Amaç: Ağır metaller özgül ağırlıkları5g cm-3’ten yüksek olan en yaygın kirleticilerdir. Insanların ağırmetallere maruziyeti genellikle ağır metal içeren su ve besinlerin tüketilmesiyolu ile olmaktadır. Içme suyu yolu ile vücuda giren metaller genotoksisite veçeşitli hastalıklara neden olurlar. Bu nedenle insani tüketim amaçlı kullanılansularda ağır metallerin kabul edilebilir limitleri kanun ve yönetmelikler ilebelirlenmiştir. Bizim çalışmamız içme suyunda izin verilen limitlerde metalmiksinin (Cu, Zn, Pb ve Fe) insan aort düz kas hücrelerinde (TG-HA-VSMC)etkilerini moleküler düzeyde araştırmayı amaçlamıştır. Materyal-Metot: İnsan aort düz kashücrelerine içme sularında izin verilen düzey ve 10 kat yüksek konsantrasyondametal miksi 24 ve 48 saat süre ile uygulandı ve RAPD-DNA polimorfizmine ilavetenantioksidan enzimler, ısı şok proteinleri ve apoptoz yolak genlerinin ifadeleribelirlendi. Bulgular: Ağır metal uygulamasının DNApolimorfizminde, apoptotic hücre oranında, antioksidan ve apoptoz yolağı genifadelerinde artışa neden olduğu belirlendi. |

Objective: Heavy metals are the mostcommon pollutants with specific gravity greater than 5 g cm-3. Humanexposure to heavy metals occurs mostly through the consumption of food andwater containing heavy metals. Metals that enter the body via drinking watermay cause genotoxicity and various diseases. Therefore, permissible limits ofheavy metals in water for human consumption have been determined by laws andregulations. Our study aimed to investigate the effects of a mix of metals (Cu,Zn, Pb and Fe) at permissible limits in drinking water on aortic smooth musclecells (TG-HA-VSMC) with at molecular level. Material-Method: A metal mix at thepermissible concentration for drinking water and 10-fold higher concentrationwere exposed to human aortic smooth muscle for 24 and 48 hours, and geneexpression of antioxidant enzymes, heat-shock proteins and apoptosis pathwaywere investigated in addition to RAPD-DNA polymorphism. Results: It was determined that heavymetal exposure caused increase in the DNA polymorphism, apoptotic cell ratiosand enhanced antioxidant and apoptosis pathway gene expression in cells. Conclusion: In conclusion, the mixconsisting of heavy metals at permissible limits in drinking water has beenshown to cause oxidative stress, genotoxicity and apoptosis in human aorticsmooth muscle cells. Considering that apoptosis in vascular smooth muscle cellsplays a role in the pathophysiology of diseases such as arteriosclerosis andhypertension, we believe it would be beneficial to assess these limits throughextensive investigations in further studies. |
