Çalışmamızda; melatonin uygulanan ratlarda, melatoninin hipokampusta NR 2A ve NR 2B reseptörlerine etkisi araştırıldı. Bunun için 4 farklı rat grubu oluşturuldu. Kontrol (Grup 1), 5mg/kg/gün melatonin (Grup II), 10mg/kg/gün melatonin (Grup III), 20mg/kg/gün melatonin (Grup IV). Ratlara 1 ay süresince her akşam melatonin uygulandı. 1 ay sonunda kan, beyin ve hipokampus örnekleri alınarak, hipokampus NR 2A ve NR 2B reseptörleri ve antioksidan enzim aktiviteleri ölçüldü. Hipokampus GSH-Px düzeylerinin grup IV'de, grup I ve grup II'ye göre istatistiksel olarak anlamlı bir şekilde arttığı görüldü. Hipokampus CAT düzeylerinin grup IV'de, grup I ve grup II'ye göre istatistiksel olarak anlamlı bir şekilde arttığı görüldü. Hipokampusta MDA düzeyleri; grup II, grup III ve grup IV'de, grup I'e göre anlamlı olmayan değişiklik gösterdi. Hipokampus NR 2A ve NR 2B reseptörleri düzeylerinde grup II, grup III ve grup IV'de, grup I'e göre istatistiksel olarak anlamlı artış saptandı. Bu çalışmanın sonuçları; melatoninin hipokampus NR 2A ve NR 2B reseptörlerine olumlu etkisini göstermektedir. In this study we investigated the effect of melatonin on hippocampus NR 2A and NR 2B receptors in rats exposed to melatonin. Rats were divided in to four different groups. These groups are: Control (Group I), 5mg/kg/day melatonin (Group II), 10mg/kg/day melatonin (Group III), 20mg/kg/day melatonin (Group IV). Rats were exposed to melatonin for every evening during 1 month. At the end of 1 month hippocampus NR 2A and NR 2B receptors and antioxidant enzyme levels were assayed in erithrocyte, brain and hippocampus samples. It was observed that the hippocampus GSH-Px levels of group IV is significantly higher than the hippocampus GSH-Px levels of group I and group II. It was observed that the hippocampus CAT levels of group IV is significantly higher than the hippocampus CAT levels of group I and group II. İn hippocampus increased levels of MDA group II, group III and group IV were not significantly different from that of group I. Hippocampus NR 2A and NR 2B receptors levels of group II, group III and group IV were significantly higher than group I. The results of our study suggested the positive effects of melatonin on hippocampus NR 2A and NR 2B receptors.
