Türkiye kendiliğinden yetişen bitki örtüsü açısından zengin bir çeşitliliğe ve birçok tıbbi ve aromatik bitkiye sahiptir. Rumex türlerinden Rumex acetosella L. ve Rumex crispus L. halk tarafından çeşitli amaçlarla (gıda, tedavi, boya vb.) kullanılmaktadır. Bu bitkiler antrakinon içerik bakımından zengindir ve bu içeriklerinin belirlenmesi ve tayini oldukça önemlidir. Bu çalışmada farklı bölge ve zamanlarda toplanan Rumex acetosella L. ve Rumex crispus L. bitkilerindeki antrakinonların (aloe-emodin, rhein, emodin, chrysophanol ve physcion) eş zamanlı kalitatif ve kantitatif tayini için yüksek ayırma verimliliği, yüksek tespit hassasiyeti ve seçiciliğine sahip bir HPLC-DAD yöntemi geliştirildi ve valide edildi. Geliştirilen HPLC-DAD yönteminde kromatografik ayırma Supelcosil LC-18 (250×4.6mm, 5μm) kolonunda ve gradient program ile gerçekleştirilmiştir. Optimum metot koşulları, farklı mobil faz bileşimleri, mobil faz akış hızı, kolon sıcaklığı, enjeksiyon hacmi ve bileşiklerin dalga boyu incelenerek belirlenmiştir. Mobil faz A ve B olarak %0,5 ortofosforik asit içeren deiyonize su ve metanol kullanılmıştır. Prosedür, 225 nm'de DAD dedektörlü HPLC yöntemi kullanılarak 1,0 mL/dakika akış hızı ve 20 μL enjeksiyon hacmi ile 20 °C'de gerçekleştirilmiştir. Geliştirilen metodun geçerliliğinin kanıtlanması amacıyla validasyon çalışmaları yapılmıştır. Tüm bileşikler için korelasyon katsayısı (r) 0,9991-0,9997 aralığında ve kalibrasyon eğrisi 0,1-1 μg/mL derişim aralığında doğrusal olarak bulunmuştur. Tayin limiti ve kantitatif tayin limitleri sırasıyla, 0,014–0,017 μg/mL ve 0,017–0,057 μg/mL aralığında tespit edilmiştir. RSD'leri %1'den düşük olan antrakinonlar için kabul edilebilir gün içi (%99,167-100,810) ve günler arası (%99,375-100,919) geri kazanımlar elde edilmiştir. Doğruluk çalışmasında bileşiklerin % geri kazanım değerleri %99,215 ile %100,915 aralığında bulunmuştur. Kararlılık parametresinde, bileşiklerin oda sıcaklığında 48 saat bozunmadan stabil kaldığı ve sağlamlık testinde ise metodun güvenilir ve sağlam olduğu gösterilmiştir. Rumex acetosella L. ve Rumex crispus L. bitkilerinde başlıca emodin, chrysophanol ve physcion az miktarda aleo-emodin ve rhein tayin edilmiştir. Geliştirilen ve valide edilen yöntemin, Rumex acetosella L. ve Rumex crispus L. bitkilerinin ekstraktlarında bulunan aloe-emodin, v rhein, emodin, chrysophanol ve physcion kalitatif ve kantitatif olarak belirlenmesi için etkili ve uygun olduğunu göstermiştir. Turkey has a rich diversity in terms of vegetation that grows spontaneously and many medicinal and aromatic plants. Rumex species Rumex acetosella L. and Rumex crispus L. are used by the public for various purposes (food, treatment, dye, etc.). These plants are rich in anthraquinone content and the specification and determination of these contents is very important. In this study, a HPLC-DAD method with high separation efficiency, high detection sensitivity and selectivity was developed and validated for the simultaneous qualitative and quantitative determination of anthraquinones (aloe-emodin, rhein, emodin, chrysophanol ve physcion) in Rumex acetosella L. and Rumex crispus L. plants collected at different regions and times. In the developed HPLC-DAD method, chromatographic separation was performed on a Supelcosil LC-18 column (250×4.6mm, 5μm) and a gradient program. Optimum method conditions were determined by studying different mobile phase compositions, mobile phase flow rates, column temperatures injection volume and wavelength of the compounds. Deionized water containing 0.5% orthophosphoric acid and methanol was used as the mobile phases A and B. The procedure was carried out at 20 °C with the flow rate of 1.0 mL/min and the injection volume of 20 μL utilizing an HPLC method with DAD detector at 225 nm. Validation studies were carried out to prove the validity of the developed method. The correlation coefficient (r) for all compounds was found to be in the range of 0.9991-0.9997 and the calibration curve was linear in the concentration range of 0.1-1 μg/mL. LOD and LOQ values were calculated in the range of 0,014–0,017 μg/mL and 0,017–0,057 μg/mL, respectively. Acceptable intraday (99,167-100,810) and interday (99,375-100,919) recoveries were acquired for anthraquinones with RSDs less than 1%. In the accuracy study, the % recovery values of the compounds were found between 99.215% and 100.915%. In the stability parameter, it was shown that the compounds remained stable for 48 hours at room temperature without decomposition, and the method was reliable and robust in the robustness test. Major emodin, chrysophanol and physcion, and small amounts of aleo-emodin and rhein were determined in Rumex acetosella L. and Rumex crispus L. plants. The developed and vii validated method showed that it is effective and suitable for the qualitative and quantitative determination of aloe-emodin, rhein, emodin, chrysophanol and physcion in extracts of Rumex acetosella L. and Rumex crispus L. plants.
