Bu tez çalışmasında, kemometrik kalibrasyon yöntemleri (temel bileşen analizi yöntemi (PCA), temel bileşen regresyonu yöntemi (PCR), kısmi en küçük kareler yöntemi (PLS), farmasötik preparattaki Amlodipin ve Atorvastatin'in aynı anda miktar tayinlerine hiçbir ayırma işlemi kullanmaksızın başarıyla uygulanmıştır ve bu yöntemlerle UV Görünür Alan Spektroskopisi yöntemlerinden elde edilen veriler kemometrik olarak değerlendirilmiştir. Belirtilen kemometrik yöntemlerin geçerliliğin belirlenmesinde, Amlodipin ve Atorvastatin'in bileşiklerini içeren 16 adet karışımdan oluşan kalibrasyon (konsantrasyon) seti, hazırlandı. Kalibrasyon setinin 200‐500 nm aralığında absorpsiyon spektrumu kaydedildi. Kalibrasyon seti ve bu sete karşılık 215‐355 nm aralığında elde edilen absorpsiyon verileri arasındaki ilişkiden yararlanılarak üç kemometrik kalibrasyon oluşturuldu. PCA, PCR ve PLS yönteminin geçerliliği Amlodipin ve Atorvastatin içeren sentetik karışımların analiziyle gerçekleştirildi. Uyguladığımız yöntem, Amlodipin ve Atorvastatin için yüzde (%) geri kazanımı ortalaması sentetik karışım için ve ilaç numunesi için hesaplandı. Hesaplanan değerler, oldukça yüksek bulundu. Grup içi ve gruplar arası serbestlik dereceleri yardımıyla F testi yapıldı. F testi sonucuna göre kullandığımız modeli ilaç numunesi karışımına uygulayıp uygulayamayacağımıza karar verildi. FH < FT olduğunda model uygulandı. Geri kazanım değerleri kabul edilebilir boyutta büyük, standart sapmalar da çalışmaya uygun olarak hesaplanmıştır. PLS ve PCR yöntemleri uygulanmış ve bulunan değerlerin doğruluğu F testi ile ölçüldü. Hesaplanan F değerlerinin, teorik değerlerden düşük olduğu görüldü. İkili ilaç maddesini içeren karışımlarda bu maddelerin miktar tayini için PLS ve PCR kalibrasyonlarının kurulmasında çapraz validasyon işleminde tahmin edilen hataların karelerinin toplamının (Predicted Resudiual Error Some of Squares→ PRESS) ve kalibrasyonun standart hatası (SEC) değerlerinin sıfıra yakın olması doğruluğu ve güvenilirliği arttırdı. Kullanılan PLS ve PCR kalibrasyon tekniklerinden ulaşılan sonuçlara ANOVA testi uygulanarak yöntemlerin kontrolü sağlanmıştır. Geliştirilmiş yöntem, tekrar edilebilir, duyarlılığı hassas ve doğru sonuç veren yöntemlerden biri olup Amlodipin ve Atorvastatin içeren ilaç numunelerinin analizinde önerilebilir olduğu tespit edildi. In this thesis, chemometric calibration methods (principal component analysis method (PCA), principal component regression method (PCR), partial least squares method (PLS) were successfully applied to the simultaneous quantification of Amlodipine and Atorvastatin in pharmaceutical preparation without any separation process and the data obtained from UV Visible Field Spectroscopy methods were evaluated chemometrically. To determine the validity of these chemometric methods, a calibration (concentration) set of 16 mixtures containing compounds of Amlodipine and Atorvastatin was prepared. The absorption spectrum of the calibration set in the 200-500 nm range was recorded. Three chemometric calibrations were established using the relationship between the calibration set and the absorption data obtained in the 215-355 nm range. The PCA, PCR and PLS method was validated by analyzing synthetic mixtures containing Amlodipine and Atorvastatin. We calculated the mean percentage (%) recovery for Amlodipine and Atorvastatin for the synthetic mixture and for the drug sample. The calculated values were found to be quite high. F test was performed with the help of degrees of freedom within and between groups. According to the result of the F test, we decided whether we could apply the model we used to the drug sample mixture. The model was applied when FH < FT. The recoveries values were acceptably large and the standard deviations were calculated in accordance with the study. . PLS and PCR methods were applied and the accuracy of the values found was measured by F test. The calculated F values were lower than the theoretical values. In the cross-validation of PLS and PCR calibrations for the quantification of binary drug substances in mixtures containing these substances, the sum of squares of predicted errors (Predicted Residual Error Some of Squares→ PRESS) and standard error of calibration (SEC) values were close to zero, which increased the accuracy and reliability. ANOVA test was applied to the results obtained from the PLS and PCR calibration techniques used to check the methods. The developed method was found to be reproducible, sensitive and accurate and can be recommended for the analysis of drug samples containing Amlodipine and Atorvastatin.
