Alkol bağımlılığı büyük toplumsal etkileri olan kronik, yineleyici bir hastalıktır. Alkolizmin nasıl bir hastalık olduğu, etyolojisinde hangi faktörlerin rol oynadığı ve tedavisi hakkında bilgilerimiz yeterli değildir. Artan bilgiler alkolün nörotrofik faktörler, glutamaterjik ve serotonerjik sistemler ve hücre içi sinyal yolları üzerinde etkisi olduğunu düşündürmektedir. Bu çalışmanın amacı etanol ile alkol bağımlılığı oluşturulmuş ratlarda akamprosatın beyi korteks MDA, GSH-Px, GSH, mikrozomal Ca+2-ATPaz aktivitelerini ve hipokampal NMDAR konsantrasyonunu ölçmektir. Çalışma için 40 erkek Wistar rat alındı ve dört eşit gruba ayrıldı. İlk grup kontrol grubu ve ikinci grup etanol (10 mg/kg/g) verilen gruptu. 3. gruba 200 mg/kg/g den akamprosat, 4. gruba ise 200 mg/kg/g.den akamprosat ve 10 mg/kg/g.den etanol intragastrik olarak 21 gün boyunca verildi. 21. günün sonunda tüm ratlar sakrifiye edildi. Beyin dokusu ve hipokampusları alındı. Sonuçlarımız etanolle oluşturulan alkol bağımlılığı modelinde GSH-Px, Ca+2-ATPaz aktivitesinin ve NMDA subünit konsantrasyonlarının azaldığını, MDA düzeyinin arttığını desteklemektedir. Akomprasat tedavisi GSH-Px, Ca+2-ATPaz aktivitesini ve NMDA subünit konsantrasyonlarının azaltıp MDA düzeylerini arttırırken, akamprosat ve alkolün beraber verilmesi GSH-Px, Ca+2-ATPaz enzim aktivitesini azaltıp, MDA düzeylerini ve NMDA subünit konsantrasyonlarını artırmaktadır. Sonuç olarak, bu çalışmada elde edilen veriler akamprosatın alkol bağımlılığı modelinde NMDA reseptörleri üzerinde bazı olumlu etkilerinin ve antioksidan sistemler üzerinde olumsuz etkilerinin olduğunu göstermiştir. Alcohol addiction is a chronic, relapsing disorder with a huge societal impact. We do not have adequate information on the factors taking part in the etiology of alcoholism, what sort of disease it is and on its treatment. Increasing evidences suggest that alcohol interferes with the action of neurotrophic factors, glutamatergic and serotonergic systems, intracellular signaling pathways. The aim of this study was to investigate the brain cortex MDA, GSH-Px, GSH, microsomes Ca+2-ATPase activities of acamprosate and hippocampal NMDAR consantration in rats induced alcohol dependence by ethanol. 40 male Wistar rats were randomly divided into four equal groups. First group was used as control and second group received 10 mg/kg/d ethanol. 200 mg/ kg/d of acamprosate and 200 mg/ kg/d of acamprosate with 10 mg/kg/d ethanol each day were intragastrically administrated to rats constituting third and forth groups for 21 days, respectively. At the end of 21. day, all rats will be sacrificed and brain tissue and hippocampus were taken. Our results support the that decreased GSH-Px, Ca+2-ATPase activities and NMDA subunit concentrations, increased MDA levels following induction of alcohol dependence with ethanol. Acamprosate treatment decraesed GSH-Px, Ca+2-ATPase activities and NMDA subunit concentrations, increased MDA levels while giving acamprosate together with alcohol caused decrease in the activity of the enzyme GSH-Px, Ca+2-ATPaz; increase MDA levels and NMDA subunit concentrations. In conclusion, the data obtained in this study revealed that in alcohol dependence model acamprosate has some beneficial effects on NMDA receptors although acamprosate induced nagative effects on antioxidant defence systems.
