Sisplatin, en çok kullanılan kemoteröpatik ajanlardan birisidir. Birçok organda toksisiteye neden olmaktadır. Düşük dozlarda verilen sisplatin tedavinin etkinliğini azaltır. Hastalara yüksek oranda verilen kemoterapi ilaçları ise toksisiteye neden olur. Özellikle sisplatinin toksik etkisinin en yüksek oranda görüldüğü doku nefronlardır. Böbreklerde, renal hasara sebebiyet vermekte ve böbrek fonksiyonlarını olumsuz etkilemektedir. Ovaryum dokusunda da toksisite etkisi yüksektir. Özellikle kadınlarda sisplatin kullanılarak yapılan kemoterapi tedavisinde, ovaryumda en sık rastlanan durumların başında folikül rezervlerinin sayısının azalması gelmektedir. Primordiyal foliküllerin, primer foliküllerin ve sekonder foliküllerin sayısının azalması, kadınlarda kısırlığın ortaya çıkmasına neden olmaktadır. Son yıllarda kemoterapötik ajanlar ile antioksidan maddelerin kullanımı da artmıştır. Çalışmalarda sıklıkla sisplatinin neden olduğu toksik etkileri azaltmak adına antioksidan bir maddenin kullanılmasının gerekliliği vurgulanmıştır. Resveratrol ise fenil alaninden sentezlenen doğal bir polifenoldür. Polygonium cuspidatum bitkisinin köklerinde ve üzümün kabuğunda çok yüksek miktarlarda bulunmaktadır. Ovaryum dokusunda resveratrolün etkisi incelendiğinde foliküler atrezi sırasında granüloza hücrelerinde apoptozun indüklediği görülmüştür. Sıçanlarla yapılan bir çalışmada ise ovaryumda resveratrol, oosit sayısını arttırmış, apoptozu engellemiş ve primordiyal folikül ile primer folikül sayısını arttırmıştır. Böylece sisplatin ile yapılan tedavilerde resveratrol gibi antioksidan maddelerin kullanımının, sisplatinin neden olabileceği toksik etkileri azalttığı vurgulanmıştır. Mezenkimal kök hücreler ise mezoderm tabakasından kökenlenen ve multipotent özellikte olan yetişkin kök hücreleridir. Yağ dokusunda ve kemik iliğinde yüksek oranda MKH bulunmaktadır. Son dönemde kök hücrelerin tedavilerde kullanılmaya başlaması ile MKH'lerin kullanımı artmıştır. Çalışmamızda 36 adet dişi sıçan kullanılmış olup her grup 6'şarlı olacak şekilde 6 gruba ayrıldı (n=6). Grup 1 kontrol grubuna deney süresi boyunca intraperitonal (i.p.) olarak resveratrol ile eşit hacimde izotonik salin solüsyonu verildi. Grup 2 resveratrol grubuna 14 gün boyunca 20 mg/kg i.p. olarak verildi. Grup 3 sisplatin grubuna 3. günde 7 mg/kg sisplatin i.p. enjekte edildi. Grup 4 sisplatin ve resveratrol grubuna 3. gün sisplatin 7 mg/kg tek doz olarak i.p. uygulandı. 14 gün boyunca resveratrol 20 mg/kg olarak i.p. verildi. Grup 5 sisplatin ve MKH grubuna sisplatin uygulamasından sonra 7. gün tek doz 3x104 hücre olacak şekilde dokuya i.p. olarak enjekte edildi. Grup 6 sisplatin, resveratrol ve MKH grubuna sisplatin uygulamasından sonra 7. gün tek doz 7. gün tek doz 3x104 hücre olacak şekilde MKH, 14 gün boyunca 20 mg/kg resveratrol i.p. olarak uygulandı. 14. günün sonunda sıçanlar sakrifiye edilerek ovaryum doku örnekleri alındı. Histopatolojik inceleme için hematoksilen-eozin (HE) ile boyamalar yapıldı. Çalışmamızın sonucunda sisplatin ile oluşturulan ovaryum hasarında Grup 1 kontrol grubunda normal bir görünüm izlenmiştir. Grup 2 resveratrol grubunda normale yakın bir görünüm izlenmiştir. Grup 3 sisplatin grubunda ödem, hemoraji ve aynı zamanda atretik foliküller gözlenmiştir. Grup 4 sisplatin ve resveratrol grubunda hemorajik alanlar görülmeye devam etse de atretik foliküllerin sayısında azalma görülmüştür. Grup 5 sisplatin ve MKH grubunda atretik folikül miktarında iyice gerileme gözlenmiştir. Grup 6 sisplatin, resveratrol ve MKH grubunda kontrol grubunda gözlenen normal görünüm görülmüştür.
