Epilepsi tam olarak anlaşılmayan biyokimyasal ve moleküler olaylardan kaynaklanan yaygın ve heterojen bir nörolojik hastalıktır. Oksidatif stresin epilepsi patofizyolojisinde önemli rol oynadığı ileri sürülmektedir. Selenyumun (Se) epileptik hastalarda antioksidan savunma mekanizmasını düzenleyerek nöronal hasara karşı koruma sağladığı bildirilmiştir. Topiramatın (TPM) nöroprotektif etkisi birçok çalışmada rapor edilmiş olmasına rağmen muhtemel etki mekanizması tam olarak bilinmemektedir. Bu çalışmanın amacı, TPM ve Se.nin PC12 hücrelerinde, lipid peroksidasyonu, glutatyon (GSH), glutatyon peroksidaz (GSH-Px), sitozolik Ca+2 salınımı ve hücre canlılığı üzerine etkilerini incelemektir. PC12 hücreleri; kontrol, TPM, Se, H2O2, TPM+H2O2, Se+H2O2, Se+TPM ve Se+TPM+H2O2 olmak üzere sekiz gruba ayrıldı. H2O2, TPM and Se.nin doz ve süreleri hücre canlılığını değerlendirmede kullanılan MTT analizine göre belirlendi. Hücreler 10 .M TPM ile 5 saat ve 500 .M Se ile 10 saat inkübe edildi. Ardından hücreler analizden önce 10 saat boyunca 100 .M H2O2 ye maruz bırakıldı. Lipid peroksidasyon düzeyi; kontrol, TPM, Se, Se+TPM, Se+H2O2, TPM+H2O2 ve Se+TPM+H2O2 gruplarında H2O2 grubuna göre anlamlı olarak düşük saptandı (p<0,05). GSH-Px aktivitesi; Se ve Se+TPM gruplarında H2O2 grubuna göre anlamlı olarak yüksek saptandı (p<0,01). GSH seviyesi; kontrol, TPM, Se, Se+TPM, Se+H2O2, TPM+H2O2 ve Se+TPM+H2O2 gruplarında H2O2 grubuna göre anlamlı olarak yüksek saptandı (p<0,05). Sitozolik Ca+2 salınımı, H2O2 ve TPM+H2O2 gruplarında kontrol, TPM, Se, Se+TPM, Se+H2O2, Se+TPM+H2O2 gruplarına göre anlamlı olarak yüksek saptandı (p<0,001). Sitozolik Ca+2 salınımı, Se+H2O2 grubunda kontrol, Se ve Se+TPM gruplarına göre anlamlı olarak yüksek saptandı (p<0,05). Sonuç olarak, bu çalışmada TPM ve Se.nin PC12 hücrelerinde oksidatif stres üzerine koruyucu ekileri olduğu gösterilmektedir. Epilepsy is a common and heterogeneous neurological disorder arising from biochemical and molecular events that are incompletely understood. It has been widely suggested that oxidative stress products play an important role in the pathophysiology of epilepsy. It has been reported that selenium (Se) provides protection against the neuronal damage in patients with epilepsy by restoring the antioxidant defense mechanism. The neuroprotective effects of topiramate (TPM) have been reported in several studies but the putative mechanism of action remains elusive. The aim of this study is to investigate the effects of TPM and Se on lipid peroxidation, glutathione (GSH), glutathione peroxidase (GSH-Px), cytosolic Ca2+ release and cell viability in PC12 cells. PC12 cells were divided into eight groups namely control, TPM, Se, H2O2, TPM+H2O2, Se+H2O2, Se+TPM, Se+TPM+H2O2. The doses and times of H2O2, TPM and Se were determined by MTT assay which is used to evaluate cell viability. Cells were incubated with 10 .M TPM for 5 hours and 500 .M Se for 10 hours. Then, cells were exposed to 100 .M H2O2 for 10 hours before analysis. Lipid peroxidation levels were significantly lower in the control, Se, TPM, TPM+H2O2, Se+H2O2, Se+TPM, Se+TPM+H2O2 groups than in the H2O2 group (p<0,05). GSH-Px activities were significantly higher in the Se and Se+TPM groups than in the H2O2 group (p<0,01). GSH levels were significantly higher in the control, Se, TPM, TPM+H2O2, Se+H2O2, Se+TPM, Se+TPM+H2O2 groups than in the H2O2 group (p<0,05). Cytosolic Ca2+ release was significantly higher in the H2O2 and TPM+H2O2 groups than in the control, Se, TPM, Se+H2O2, Se+TPM, Se+TPM+H2O2 groups (p<0,001). Cytosolic Ca2+ release was significantly higher in the Se+H2O2 group than in the control, Se, Se+TPM (p<0,05). In conclusion, the present study demonstrates that TPM and Se have protective effects on oxidative stress in PC12 cells.
