Amaç: Bu çalışmanın amacı, FoxO1, Anneksin 2 ve p53’ün immünohistokimyasal ekspresyonlarının klinikopatolojik parametrelerle ilişkisini araştırıp, renal hücreli karsinom (RHK) gelişimindeki rollerini belirlemektir.Yöntem: On normal böbrek ve 70 berrak hücreli RHK (BHRHK)'de immünohistokimyasal yöntemle FoxO1, Anneksin 2 ve p53 ekspresyonları incelendi. FoxO1, Anneksin 2 ve p53 ekspresyonları ve klinikopatolojik parametreler arasındaki ilişki yanı sıra bu belirleyiciler arasındaki korelasyon değerlendirildi.Bulgular: Anneksin 2 ve p53 ekspresyonları BHRHK olgularında normal böbrek dokularına göre anlamlı olarak artmıştı (p<0,0001, p=0,014, sırasıyla). Normal böbrek dokuları ile BHRHK arasında FoxO1ekspresyonu açısından anlamlı bir fark yoktu (p> 0,05).Anneksin 2 ve p53 ekspresyonları yüksek nükleer dereceli (p=0,001, p<0,0001, sırasıyla ) ve yüksek pT evreli (p<0,0001, p < 0,0001, sırasıyla) olgularda daha güçlüydü. Bu belirteçler ile lenfovasküler invazyon ve tümör boyutu arasında da anlamlı ilişki bulduk (p<0,05). FoxO1 ekspresyonu düşük nükleer dereceli (p <0,0001) ve düşük pT evreli (p = 0,001) olgularda daha güçlüydü. FoxO1 ekspresyonu ile lenfovasküler invazyon ve tümör boyutu arasında da anlamlı bir ilişki bulduk (p<0,05). Ek olarak, BHRHK olgularında, Anneksin 2 ve p53 ekspresyonları (p< 0,0001, r=0,522) arasında pozitif korelasyon, Anneksin 2 ve FoxO1 ekspresyonları (p<0,0001, r=-0,587) yanısıra FoxO1 ve p53 ekspresyonları (p<0,0001, r=-0,690) arasında da negatif korelasyon bulundu.Sonuç: Bulgularımız FoxO1, Anneksin 2 ve p53 ekspresyonlarının BHRHK'lerin diferansiyasyonu, progresyonu ve agresif davranışında rol oynayabileceğini düşündürmektedir. Bununla birlikte, bulgularımızı doğrulamak ve bu belirteçlerin BHRHK gelişimindeki rolünü belirlemek için daha fazla çalışmaya ihtiyaç bulunmaktadır. Aim: The aim of the present study was to investigate the immunohistochemical expressions of FoxO1, Annexin 2 and p53 in association with the clinicopathological parameters in order to define their roles in the development of renal cell carcinoma (RCC).Method: Immunohistochemical analyse of FoxO1, Annexin 2 and p53 expressions were performed on the sections of 10 normal kidney and 70 clear cell RCC (cRCC) tissues.The association between FoxO1, Annexin 2 and p53 expressions and the clinicopathological parameters as well as, the correlation between these markers were analysed.Results: The expressions of Annexin 2 and p53 were significantly higher in cRCC than the normal kidney tissues (p<0,0001, p=0,014, respectively). There was not any significant difference between cRCC and normal kidney groups regarding the expression of FoxO1 (p>0.05). Annexin 2 and p53 expressions were significantly stronger in cases with higher nuclear grade (p=0,001, p<0,0001 respectively) and pT stage (p<0,0001, p<0,0001 respectively). We also found significant association between these markers and lymphovasculer invasion and tumor size (p<0,05). FoxO1 expression was significantly stronger in cases with lower nuclear grade (p<0,0001) and pT stage (p=0,001). There was a significant association between FoxO1 expression and lymphovasculer invasion and tumor size (p<0,05). In addition, a positive correlation was found between Annexin 2 and p53 expressions (p<0,0001, r=0,522), furthermore a negative correlation was found between Anneksin 2 and FoxO1 (p<0,0001, r=-0,587), as well as FoxO1 and p53 expressions (p<0,0001, r=-0,690) in cRCC cases.Conclusion: According to our results we suggest that FoxO1, Annexin 2 and p53 expressions may involve in differentiation, progression, and the aggressiveness of cRCC. However, further studies are needed to approve our findings and to clarify the role of these markers in the development of cRCC.
