RESVERATROL AND AVOCADO OIL PRETREATMENT ATTENUATES PARACETAMOL-INDUCED LIVER INJURY IN RATS

dc.description <p>Paracetamol (Acetaminophen; APAP) is one of the most widely used analgesic and antipyretic agent today. Paracetamol toxicity due to paracetamol overdose is primarily seen in the liver. In this study, it was aimed to investigate the effects of Resveratrol and Avocado Oil on paracetamol-induced hepatotoxicity in rats. In our study, using a total of 30 male Wistar Albino rats weighing between 400 and 550 grams, 5 separate groups (Group I; Control, Group II; Paracetamol, Group III; Paracetamol + Resveratrol, Group IV; Paracetamol + Avocado oil, Group 5; Paracetamol + Resveratrol + Avocado oil) were created. Only isotonic sodium chloride solution was administered by oral gavage to the rats in the control group. Resveratrol (10 mg/kg) and Avodako oil (200 mg/kg) were administered by oral gavage for 5 days. Paracetamol was administered intraperitoneally at a dose of 600 mg/kg on the 4th day. Animals were sacrificed at 24th hour following paracetamol administration. In our study, Hematoxylin &amp; Eosin staining was performed on liver tissues taken from rats and liver damage was evaluated under light microscope. In the control group (Group I), no findings other than normal histological structures were found in the histological examination of liver tissue sections. When the liver tissues of the experimental group given paracetamol (Group II) were examined, compared to the control group; Granular and vacuolar degenerations in hepatocytes, pycnotic nuclei, mononuclear cell infiltrations in the portal area and parenchyma, hemorrhagic areas, sinusoidal dilatations, vascular congestions, macro and micro vesicular fat deposits were observed (p &lt;0.05). In the group treated with Paracetamol and Resveratrol (Group III), an improvement was observed in histopathological findings compared to the group given Paracetamol (Group II). In the group that received Avocado with Paracetamol (Group IV), an improvement in histopathological findings was observed compared to the group given Paracetamol (Group II), but this improvement was less than the group that received Resveratrol (group III). Paracetamol, Resveratrol and Avocado were given together in the group (Group V) compared to the group (Group II) given paracetamol, an improvement was observed in histopathological findings and this improvement was higher than in group III and group IV. The results indicated that the combined use of avocado oil, which is rich in&nbsp;oleic acid, having antioxidant properties, known as a healthy fat store, and Resveratrol, a powerful antioxidant, exhibits protective effects on high dose paracetamol-induced liver injury compared to their use alone.
