Amaç: Demans, yaşlanan nüfus ile birlikte dünya çapında giderek yaygınlaşmaktadır. Alzheimer Hastalığı (AH) demansın en sık sebebidir. AH'nın etyolojisinde oksidatif stresin önemli bir rol oynadığı bildirilmektedir. AH'nda artan oksidatif stres ile ortaya çıkan serbest oksijen radikalleri, nöronlarda dejenerasyona sebep olmaktadır. Eser element olan selenyumun, antioksidan sistemin bir bileşeni olarak bu dejenerasyonu azalttığı düşünülmektedir. Ancak, selenyumun hangi mekanizma ile bilişsel fonksiyonlar üzerinde olumlu etkileri olduğu tam olarak bilinmemektedir. Bu çalışmada skopolamin uygulamasıyla oluşturulan demans modelinde selenyumun bellek, apopitozis, oksidatif stres, kalsiyum iyon düzeyleri, TRPM2 ve TRPV1 katyon kanalları üzerine etkilerinin ortaya çıkarılması amaçlanmıştır. Materyal metod: Çalışmada Wistar albino cinsi dişi ratlar, kontrol, skopolamin, selenyum ve skopolamin+selenyum olmak üzere 4 gruba ayrıldı. Tüm ratlara 2 hafta boyunca 8 kollu labirentte eğitim verildi ve 15. gün başlangıç bellek kayıtları alındı. Kontrol grubuna 2 hafta intraperitoneal serum fizyolojik (1 mg/kg) uygulandı, 30.gün bellek kayıtları alınarak dekapite edildi. Selenyum grubuna 2 hafta boyunca gün aşırı intraperitoneal selenyum (1,5 mg/kg) uygulandı, 30.gün bellek kayıtları alınarak dekapite edildi. Alzheimer modeli oluşturmak amacıyla skopolamin grubuna 3 hafta boyunca günlük skopolamin (1 mg/kg) intraperitoneal uygulandı, 37. gün bellek kayıtları alınarak dekapite edildi. Skopolamin+selenyum grubu ratlara 3 hafta boyunca skopolamin (1 mg/kg) intraperitoneal uygulandı, 37. gün bellek kayıtları alındı, takiben 2 hafta boyunca gün aşırı intraperitoneal selenyum (1,5 mg/kg) uygulandı ve çalışmanın 52. gününde bellek kayıtları alınarak dekapite edildi. Dekapite edilen ratlarda bellek, apopitozis, oksidatif stres, kalsiyum iyon düzeyleri, TRPM2 ve TRPV1 katyon kanalları üzerine selenyum ve skopolaminin etkileri araştırıldı. Bulgular: Skopolamin uygulanan ratlarda kontrole göre, 8 kollu ışınsal labirent testinde kısa ve uzun süreli bellek hatalarının arttığı, selenyum uygulanan ratlarda kısa ve uzun süreli bellek hatalarının azaldığı bulundu. Skopolamin+selenyum grubunda skopolamin ile oluşan bellek bozukluğunun selenyum uygulanması ile gerilediği görülmüştür. Laboratuvar incelemelerinde skopolamin uygulanan ratlarda lipid peroksidasyonu artarken, GSH düzeyi ve GSH-Px aktivitesinin azaldığı, TRPV1 ve TRPM2 kanalları aracılığıyla hücre içine daha fazla kalsiyum girdiği bu nedenle MTT düzeyinin azaldığı, apopitozis, kaspaz 3-9, ROS, JC-1, prokaspaz 3-9 ve PARP aktivitesinin arttığı bulunmuştur. Selenyum uygulaması, skopolamin uygulamasının neden olduğu bu olumsuz etkileri azaltmıştır. Sonuç: Skopolamin ile oluşturulan demans modelinde selenyum tedavisinin bellek hatalarını anlamlı ölçüde azalttığı bulunmuştur. Ek olarak, selenyum tedavisinin oksidatif parametreleri, TRPV1 ve TRPM2 kanalları aracılığı ile hücre içine giren kalsiyum düzeyini ve nöronal ölümü azalttığı gösterilmiştir. TRPV1 ve TRPM2 kanalları, oksidatif hipokampal hasarın indüklediği demansın tedavisinde önemli bir farmakolojik hedef olarak görünmektedir. Purpose: Dementia is becoming increasingly common around the world with an aging population. Alzheimer's disease (AD) is the most common cause of dementia. It is reported that oxidative stress plays an important role in the etiology of AD. Oxygen free radicals associated with increased oxidative stress in AD, leads to the degeneration of neurons. Trace element selenium, as a component of the antioxidant system is thought to be reducing this degeneration. However, mechanism of this positive effect on cognitive function facilitated by selenium is not known exactly. In this study, in the dementia model created with scopolamine, it is intended to elucidate the effects of selenium on memory, apoptosis, oxidative stress, calcium ion levels, and TRPV1, TRPM2 cation channels. Materials and Methods: Wistar female rats in the study were divided into 4 groups; which were control, scopolamine, scopolamine+selenium and selenium. All rats were trained in the 8-arm maze for 2 weeks and in the 15th day initial memory records were taken. Intraperitoneal saline (1 mg / kg) was administered for two weeks to the control group, in the 30th day memory records were obtained and the rats were decapitated. Selenium (1.5 mg / kg) was administered intraperitoneally every other day for 2 weeks to the selenium group, in the 30th day memory records were obtained and the rats were decapitated. To create a model of Alzheimer's, scopolamine (1 mg / kg) was administered intraperitoneally every day for 3 weeks to scopolamine group, in the 37th day memory records were obtained and the rats were decapitated. Scopolamine (1 mg/kg) was administered intraperitoneally every day for 3 weeks to scopolamine+selenium group, in the 37th day memory records were obtained, then selenium (1.5 mg / kg) was administered intraperitoneally every other day for 2 weeks and in the 52th day of trial, memory records were obtained and the rats were decapitated. In rats which were decapitated, the effects of selenium and scopolamine on memory, apoptosis, oxidative stress, calcium ion levels, TRPV1 and TRPM2 cation channels were investigated. Results: It was determined that in the radial-arm maze test on the scopolamine administered rats, there were more short and long term memory errors; and on the selenium administered rats, there were less short and long term memory errors compared to control. In the scopolamine + selenium group, it has been shown that the memory impairment caused by scopolamine administration was regressed by administering selenium. In the laboratory examinations on the scopolamine-treated rats, it was found that lipid peroxidation increased, GSH levels and GSH-Px activity decreased and more calcium entered the cells via TRPV1 and TRPM2 channels; therefore levels of MTT decreased, apoptosis, caspase 3-9, ROS, JC-1, procaspase 3-9 and PARP activity increased. Selenium application has reduced the negative effects caused by administration of scopolamine. Conclusion: In dementia model induced by scopolamine, treatment of selenium has been found to be significantly reducing memory errors. In addition, selenium administration has been shown to be reducing oxidative parameters, the level of calcium entering the cell via TRPV1 and TRPM2 channels and neuronal death. It seems that TRPM2 and TRPV1 channels may become important pharmacological targets in the treatment of memory impairment induced by oxidative hippocampal injury.
