Sıçanlarda Lacticaseibacillus casei Kullanımının Yeme Davranışı, Anksiyete ve Öğrenme Üzerine Etkilerinin Araştırılması Mikrobiyota; insanlarla birlikte yaşayan özel türlerin tamamını ifade etmektedir. Bağırsak mikrobiyotası, pek çok faktörden etkilenebilir. Probiyotikler, konak için sağlığa yararlı canlı mikroorganizlardır. Probiyotik kullanımı bağırsak mikrobiyotasını doğrudan etkiler. Postbiyotik ise, probiyotiklerin ürettiği canlı olmayan bakteriyel veya metabolik yan ürünler olarak bilinir. Bağırsak ile beyin arasındaki iletişim ve etkileşim, beyin-bağırsak ekseni olarak adlandırılan çift yönlü bir ağ tarafından gerçekleştirilmektedir. Bağırsak mikrobiyotasındaki herhangi bir değişiklik bu çift yönlü ağ vasıtası ile beyinde değişikliklere neden olmaktadır. Dolayısıyla çalışmamızda sıçanlarda Lacticaseibacillus casei (L. casei) probiyotik ve postbiyotiğin yeme davranışı, anksiyete ve öğrenme üzerine etkilerini araştırmayı amaçladık. Sıçanlar; kontrol, probiyotik ve postbiyotik olarak üç gruba ayrıldı. Kontrol grubu sıçanlara 42 gün süresince 0,5 ml/gün musluk suyu gavaj yolla verildi. Probiyotik grubu sıçanlara 42 gün süresince L. casei 10 log kob/ml olarak hazırlanmış kültür takviyesi gavaj yoluyla 0,5 ml/gün (9.6 log kob/sıçan) olarak verildi. Postbiyotik grubu sıçanlara, L. casei 10 log kob/ml olarak hazırlanmış kültüre 121ºC'de 20 dakika ısıl işlem uygulanmasıyla canlı bakteri inaktif edilerek hazırlanan sıvı gavaj yoluyla 0,5 ml/gün olarak 42 gün süresince uygulandı. Probiyotik ve postbiyotik gruplarına gavaj uygulaması devam ederken sıçanlara Elevated Plus Maze (EPM), Novel Object Recognation (NOR), Morris Water Maze (MWM), Forced Swimming Test (FST) davranış testleri yapıldı. Sıçanlar anestezi altında dekapite edilerek beyin dokuları alındı. Malondialdehit (MDA) ve Katalaz (CAT) bakımından biyokimyasal analizler yapıldı. Beyin prefrontal korteks, hipokampüs ve amigdala dokuları histopatolojik olarak incelendi ve Serotonin (5-HT) ekspresyonları immunohistokimyasal olarak incelendi. Yeme davranışının değerlendirilmesinde sıçanların vücut ağırlığındaki artış ve su tükemi takibinde her 3 grupta da anlamlı bir farklılık bulunmadı. Probiyotik takviyesinin uygulandığı gruptaki sıçanların anksiyete ve öğrenmenin değerlendirilmesi için yapılan davranış testlerinde istatistiksel olarak anlamlı farklılıklar bulundu. Probiyotik takviyesinin oksidatif stres belirteci olan MDA seviyelerini düşürdüğü saptandı. 5-HT seviyelerinde ise, gruplar arasında anlamlı bir farklılık saptanmadı. Sonuç olarak, probiyotik L. casei uygulamasının anksiyete ve öğrenme üzerinde olumlu etkilerinin olduğu gözlenirken; bu bakteriden üretilen postbiyotik uygulamasıyla ilgili daha fazla çalışma yapılması gerektiğini düşünmekteyiz. Investigation of the Effects of Lacticaseibacillus casei Use on Eating Behavior, Anxiety and Learning in Rats Microbiota; refers to all of the special species that live with humans. The gut microbiota can be affected by many factors. Probiotics are live microorganisms that are beneficial to the health of the host. The use of probiotics directly affects the intestinal microbiota. Postbiotics, on the other hand, are known as non-living bacterial or metabolic byproducts produced by probiotics. Communication and interaction between the gut and the brain is carried out by a bidirectional network called the brain-gut axis. Any change in the gut microbiota causes changes in the brain through this bidirectional network. Therefore, in our study, we aimed to investigate the effects of Lacticaseibacillus casei (L. casei) probiotic and postbiotic on eating behavior, anxiety and learning in rats. Rats; They were divided into three groups as control, probiotic and postbiotic. Control group rats were given 0.5 ml/day tap water by gavage for 42 days. L. casei culture supplement prepared as 10 log cfu/ml was given to rats in the probiotic group by gavage as 0.5 ml/day (9.6 log cfu/rat) for 42 days. In the postbiotic group, L. casei was applied to the culture prepared as 10 log cfu/ml, at 121ºC for 20 minutes, and the liquid prepared by inactivating the live bacteria was applied by gavage at 0.5 ml/day for 42 days. While gavage was applied to probiotic and postbiotic groups, Elevated Plus Maze (EPM), Novel Object Recognation (NOR), Morris Water Maze (MWM), Forced Swimming Test (FST) behavioral tests were performed on rats. The rats were decapitated under anesthesia and their brain tissues were removed. Biochemical analyzes were performed for malondialdehyde (MDA) and catalase (CAT). Brain prefrontal cortex, hippocampus and amygdala tissues were examined histopathologically and 5-HT expressions were examined immunohistochemically. In the evaluation of eating behavior, no significant difference was found in all 3 groups in the follow-up of the rats' body weight gain and water consumption. Statistically significant differences were found in behavioral tests for the assessment of anxiety and learning in the rats in the probiotic supplementation group. It was determined that probiotic supplementation decreased MDA levels, which is a marker of oxidative stress. There was no significant difference between the groups in 5-HT levels. As a result, it was observed that the application of probiotic L. casei had positive effects on anxiety and learning; We think that more studies should be done on the application of postbiotics produced from this bacterium.
