Çalışmada, 73 adet geleneksel turşu örneğinden 145 laktik asit bakterisi izole edilmiş ve seçilen izolatların turşu üretimlerinde kullanılmak üzere endüstriyel özellikleri ( laktik asit üretimleri, yüksek tuz ve düşük pH ortamlarında gelişimleri, biyojen amin üretim özellikleri) belirlenmiştir. Testler sonucunda starter kültür olarak seçilen izolatlar, 16S rDNA dizi analizleri sonucunda Lactiplantibacillus plantarum AK 87 (L. plantarum AK 87), Lactiplantibacillus plantarum AK 91 (L. plantarum AK 91), Levilactobacillus brevis AK 72 (L. brevis AK 72), Enterococcus casseliflavus AK 114 (E. casseliflavus AK 114) olarak tanımlanmıştır. Seçilen kültürler, liyofilize edilmiş ve bunlar, kornişon turşusu ve karışık turşu üretiminde kullanılmıştır. Turşu üretimlerinde, kontrol grupları haricinde starter kültürlerin bireysel grupları ve karışık kültür grupları ile kontrollü fermantasyonlar gerçekleştirilmiştir. Kornişon turşusu üretimleri, starter kültürlerin inokülasyon oranı ve salamura içeriklerine göre üç farklı deneme şeklinde, karışık turşu üretimi ise tek deneme planında gerçekleştirilmiştir. Oda sıcaklığında gerçekleştirilen denemelerde fermantasyonu tamamlanan turşular, +4 ͦC'de 6 ay süreyle depolanmıştır. Fermantasyon ve depolama sürecinde alınan örneklerde kimyasal ve mikrobiyolojik analizler yapılmış ve sonuçlar, kontrol gruplarıyla karşılaştırılmıştır. Ayrıca örneklerde biyojen amin analizi, aroma profili ve organik asit analizleri ile sertlik ve renk analizleri yapılmış, turşu yüzeyleri taramalı elektron mikroskobunda görüntülenmiştir. Çalışma sonunda fermantasyon süresince starter kültür kullanılan turşularda, kontrol gruplarına göre daha yüksek asit üretimi ve daha düşük pH değerlerine ulaşılmıştır. Kornişon turşusu üretimlerinde starter kültür gruplarında %0,92 laktik asit ile 3,2 pH değerleri tespit edilirken, karışık turşu üretimlerinde %1,03 laktik asit ile 3,01 pH değerleri tespit edilmiştir. Starter kültür gruplarından L. plantarum suşlarının ve karışık kültür kullanımının daha etkili olduğu belirlenmiştir. Tüm üretimlerde starter kültürlerin kullanımları ile enterobakterler kısa sürede inhibe edilmiştir. Starter kültürlerin bireysel olarak 106 kob/ml kullanıldıklarında maya ve küfler üzerinde inhibe edici etkileri belirlenirken, 104 kob/ml inokülasyon oranlarında maya ve küfler üzerinde etkili olmamışlardır. Karışık kültürlerin kullanımında ise tüm üretimlerde kısa sürede maya ve küfler inhibe edilmiştir. Starter kültürlerin fermantasyon sürecindeki etkileri depolama boyunca da devam etmiş, böylece depolamada bozulma yapabilecek olası mikroorganizma gruplarının oluşturabileceği sorunlar engellenmiştir. Yapılan duyusal analizlerde kültür kullanılarak üretilen turşular, kontrol örneklerine göre daha çok beğenilmiştir. Turşu salamuralarındaki biyojen amin analizi sonuçlarına göre, starter kültür kullanılarak üretilen turşulardaki biyojen amin miktarlarının toksik limitlerin altında olduğu tespit edilmiştir. Çalışmada turşu salamuralarında organik asit miktarının ve çeşitliliğinin arttırılmasında starter kültür kullanımının etkili olduğu, starter kültürlerin ürettiği aroma maddelerinin turşu kalitesine ve duyusal özelliklerine de etkili olduğu belirlenmiştir. Fermantasyonu tamamlanan turşuların yüzeyinde çok sayıda laktik asit bakterilerinin var olduğu taramalı elektron mikroskobuyla gösterilmiştir. Renk ve sertlik analizleri sonucuna göre, turşuların duyusal olarak kabul edilir özellikte olduğu, depolama sürecinde ise salamuraya sitrik asit ve glikoz kullanılan turşularda yumuşama ve renkte sararmaların olduğu belirlenmiştir. Endüstriyel turşu üretiminde kullanılmak üzere liyofilize edilen kültürlerin bir yıl süresince canlılık oranları belirlenmiş ve bu süre içerisinde canlılıklarını fazla kaybetmeden korudukları tespit edilmiştir. Starter kültürlerin liyofilize formlarının turşu üretimlerinde kullanıldığı bu çalışmadan elde edilen veriler, starter kültür kullanımının turşu fermantasyonunun gidişatı ve kalitesi üzerinde olumlu etkilerinin olduğunu göstermekle birlikte, starter kültürlerin liyofilize fromlarının uzun süre canlılıklarını sürdürmeleri, turşu üretimlerinde kullanımının pratik ve etkili olduğunu göstermektedir. In the study, 145 isolates of lactic acid bacteria were selected from 73 traditional pickle samples. Industrial properties (lactic acid production, growth in high salt and low pH environments, biogenic amine production characteristics) of these isolates to be used in pickle production were determined. After 16S rDNA sequence analysis of selected isolates were identified as Lactiplantibacillus plantarum AK 87 (L. plantarum AK 87), Lactiplantibacillus plantarum AK 91 (L. plantarum AK 91), Levilactobacillus brevis AK 72 (L. brevis AK 72), Enterococcus casseliflavus AK 114 (E. casseliflavus AK 114). These cultures were lyophilized and further used in the production of pickled gherkins and mixed pickles. In pickle production, controlled fermentations were carried out with individual groups of starter cultures and mixed culture groups. The production of pickled gherkins was carried out in three different production trials depending on the inoculation rate of the starter cultures and the brine contents, while mixed pickles was carried out in a single production. The fermentation process was carried out at room temperature and after completion of fermentation stage pickles were stored at +4 ͦC for 6 months. Chemical and microbiological analyzes were performed during fermentation and storage steps and the results were compared with the control groups. In addition, biogenic amine, aroma profiles and organic acid analyzes along with the hardness and color analyzes were done. Finally the surface of the pickles were visualized under scanning electron microscopy. At the end of the study, higher acid production and lower pH values were achieved in the pickles produced by addition of starter cultures in fermentation process compared to the control groups. While 0.92% lactic acid and 3.2 pH values were determined in starter culture groups in pickled gherkins, 1.03% lactic acid and 3.01 pH values were determined in mixed pickles. It was also determined that the use of L. plantarum strains and mixed cultures for fermentation were more effective. Enterobacteriaceae were inhibited in a short time with the use of starter cultures in all productions. While the inhibitory effects of starter cultures were determined on yeast and molds when they were used individually at 106 cfu/ml, they were not effective on yeast and molds at 104 cfu/ml inoculation rates. In the use of mixed cultures, yeast and molds were inhibited in a short time in all productions. The antimicrobial effects of the starter cultures used in the fermentation process continued throughout the storage period, thus preventing possible problems caused by microorganisms which can cause deteriorations during storage. In the sensory analysis, the pickles produced by using the culture were more appreciated than the control samples. According to the results of biogenic amine analysis in pickle brines, the amounts of biogenic amines in pickles produced using starter cultures were below toxic limits. It was also determined that using starter cultures was effective in increasing the amount and variety of organic acids in pickle brines, and the aroma substances produced by starter cultures were also effective on pickle quality and sensory properties. The presence of a large number of lactic acid bacteria on the surface of the pickles was demonstrated by scanning electron microscopy. According to the results of the color and hardness analysis, it was determined that the pickles were acceptable regarding their sensory quality. The pickles produced by addition of citric acid and glucose in brine softened and turned to yellow during storage. The viability rates of the cultures lyophilized for use in industrial pickle production were determined for one year and it was determined that they preserved their vitality without losing much during this period. The study showed that lyophilized forms of starter cultures can be used in pickle production, showing that the use of starter cultures has positive effects on the course and quality of pickle fermentation. Prolonged term survival of lyophilized cultures showed that their use in pickle production is practical and effective.
