Yüksek Fruktozlu Mısır Şurubu ile Beslenen Genç Sıçanlarda Prefrontal Korteks Hasarı ve D Vitamininin Koruyucu Rolü Günümüzde kullanımı oldukça artan Yüksek Fruktozlu Mısır Şurubu (YFMŞ); çok tatlı olması, gıdaların raf ömrünü uzatması ve ucuz olması gibi birçok nedenden dolayı tercih edilmektedir. Yapılan son araştırmalar D vitamininin antioksidan ve nöroprotektif etkilerini göstermektedir. Çalışmamızda YFMŞ ile beslenen genç sıçanlarda PFK ilişkili kolinerjik aktivitenin bozulmasına bağlı hafıza ve öğrenmeyi zayıflatan olumsuz etkilerin ve D vitamininin koruyucu rolünün araştırılması amaçlanmıştır. Wistar albino türü erkek 6-8 haftalık sıçanlar, her grupta 8 adet olmak üzere rastgele 3 gruba ayrıldı; kontrol, YFMŞ ve YFMŞ+D Vit. Kontrol grubundaki sıçanlar bir ay boyunca serum fizyolik ile gavaj stresine tabi tutuldu. YFMŞ grubundaki sıçanlara bir ay boyunca YFMŞ-55 (%11) içme sularına eklendi ve serum fizyolik ile gavaj stresine tabi tutuldu. YFMŞ+D Vit grubundaki sıçanlara YFMŞ-55 (%11) ve 42 µg/kg/gün D vitamini gavaj yoluyla verildi. Sıçanların sıvı tüketimleri günlük olarak, ağırlık kazanımı ve kan glikoz ölçümü haftalık olarak yapıldı. Sıçanlara son 9 günde Elevated Plus Maze (EPM), Forced Swimming Test (FST) ve Morris Water Maze (MWM) davranış testleri uygulandı. Sıçanlar anestezi altında sakrifiye edilerek beyin dokusu çıkartıldı. Biyokimyasal olarak oksidatif stres belirteçlerinden Malondialdehit (MDA), Katalaz (CAT) enzim aktivitesi, histopatolojik analizler, Kolin asetiltransferaz (ChAT) ve Beyin Kaynaklı Nörotrofik Faktör (BDNF) ekspresyonları immunohistokimyasal olarak incelendi. YFMŞ grubunda sıvı tüketiminin ve kan glikozu düzeylerinin 2.haftadan itibaren anlamlı olarak arttığı saptandı (sırasıyla p=0,001, p=0,001, p=0,001, p=0,001). EPM'de kapalı kolda daha fazla zaman geçirildiği, FST'de immobilite sürelerinin arttığı ve MWM'de hedef kadranda geçirilen zamanın azaldığı tespit edildi (sırasıyla p=0,010, p=0,002, p=0,002). MDA seviyesinin arttığı ve CAT enzim aktivitesinin azaldığı bulundu (sırasıyla p=0,003, p=0,009). Ayrıca PFK'de dejeneratif nöron sayısında ve ChAT ekspresyonunda artış; BDNF ekspresyonunda azalma görüldü. YFMŞ+D Vit grubunda sıvı tüketiminin arttığı, kan glikozu düzeylerinin 2. ve 4.haftalarda anlamlı olarak azaldığı saptandı (sırasıyla p=0,001, p=0,002, p=0,001). EPM'de kapalı kolda geçirilen sürenin azaldığı, FST'de immobilite sürelerinin azaldığı, MWM'de hedef kadranda geçirilen sürenin arttığı tespit edildi (sırasıyla p=0,047, p=0,006, p=0,002). Sonuçta YFMŞ'nin PFK'de dejeneratif nöron sayısında artışa sebep olarak kolinerjik aktiviteyi bozduğu ve öğrenme-bellek fonksiyonlarını olumsuz etkilediği görüldü. D vitamininin ise dejeneratif nöron sayısını azaltarak kolinerjik aktiviteyi arttırdığı ve öğrenme-bellek fonksiyonlarını olumlu etkilediği ortaya kondu. Prefrontal Cortex Injury and Protective Role of Vitamin D in Young Rats Fed High Fructose Corn Syrup High Fructose Corn Syrup (HFCS), which is widely used today; It is preferred for many reasons such as being very sweet, extending the shelf life of foods and being cheap. Recent studies show the antioxidant and neuroprotective effects of vitamin D. In our study, it was aimed to investigate the negative effects of weakening memory and learning due to the deterioration of PFC-related cholinergic activity in young rats fed with HFCS and the protective role of vitamin D. Wistar albino male 6-8 weeks old rats were randomly divided into 3 groups, 8 in each group; control, YFŞ and HFCS+D Vit. The rats in the control group were subjected to gavage stress with serum physiol for one month. HFCS-55 (11%) was added to the drinking water of rats in the YFMS group for one month and subjected to gavage stress with serum physiolic. HFCS-55 (11%) and 42 µg/kg/day vitamin D were given by gavage to rats in the HFCS+Vit group. Fluid consumption of rats was done daily, weight gain and blood glucose measurement were done weekly. Elevated Plus Maze (EPM), Forced Swimming Test (FST) and Morris Water Maze (MWM) behavioral tests were applied to rats in the last 9 days. The rats were sacrificed under anesthesia and the brain tissue was removed. Biochemically, oxidative stress markers Malondialdehyde (MDA), Catalase (CAT) enzyme activity, histopathological analysis, Choline acetyltransferase (ChAT) and Brain Derived Neurotrophic Factor (BDNF) expressions were examined immunohistochemically. Fluid consumption and blood glucose levels were found to increase significantly in the HFCS group from the 2nd week (p=0.001, p=0.001, p=0.001, p=0.001, respectively). It was determined that more time was spent in the closed arm in EPM, the duration of immobility increased in FST, and the time spent in the target quadrant was decreased in MWM (p=0.010, p=0.002, p=0.002, respectively). It was found that MDA level increased and CAT enzyme activity decreased (p=0.003, p=0.009, respectively). In addition, an increase in the number of degenerative neurons and ChAT expression in PFC; BDNF expression was decreased. Fluid consumption increased and blood glucose levels decreased significantly in the 2nd and 4th weeks in the HFCS+D Vit group (p=0.001, p=0.002, p=0.001, respectively). It was determined that the time spent in the closed arm decreased in EPM, the duration of immobility decreased in FST, and the time spent in the target quadrant increased in MWM (p=0.047, p=0.006, p=0.002, respectively).
