Omurilik Yaralanması, yıkıcı ve yaşamı değiştiren bir nörolojik bozukluk olup en ciddi sağlık sorunlarından biridir. Günümüzde akut omurilik yaralanması tedavisi, cerrahi dekompresyonu ve farmakoterapi içerir. Her iki yaklaşımın da omurilik yaralanması hastaları üzerinde olumsuz fizyolojik etkileri olduğu gözlemlenmiştir. Bu nedenle, omurilik yaralanması yönetimi için hücre bazlı tedaviler geliştirmek üzere yeni terapötik hedeflere acilen ihtiyaç vardır. Mezenkimal kök hücrelerin kullanımı, en önemli ve umut verici test edilmiş stratejilerden biridir. Nöroinflamasyon, omurilik yaralanmasının iyileşmesini engelleyen baskın süreçlerden biridir. Resveratrol, antibakteriyel, antioksidan ve antiinflamasyon gibi çeşitli biyolojik özelliklere sahip bir bileşiktir. Resveratrol, nöroinflamatuar faktörlerin azaltılmasında güvenli ve etkilidir. Çalışmamızın amacı sıçanlarda omurilik yaralanması tedavisinde mezenkimal kök hücrenin ve resveralrolün etkisini incelemektir. Çalışmamızda 28 sıçan kullanılmış olup 4 gruba ayrıldı (n=7). Bütün gruplara laminektomi sonrası alt torakal bölgeden tahmini T9-T11 arası anevrizma kliple omurilik travması oluşturuldu. Grup Ⅱ'de omurilik travması oluşturulduktan hemen sonra intramusküler yolla sadece sıçan adipoz dokudan izole edilmiş mezenkimal kök hücre verildi. Grup Ⅲ'de omurilik travmasından hemen sonra intraperitonel yolla resveratrol verildi. Grup Ⅳ'de omurilik travmasında hemen sonra intramusküler yolla sıçan adipoz dokudan izole edilmiş mezenkimal kök hücre (MKH) ve intraperitonel yolla resveratrol verildi. Resveratrol (RES) verilen gruplara, travma oluşturulduktan sonraki 5 gün boyunca resveratrol verilmeye devam edildi. 5. gün sonunda tüm sıçanlar sakrifiye edilerek omurilik travması oluşturulan bölgeden doku örnekleri alındı. Her hayvandan alınan omurilik dokularından longutindal ve transvers makraskopi yapılarak histopatolojik inceleme için HE (Hematoksilen-Eozin) ile boyalamalar yapıldı. Çalışmamızda laminektomi sonrası anevrizma kliple hasar oluşturduğumuzda elde ettiğimiz bulgular diğer çalışmalardaki akut faz patolojik değişikliklerle uyumlu gözlenmektedir. Oluşturduğumuz deneysel modelde 5 gün boyunca subakut dönemin başlamasını bekledik. Ancak elde edilen bulgular literatürde subakut dönemdeki histopatolojik bulgularla çok benzerlik taşımamaktaydı. Grup Ⅱ ve grup Ⅲ grubundaki sıçanların histolojik kesitlerinde glial hücrelerdeki sayı artışı dikkat çekiciydi. Anahtar Kelimeler : Mezenkimal kök hücre, Omurilik yaralanması, Resveratrol.
Spinal Cord Injury, as a distruptive and life-changing neurological disorder, is one of the most critical health issues. At present, the administration of acute spinal cord injury comprises surgical decompression and pharmacotherapy. Both the approaches have been observed to possess negative physiological effects on spinal cord injury patients. Thus, original therapeutic aims for the adminastration of spinal cord injury are immediately necessary for growing cell-based therapies. The use of mesenchymal stem cells (MSCs) typifys one of the most substantial and up and coming tested strategies. Neuroinflammation is one of the predominant processes that hamper spinal cord injury recovery. Resveratrol is a compound that has several biological features, such as antibacterial, antioxidation, and antiinflammation. Resveratrol is confidental and effective in the outstanding reduction of neuroinflammatory factors. The aim of our study is to examine the effect of mesenchymal stem cell and resveralrol in the treatment of spinal cord injury in rats. In our study, 28 rats were used and they were divided into 4 groups (n=7). All groups underwent spinal cord trauma with an estimated T9-T11 aneurysm clip from the lower thoracic region after laminectomy. In group Ⅱ, mesenchymal stem cells isolated from rat adipose tissue were administered intramuscularly immediately after spinal cord trauma was created. In group Ⅲ, resveratrol was administered intraperitoneally immediately after spinal cord trauma. In group Ⅳ, mesenchymal stem cell (MSC) isolated from rat adipose tissue and resveratrol intraperitoneally were administered intramuscularly immediately after spinal cord trauma. Resveratrol (RES) given groups continued to be given resveratrol for 5 days after trauma was created. At the end of the 5th day, all rats were sacrificed and tissue samples were taken from the area where spinal cord trauma was created. Longitudinal and transverse macroscopy was performed from the spinal cord tissues taken from each animal, and staining with HE (Hematoxylin-Eosin) was performed for histopathological examination. In our study, the findings we obtained when we inflicted damage with aneurysm clip after laminectomy are observed to be compatible with acute phase pathological changes in other studies. In the experimental model we created, we waited for the subacute period to begin for 5 days. However, the findings were not very similar to the histopathological findings in the subacute period in the literature. The increase in the number of glial cells was remarkable in the histological sections of rats in group Ⅱ and group Ⅲ groups. Keywords : Mesenchymal stem cell, Spinal cord injury, Resveratrol.
