Enjekte edilebilir doku iskeleleri kemik dokusu mühendisliğinde yaygın olarak kullanılmaktadır. Kusur geometrisinden bağımsız enjekte edildiği yerde kusurları dolduracak osteo iletken ve hasarlı kemik dokusunu iyileştirebilecek hidroksiapatit (HAp) ve silimarin ile birleştirilmiş sodyum aljinat (SA)-jelatin (GE) bazlı jellerle enjekte edilebilir doku iskeleleleri hazırlanmıştır. Deve dikeninden (silybum marianum) çözücü ile ekstraksiyon yöntemi uygulanarak %7,3 verimle %57,5 silimarin içeren toz (ES) elde edilmiştir. Çapraz bağlanmayı ve jelleşmeyi kontrol edebilmek için glukono-d-lakton (GDL) kullanılmıştır. Şişme ve jelleşme süresi göz önünde bulundurularak SA/GDL (hacim/hacim) oranının 5/1 olmasına karar verilmiştir. Bu oran sabit tutularak SA jellerinin optimizasyonu jelleşme süresi ve şişme yüzdesine göre yapılmıştır. Jelleşme süresi ve şişme yüzdesine etki eden GE, GDL, çapraz bağlayıcı ve HAp miktarları (ağırlık/hacim) değiştirilerek optimizasyon yapılmıştır. Optimum jel içeriği (% ağırlık/hacim) SA/GE/HAp:1,5/0,5/0,75 bulunmuştur. Jellerin yüzey morfolojileri SEM ile, kristal yapıları XRD ile karakterize edilmiştir. Jellerin 1 saat jelleşme sonrasında ölçülen pH değerleri 7,10-7,26 aralığında bulunmuştur. Hidrojellerin şişme davranışı 37°C'de 1 mM CaCl2 ilaveli fosfat tampon çözeltisi (PBS) içerisinde 24 saat süresince bekletilerek incelenmiştir. Optimum jelin şişme yüzdesi %27,33 olarak bulunmuştur. Jellerin jelleşme süreleri reometre ile 0,5 Hz sabit frekansta ve 0,05 sabit gerilimde frekans tarama testlerinden elde edilmiştir. Optimum jelin jelleşme süresi 317 s olarak ölçülmüştür. Tüm örneklerin reometre ile açısal frekans tarama testi sonucunda kayıp modüllerinin (G') depolama modüllerinden (G'') büyük olduğu görülmüştür. Örneklerin ASTM-D1621'e göre yapılan sıkıştırma testlerinden elastik modülleri ve akma gerilimleri elde edilmiştir. Jellere HAp eklenmesi, elastik modülü 25.6 MPa'dan 23.44 MPa'ya düşürmüştür. Jelatin ve HAp ilavesi aljinat jelinin elastikiyetini azaltmıştır. Örneklerin elastik ve mekanik olarak dayanıklı olduğu bulunmuştur. Bozunma deneyleri hidrojellerin vücut içerisinde hücrelerin büyümesi için yeterli sürede bozunmadan ve parçalanmadan kalabileceğini göstermiştir. OS ve ES eklenen optimum SA/GE/HAp jeller de silimarin salım deneyleri gerçekleştirilmiştir. Silimarin salım sonuçlarına göre, ilk 6 saatte kümülatif silimarin salımı %30, 21 gün sonunda ise %85 olarak bulunmuştur. ES eklenen optimum SA/GE/HAp jelin toksisitesi laktat dehidrogenaz (LDH) analizi ile sitotoksisitesi de suda çözünen formazan-1 (WST- 1) analizi ile belirlenmiştir. WST-1 ve LDH aktivite testleri hidrojel içerisindeki katkıların hücre metabolizmasını hızlandıracak yönde etki yaptığını ve HAp ve ES ile birleştirilmiş aljinat-jelatin bazlı hidrojelin toksik olmadığını göstermiştir. Injectable scaffolds are widely used in bone tissue engineering. Injectable scaffolds have been prepared with sodium alginate (SA)-gelatin (GE) based gels combined with hydroxyapatite (HAp) and silymarin, which can fill the defects and heal the damaged bone tissue, regardless of the defect geometry. Powder (ES) containing 57.5% silymarin was obtained with a yield of 7.3% by applying solvent extraction method from milk thistle (silybum marianum). Glucono-d-lactone (GDL) was used to control crosslinking and gelation. Considering the swelling and gelling time, the SA/GDL (v/v) ratio was decided to be 5/1. By keeping this ratio constant, optimization of SA gels was made according to gelation time and swelling percentage. Optimization was made by changing the amount of GE, GDL, crosslinker and HAp (weight/volume) that affect the gelation time and swelling percentage. The optimum gel content (% w/v) was found to be SA/GE/HAp: 1.5/0.5/0.75. The surface morphologies of the gels were characterized by SEM and the crystal structures were characterized by XRD. The pH values of the gels measured after 1 hour of gelling were found to be in the range of 7.10-7.26. The swelling behavior of the hydrogels was investigated by incubating in phosphate buffer solution (PBS) with the addition of 1 mM CaCl2 at 37°C for 24 hours. The swelling percentage of the optimum gel was found to be 27.33%. The gelation times of the gels were obtained from frequency scanning tests with a rheometer at a constant frequency of 0.5 Hz and a constant strain of 0.05. The gelation times of the optimum gel was measured as 317 s. As a result of the angular frequency scanning test of all samples with rheometer, it was seen that the loss modules (G') were larger than the storage modules (G''). Elastic moduluses and yield stresses were obtained from the compression tests of the samples according to ASTM-D1621. The HAp addion to gels decreased the elastic modulus from 25.6 MPa to 23.44 MPa. The addition of gelatin and HAp decreased the elasticity of the alginate gel. The samples were found to be elastic and mechanically stable. The effects of adding HAp, ES (or original silymarin (OS)) to the optimum SA/GE hydrogel on swelling percentage, gelling time, pH, mechanical strength and degradation were investigated. Degradation experiments of hydrogels were carried out at 37°C in PBS supplemented with 1 mM CaCl2 for 21 days. Degradation experiments showed that hydrogels can remain undegraded and unbroken within the body long enough for cells to grow. Silymarin release experiments were carried out in optimum SA/GE/HAp gels with OS and ES added. According to the silymarin release results, the cumulative silymarin release was found to be 30% in the first 6 hours and 85% at the end of 21 days. The toxicity of the ES added to optimum SA/GE/HAp gel was determined by lactate dehydrogenase (LDH) analysis and its cytotoxicity was determined by water-soluble formazan-1 (WST-1) analysis.
