ObjectiveIn recent years, metallic nanoparticles have attractedthe attention in the field of nanomedicine with theirantioxidant, antimicrobial and anticancer activities.Especially, metal nanoparticles produced by greensynthesis method from medicinal plants with knownpharmacological efficacy in cancer treatment havebeen spotlighted by researchers.. Gold nanoparticles;become prominentamong metal nanoparticleswith their unique chemical, physical and non-toxicproperties. In this study, it was aimed to performgreen synthesis of gold nanoparticles using Vitisvinifera (grape seed) aqueous extract and to evaluatecytotoxic, anti-proliferative and apoptotic effectsof gold nanoparticles (Vv-AuNP) alone and withGemcitabine co-administration on HT-29 humancolon cancer cell line.Materials and MethodsGreen synthesis of gold nanoparticles was carried outusing the aqueous extract of Vitis vinifera. Vv-AuNPswere characterized by various analytical techniquessuch as UV-visible spectrophotometer, scanningelectron microscope (SEM) and Energy DispersiveX-ray Spectroscopy (EDX). Vv-AuNPs were treatedwith HT-29 cells at a dose range of 0–80 μg / mL for72 hours, alone and in combination with Gemcitabine(100 μg / mL or 200 μg / mL fixed dose). Cytotoxiceffects were evaluated by MTT assay. In addition,the antiproliferative effects of Vv-AuNPs at increasingdoses (0-400 μg / mL) alone and combined withGemcitabine were determined by the BrdU test. Aftertreatment of gold nanoparticles with HT-29 cells,the level of Caspase-3 activation was analyzed byWestern-blot method.ResultsAccording to MTT results, when the dose of Vv-AuNPs was increased to 80 μg / mL, it was determinedthat HT-29 cells did not show the expected level ofcytotoxic effect. According to the results of the BrdUcell proliferation test, IC50 values of Vv-AuNPs aloneand combined with Gemcitabine were determinedas 147.9 and 39.43 μg / mL, respectively. It wasobserved that the activation of caspase-3 due to Vv-AuNP doses increased compared to control, and itscombined use with Gemcitabine reinforced this effect.ConclusionAs a result, green synthesized AuNPs exhibitedanti-cancer properties on colon cancer cells in vitro.The results of this study show that Vv-AuNPs canbe considered as a potential option for colon cancertreatment. |

AmaçSon yıllarda metalik nanopartiküller antioksidan, antimikrobiyalve antikanser etkinlikleri ile nanotıp alanındaoldukça dikkat çekmektedir. Özellikle, kansertedavisinde farmakolojik etkinliği bilinen tıbbi bitkilerdenyeşil sentez metodu ile üretilen metal nanopartikülleraraştırmacıların ilgi odağı haline gelmiştir. Altınnanopartiküller; kendilerine özgü kimyasal, fiziksel venontoksik özellikleri ile ön plana çıkan metal nanopartiküllerdenbiridir. Çalışmamızda, üzüm (Vitis vinifera)çekirdeği sulu ekstraktı kullanılarak yeşil sentezigerçekleştirilen altın nanopartiküllerin (Vv-AuNP) tekbaşına ve kemoterapötik bir ajan olan Gemsitabin ilekombine kullanımının HT-29 kolon kanseri hücre hattıüzerindeki sitotoksik, anti-proliferatif ve apoptotik etkilerinindeğerlendirilmesi amaçlanmıştır.Gereç ve YöntemAltın nanopartiküllerin yeşil sentezi, Vitis viniferasulu özütü kullanılarak gerçekleştirildi. Vv-AuNP’lerinUV-görünür spektrofotometre, taramalı elektronmikroskobu (SEM) ve Enerji Dağılımlı X-ışını Spektroskopisi(EDX) gibi çeşitli analitik tekniklerle karakterizasyonuyapıldı. Vv-AuNP'ler 0–80 μg/mL dozaralığında tek başına ve Gemsitabin (100 μg/mL veya200 μg/mL sabit doz) ile kombine olacak şekilde HT-29 hücreleri ile 72 saat süresince muamele edildi.Sitotoksik etkileri MTT testi ile değerlendirildi. Ayrıca,HT-29 hücreleri üzerine Vv-AuNP’lerin artan dozlarda(0-400μg/mL) tek başına ve Gemsitabin ile kombineantiproliferatif etkileri ise BrdU testi ile belirlendi. Altınnanopartiküllerin HT-29 hücreleri ile muamelesi sonrasıKaspaz-3 aktivasyon düzeyi Western-blot yöntemiile analiz edildi.BulgularMTT sonuçları değerlendirildiğinde Vv-AuNP’lerin 80μg/mL doza kadar çıkıldığında HT-29 hücrelerindeistenilen düzeyde sitotoksik etkiler göstermediği belirlendi.BrdU hücre proliferasyon testi sonuçlarına dayalıolarak, Vv-AuNP’lerin tek başına ve Gemsitabinile birlikte uygulamalarında IC50 değerleri sırasıyla147.9 ve 39.43 μg/mL olarak belirlendi. Vv-AuNP dozlarınabağlı kaspaz-3 aktivasyonunun kontrole kıyaslaarttığı, Gemsitabin ile kombine kullanımının bu etkiyigüçlendirdiği gözlendi.SonuçSonuç olarak, yeşil sentezlenmiş AuNP’ler in vitro koşullardakolon kanseri hücreleri üzerinde anti-kanserözellikler sergilemiştir. Bu çalışmanın sonuçları, Vv-AuNP'lerinkolon kanseri tedavisi için potansiyel bir seçenekolarak düşünülebileceğini göstermektedir. |
