Dünyada en yaygın genetik hastalıklardan biri olan Beta (B)-talasemi, 11. kromozomun kısa kolunda küme olarak lokalize olan (B-globin genindeki genellikle nokta mutasyonların neden olduğu otozomal resesif bir hastalıktır. Hemolitik mikrositik anemiyle ortaya çıkan (B-talasemi, diğer Akdeniz ülkelerinde olduğu gibi Türkiye'de de en yaygın genetik bozukluklardan biridir. Çalışmaya dahil edilen 64 (B-talasemi majör hastası Süleyman Demirel Üniversitesi Hastanesi Çocuk Hematoloji Polikliniği'ne başvuran hastalardan, 27 (B-talasemi taşıyıcısı Süleyman Demirel Üniversitesi Hastanesi Biyokimya Laboratuarı'na evlilik öncesi talasemi taraması yaptırmak için başvuran kişilerden seçildi. (B-talasemi taşıyıcılarında çalışmaya dahil edilme kriteri olarak HbA2 düzeyinin % 3,5'in üzerinde olması dikkate alındı. Araştırmamıza dahil edilen 91 katılımcıdan, 27'si (% 29,7) heterozigot, 36'sı (% 39,6) homozigot ve 28'nin (% 30,8) birleşik heterozigot formunda mutant allel taşıdığı tespit edildi. Elde ettiğimiz sonuçlara göre, Isparta ve çevresinde en yaygın görülen (B-talasemi mutasyonu IVS1-110 (% 60) olarak bulundu. Bunu IVS2-1 (% 11,61), IVS2-745 (% 7,1), IVS1-6 (% 3,87), kodon8 (% 3,87), IVS1-1 (% 2,58), kodon6 (% 1,94), -30(% 1,94), kodon5 (% 1,94), kodon44 (% 1,94), HbS (% 1,29), kodon39 (% 1,29), kodon30 (% 1,29), mutasyonu takip etti. -87, HbC, kodon8/9, kodon22, IVS1-2, IVS1-5, IVS1-116, IVS1-25, kodon 36i mutasyonları herhangi bir katılımcıda tespit edilmedi. Çalışmamızdaki veriler tanı koyma ve tedavi planlamada klinisyene yardımcı olabilir ve ülkemizin talasemi haritasının belirlenmesinde katkıda bulunabilir. Genetik danışmanlık esnasında moleküler patolojinin bilinmesi sayesinde kişiye açık ve güvenilir bilgiler verilebilir, çocuk sahibi olma kararlarında alınabilecek erken önlemlerle tıbbi abortusların uygulanma gerekliliği ve hasta çocuk doğma riski azaltılabilir. Beta (B)-thalassemia is the most common genetic disease in worldwide due to point mutation on (B-globin gene which is localized on short arm of 11. chromosome as a cluster. It is inherited as autosomal recessive. (B-thalassemia which is presented with hemolytic anemia is the most common genetic disease in Turkey as well as in Mediterranean countries. Sixty four patients with (B-thalassemia carriers which were admitted to Pediatric Hematology outpatient clinic of Suleyman Demirel University and 27 patients with (B-thalassemia carriers which were admitted to Biochemistry Laboratory of Suleyman Demirel University for screening before marriage were included to this study. The inclusion criteria of (3-thalassemia carriers were accepted as HbA2 levels greater than 3,5%. From total 91 participants, 27 (29,7%) were heterozygote, 36 (39,6%) were homozygote and 28 (30,8%) were detected as carrying compound heterozygote form of mutant allele. In our findings, most common (3-thalassemia mutation was IVS1-110(60%) in Isparta and around of it. Other mutations followed it respectively; IVS2-1 (11,61%), IVS2-745 (7,1%), IVS1-6 (3,87%), IVS1-1 (2,58%), codon6 (1,94%), -30 (1,94%), codon5 (1,94%), codon44 (1,94%), HbS (1,29%), codon39 (1,29%), codon30 (1,29%). Mutations of -87, HbC, codon8/9, codon22, IVS1-2, IVS1-5, IVS1-116, IVS1-25, codon 36i were not detected in any participant. Results of this study may be help to the clinicians to diagnose and plan of treatment and may contribute to determine the map of thalassemia in our country. With molecular pathology knowledge during genetic consultancy, clear and reliable information can be given to the individuals and necessity of medical abortus, with early precaution in deciding to have child, and risk of ill child birth may be reduced.
