (6U) olarak tespit edilmiştir. Hava geçirgenliği ölçümlerinde de enzim içeren yenilebilir filmlerin hava geçirgenliğinde belirgin azalma görülmüştür. Ölçüm sonuçlarına göre transglutaminaz enzimi ilave edilmeyen filmlerde hava geçirgenliği 4.05 L/m2 dk olarak ölçülürken sırasıyla 2U, 4U ve 6U transglutaminaz enzimi içeren yenilebilir filmlerde 2.89, 2.15 ve 1.45 L/m2 dk olarak azaldığı tespit edilmiştir. Transglutaminaz enzimi ile elde edilen sodyum kazeinatlı filmlerin taramalı elektron mikroskobundaki görüntüleri de incelenmiş ve enzim kullanılmayan filmlerde yoğun gözenekler bulunurken enzim kullanılan filmlerde enzim miktarının artışı ile gözenek sayılarında azalma olduğu görülmüştür. Atomik kuvvet mikroskobundaki 2 ve 3 boyutlu incelemede taramalı elektron mikroskobu ile elde edilen verileri doğrulayan sonuçlar elde edilmiştir. Enzim kullanılmadan elde edilen filmlerin yüzeyleri pürüzlü iken enzim kullanılan filmlerde enzim miktarının artışı ile film yüzeyindeki pürüzlerin ve gözeneklerin azaldığı görülmüştür. Yenilebilir film örneklerinin elemental analizleri de yapılmış; enzim kullanılmayan ve enzim kullanılan örnekler arasında belirgin bir fark gözlenmemiştir. İncelemeler sonucunda 6U transglutaminaz enzimi kullanılarak elde edilen yenilebilir filmlerin özelliklerinin diğer örneklere göre daha verimli olduğu tespit edilmiştir. In this thesis study, it is aimed to improve the properties of edible films used as packaging material in the food industry by transglutaminase enzyme obtained from Actinomycetes. For this purpose, a total of 75 different soil samples, 30 in autumn, 10 in winter, 20 in spring and 15 in summer, were taken from 5-15 cm depths of soils grown in different seasons and different plant species from different regions of Isparta. A total of 265 actinomycete isolates were isolated from the samples by inoculation into SCA medium. The samples prepared for the purpose of determining the transglutaminase enzyme activities of the 265 actinomycete isolates obtained were analyzed firstly in a UV Spectrophotometer, then in a fluorescent spectrophotometer, and the enzyme activity was determined; samples with a high probability of enzyme activity from these tests were examined in HPLC device with Q substrate. Transglutaminase enzyme activity was not found in any of the isolates. In the further stage of the thesis, sodium caseinate based edible films were obtained by using commercial transglutaminase enzyme in different ratios such as 2U, 4U and 6U. The polymerization measurement of these samples was made. In the gel images of edible films, a decrease was observed in casein bands inversely proportional to the increase in the amount of transglutaminase enzyme and in films using transglutaminase enzyme, new bands with higher molecular weight were formed. The tensile strength of edible films produced using sodium caseinate and transglutaminase enzyme were measured. The tensile strength of the films without transglutaminase enzyme was determined as 8.04 MPa, and the tensile strength of the films with enzyme was determined as 10.13 MPa (2U), 12.63 MPa (4U) and 14.81 MPa (6U) in proportion to the increase in the amount of enzyme. In the breaking strength measurements, the elongation at break values changed inversely with the tensile strength and while it was 15.09% in edible films without transglutaminase enzyme, it was determined as 13.29% (2U), 12.07% (4U) and 9.30% (6U) in those with enzyme used, respectively. In the 14-day water vapor permeability measurements of the prepared samples, 13.10% loss was observed in the films produced without the addition of transglutaminase enzyme, while the losses were determined as 12.78% (2U), 12.11% (4U) and 11.53% (6U) in the samples with added transglutaminase enzyme. Viscosity measurements were made in edible film solutions and an increase was determined in the consistency of the solutions due to the increasing amount of transglutaminase enzyme. The viscosity was determined as 74.45 cP in the films without transglutaminase enzyme, and as 100.80 cP (2U), 128.40 cP (4U) and 145.05 cP (6U) in the films including enzyme. In air permeability measurements, a significant decrease was observed in the air permeability of the edible films including enzymes. According to the measurement results, while the air permeability of films without transglutaminase enzyme was measured as 4.05 L/m2 min, it was determined that it decreased by 2.89, 2.15 and 1.45 L/m2 min in edible films including 2U, 4U and 6U transglutaminase enzyme, respectively. The scanning electron microscope images of the sodium caseinate films obtained with the transglutaminase enzyme were also examined and while there were dense pores in the films without enzyme, it was observed that the numbers of pores were decreased with the increase in the amount of enzyme in the films with enzyme. Results of the 2 and 3 dimension examination in atomic force microscope were confirming the data obtained with scanning electron microscopy. While the surfaces of the films obtained without the use of enzymes were rough, it was observed that the roughness and pores on the film surface decreased with the increase in the amount of enzyme in the films using the enzyme. Elemental analyzes of edible film samples were also made; no significant difference was observed between the samples without enzyme and with enzyme used. As a result of the review, the properties of edible films obtained using the enzyme 6U Transglutaminase were found to be more efficient than other samples.
