Bacillus cereus sensu stricto (B. cereus), Bacillaceae familyasına ait toprak kökenli bir bakteri olup özellikle meyve sebzelerde yaygın olarak bulunan, toksin üretme özelliği olan gıda kaynaklı patojen bir mikroorganizmadır. Bu çalışmanın amacı bir akdeniz ülkesi olarak günlük hayatta sıklıkla tükettiğimiz salata yapımında kullanılan yeşil yapraklı sebzelerden (nane, maydanoz, semizotu, dereotu, tere, roka, marul ve ıspanak) muhtemel Bacillus cereus grubu (B. cereus sensu lato) üyesi bakterilerin izolasyonu, izolatların tür düzeyinde moleküler tanılarının yapılması, hemolitik aktivitelerinin belirlenmesi ve toksin genlerinin multipleks Polimeraz Zincir Reaksiyonu (mPCR) ile araştırılmasıdır. Çalışma kapsamında muhtemel B. cereus grubu üyesi bakterilerin izolasyonu için Bacillus Cereus Mannitol Egg Yolk (MYP) Agar ve Bacillus cereus Agar (PEMBA formülasyonu) besiyeri ortamları kullanılmıştır. İzolasyon işlemi için 40 adet yeşil yapraklı sebze örneği incelenmiş olup, örneklerden toplam 59 adet muhtemel B. cereus grubu üyesi bakteri izole edilmiştir. B. cereus grubuna özgül primer çiftleri kullanılarak yapılan PCR denemeleri sonucunda 46 izolatın B. cereus grubu üyesi olduğu belirlenmiştir. B. cereus grubu üyesi B. mycoides, B. cereus, B. anthracis ve B. thurigiensis türlerine ait özgül primer çiftleri kullanılarak yapılan PCR denemesi sonucunda 46 B. cereus grubu izolattan 42'sinin B. cereus ve 4'ünün B. mycoides türü üyesi olduğu belirlenmiştir. Çalışmada kullanılan izolatlardan hiçbiri B. thurigiensis ve B. anthracis'e özgü primer çiftleri ile yapılan PCR denemeleri sonucu herhangi bir ampfilikasyon vermemiştir. Hemolitik aktivite testi sonucu izolatların tamamının (β) hemolitik aktivite gösterdiği belirlenmiştir. Toksin genlerinin tespiti için yapılan mPCR denemeleri sonucunda izolatların 37'sinde (% 80,44) entFM, 27'sinde (% 58,69) nheA, 20'sinde (% 43,57) hblC, 19'unda (% 41,30) cytK, 19'unda (% 41,30) hblA, 16'sında (% 34,78) hblD, 8'inde (% 17,39) nheC, 5'inde (% 10,86) nheB, 4'ünde (% 8,69) ces ve 2'sinde (% 4,34) CER genine rastlanılmıştır. Sonuç olarak izole edilen bu suşların çok sayıda (>106 kob/g) vücuda alınması sonucunda gıda zehirlenmeleri yaşanabileceği dolayısıyla insan sağlığı üzerine olumsuz etkileri olabileceği belirlenmiştir. Bacillus cereus is a soil-borne bacterium belong to the Bacillaceae family and food-borne pathogenic and is a toxin-producing microorganism commonly found in fruits and vegetables. The aim of this study is to isolate possible Bacillus cereus group microorganisms which are used in making salad from green leafy vegetables (mint, parsley, purslane, dill, cress, arugula, lettuce and spinach), which are the green vegetables we consume frequently in our daily life as a Mediterranean country, molecular diagnosis of isolates at the species level, determination of hemolytic activities and investigation of toxin genes by multiplex Polymerase Chain Reaction (mPCR). In this study, Bacillus Cereus Mannitol Egg Yolk (MYP) Agar and Bacillus cereus Agar (PEMBA formulation) medium media were used for the isolation of possible B. cereus group microorganisms. For the isolation process, 40 piece green vegetable samples were examined, and total 59 piece possible B. cereus member microorganisms were isolated from 40 green vegetable samples. As a result of the Polymerase Chain Reaction (PCR) experiment using primer pairs specific to the B. cereus group, it was determined that 46 isolates were members of the B. cereus group. B. cereus group member B. mycoides, B. cereus, B. anthracis and B. thurigiensis species as a result of the PCR experiment performed for species-level diagnosis using specific primer pairs, 42 of the 46 B. cereus group bacteria were found to be B. cereus spp. and 4 of them were determined to be B. mycoides. Species diagnosis PCR experiments with primer pairs specific to B. thuringiensis and B. anthracis used in the study did not yield any amplification. As a result of mPCR experiments for the detection of toxin genes, we found in 37 (80,44 %) isolates were entFM, 27 (58,69 %) were nheA, 20 (43,57 %) were hblC, 19 (41,30 %) were cytK, 19 (41,30 %) were hblA, 16 (34,78 %) were hblD, 8 (17,39 %) were nheC, 5 (10,86 %) were nheB, 4 (8,69 %) ces and 2 (4,34 %) CER genes. Hemolytic activity results were determined to show (β) hemolytic activity. As a result, it has been determined that as a result of ingestion of these isolated strains in large numbers (>106 cfu/g), food poisoning may occur and therefore have negative effects on human health.
