Ips sexdentatus (Boerner, 1767), ülkemizin hemen hemen her yerinde, özellikle çam ve ladin türlerinde yayılış gösteren tahripkar bir kabuk böceğidir. Literatürde çeşitli fungal türler ile simbiyotik ilişkiler içinde olduğu bilinen bu zararlının ülkemizdeki sahip olduğu fungal etkileşimler hakkında sınırlı sayıda bilgi bulunmaktadır. Bu çalışma 2020-2021 yılları arasında, Türkiye'nin Batı Akdeniz Bölgesinde karaçam meşçerelerinde dağılış gösteren I. sexdentatus'un fungal türlerle olan simbiyotik ilişkisini belirlemek amacıyla gerçekleştirilmiştir. Çalışmada 5 farklı örnek alandan 250 adet I. sexdentatus ergin bireyi toplanmış ve toplanan böceklerin yüzey ve vücut içi fungus çeşitliliği moleküler karakterizasyon çalışmaları ile belirlenmiştir. Çalışmanın sonucunda toplam 864 adet saf fungal izolat elde edilmiştir. Elde edilen izolatlar morfolojik olarak gruplandırılmış ve her bir morfolojik gruptan temsilci olarak seçilen izolatlar DNA izolasyonuna ve ardından ITS gen bölgeleri PCR reaksiyonuna tabi tutulmuştur. Moleküler karakterizasyon sonuçlarına göre, 1 izolat alt şube, 1 izolat takım, 16 izolat cins, 33 izolat ise tür düzeyinde karakterize edilmiştir. Çalışma sonucunda; Alternaria alternata, A. chlamydosporigena, A. panax, A. penicillata, A. tenuissima, Akanthomyces attenuatus, Aspergillus carneus, A. cristatus, A. hiratsukae, A. ochraceus, A. unguis, A. versicolor, A. alboluteum, Auxarthron sp., Bjerkandera adusta, Cadophora malorum, Ceriporia sp., Cladosporium dominicanum, C. ramotenellum, C. sphaerospermum, Coprinellus xanthothrix, Cyphellophora europaea, Exophiala oligosperma, Fusarium acuminatum, Gibberella avenacea, Mortierella alpina, Nakazawaea ambrosiae, Oxyporus corticola, Penicillium chrysogenum, P. fluviserpens, P. olsonii, Phaeodactylium sp., P. cucumerina, Sarocladium sp., Talaromyces pinophilus, olmak üzere toplam 35 fungal taksonun I. sexdentatus'taki varlığı bu çalışmada ilk kez kaydedilmiştir. Ayrıca entomopatojenik bir tür olan Beauveria bassiana'nın I. sexdentatus'daki varlığı Türkiye için ilk kayıt niteliği taşımaktadır. Örnek alanların fungal çeşitliliği Shannon Wiener, Simpson, Bray-Curtis indeksleri kullanılarak belirlenmiştir. Shannon-Wiener ve Simpson çeşitlilik indeks verilerine göre, örnek alanların fungal çeşitliliğin (Shannon wiener 3.28-4.49, Simpson 0.95-0.96) yakın olduğu belirlenmiştir. Ips sexdentatus (Boerner, 1767) is a destructive bark beetle that is distributed almost everywhere in Turkiye, especially in pine and spruce species. This pest is known to have symbiotic relationships with various fungal species in the literature, but there is limited information on fungal interactions in Turkiye. This study was carried out between 2020-2021 to determine the symbiotic relationship of I. sexdentatus with fungal species in larch stands in the Western Mediterranean Region of Turkiye. In the study, 250 I. sexdentatus adults were collected from 5 different sample areas and the surface and intracorporeal fungal diversity of the collected insects were determined by molecular characterization studies. As a result of the study, a total of 864 pure fungal isolates were obtained. The isolates were grouped morphologically and representative isolates from each morphological group were subjected to DNA isolation followed by PCR reaction of ITS gene regions. According to the molecular characterization results, 1 isolate was characterized at the subdivision level, 1 isolate at the order level, 16 isolates at the genus level and 33 isolates at the species level. As a result of the study; Alternaria alternata, A. chlamydosporigena, A. panax, A. penicillata, A. tenuissima, Akanthomyces attenuatus, Aspergillus carneus, A. cristatus, A. hiratsukae, A. ochraceus, A. unguis, A. versicolor, A. alboluteum, Auxarthron sp., Bjerkandera adusta, Cadophora malorum, Ceriporia sp, Cladosporium dominicanum, C. ramotenellum, C. sphaerospermum, Coprinellus xanthothrix, Cyphellophora europaea, Exophiala oligosperma, Fusarium acuminatum, Gibberella avenacea, Mortierella alpina, Nakazawaea ambrosiae, Oxyporus corticola, Penicillium chrysogenum, P. fluviserpens, P. olsonii, Phaeodactylium sp, P. cucumerina, Sarocladium sp., Talaromyces pinophilus, the presence of 35 fungal taxa in I. sexdentatus was recorded for the first time in this study.
