Vakumlu emdirme, meyve-sebzelerin gözenekli yapısına kontrollü miktarlarda çözelti emdirilmesine izin veren yeni tekniklerden biridir. Bu teknik meyve-sebze ürünlerinin probiyotikle zenginleştirilmesi gibi, farklı türde maddeler içeren birçok fonksiyonel gıdanın geliştirilmesinde kullanılmaktadır. Bu tezin amacı, vakumlu emdirme tekniği ile probiyotik bir kültür olarak su kefiri ve probiyotik bir suşun (Lacticaseibacillus casei ATCC 393) bir gıda matrisi olarak elma (Golden delicious) ve muz (Musa spp.) içine emdirilmesi ve kurutulması ile fonksiyonel yeni bir ürün elde edilmesidir. Vakum işlemi için deneysel bir vakumlu emdirme düzeneği hazırlanmıştır. Emdirme matrisi olarak küp elma parçaları ve muz dilimleri, probiyotik kültür olarak L. casei suşu ve su kefiri kullanılmıştır. L. casei liyofilize kültürü uygun besiyeri ortamında aktifleştirilmiş, tampon çözeltiyle emdirme sıvısı olarak kullanılmıştır. Normal daldırma ve vakumlu emdirme yöntemleri ile elma örneklerine L. casei emdirme işlemleri için kullanılan emdirme sıvısında Laktik asit bakterileri (LAB) toplam canlı sayısı 9,48±0,97 log kob/g olarak belirlenmiştir. LC emdirme işlemi sonrası ND yöntemi uygulanan yaş elma örneklerinde 7,89±0,16 log kob/g, VE yöntemi uygulanan yaş elma örneklerinde ise 9,32±0,44 log kob/g, toplam canlı LAB sayısı elde edilmiştir. LC emdirme işlemi uygulanmış yaş elma örneklerinde yapılan LAB toplam canlı sayımına göre, VE yöntemi ile ND yöntemine göre elmalarda 1 log'dan fazla (1,43 log) canlı bakteri varlığı tespit edilmiştir. Konvektif kurutucuda 40 °C'de 10 saat süreyle %,7,61-11,17 aralığında nem içeriğine kadar kurutulan elma örneklerinde toplam laktik asit bakteri sayısı, LCD-K örneğinde 7,08 ±0,7 log kob/g, LCV-K örneğinde 9,05 ± 0,65 log kob/g olarak bulunmuştur. Liyofilize kurutma sonrası ise toplam laktik asit bakteri sayısı LCD-L ve LCV-L örneklerinde sırasıyla; 7,75 ± 0,69 log kob/g ile 7,96 ± 0,89 log kob/g tespit edilmiştir. Her iki kurutma işleminden sonra da VE yöntemi uygulanan örneklerde ND örneklerine kıyasla daha yüksek sayıda canlı laktik asit bakteri varlığı tespit edilmiştir. Konvektif kurutma ile liyofilize kurutmaya göre daha yüksek sayıda bakteri varlığı gözlenmiştir. Elma örneklerinde emdirme sıvısı olarak kullanılan su kefiri örneğinin LAB toplam canlı sayısı, 7,82±1,82 log kob/g; maya sayısı 7,04±0,38 log kob/g olarak belirlenmiştir. ND ve VE yöntemleriyle su kefiri emdirme işlemi uygulanmış SKD-Y ve SKV-Y örneklerinde (yaş elma örneklerinde) sırasıyla LAB toplam canlı sayısı, 6,51±2,13 ve 6,87±2,29 log kob/g; maya sayısı, 6,11±0,41 ve 6,49±0,18 log kob/g olarak tespit edilmiştir. Elma örneklerine alternatif bir ürün olarak çalışılan muz örneklerinde ND ve VE yöntemleriyle su kefiri emdirme işlemi uygulanmış, emdirme sonrası örnekler konvektif kurutucuda 40 °C'de 10 saat, %8,55±0,78 - %11,62±1,3 aralığında nem içeriğine kadar kurutulmuştur. ND yöntemiyle su kefiri emdirilen yaş muz örneklerinde, emdirme sıvısında bulunan bakteri sayısından 2,53 log kob/g az, VE uygulanan yaş muz örneklerinde ise, 0,97 log kob/g daha az sayıda canlı bakteri bulunduğu tespit edilmiştir. Konvektif kurutma sonrası canlı bakteri sayısı, SKD-K örneğinde; 6,37±0,86, SKV-K örneğinde; 6,57±0,27 log kob/g olarak bulunmuştur. Maya sayıları ise yaş örneklerde sırasıyla; 5,21±0,37log kob/g, 5,78±0,13 log kob/g, kuru örneklerde; 5,93±0,86 log kob/g, 6,57±0,27 log kob/g olarak tespit edilmiştir. Elma örneklerinde yapılan renk analizi sonuçlarına göre, liyofize örneklerde konvektif örneklerde göre daha yüksek L ve b değerleri, daha düşük a değerleri ölçülmüştür. Hem su kefiri hem de L. casei emdirilen örneklerde en açık renkli örnek K-L örneği, en koyu renkli olan ise vakumlu emdirme-konvektif kurutma örneği olmuştur. Muz örneklerinde yapılan renk ölçümünde de sonuçlar benzer şekilde en açık renkli kontrol örneği, en koyu örnek ise vakumlu emdirme yapılan örnek olmuştur. Elma örneklerinde yapılan antioksidan kapasite ve toplam fenolik madde analiz sonuçlarına göre konvektif kurutma örneklerinin liyofilize örneklere göre, su kefiri emdirilen örneklerin de L. casei emdirilen örneklere göre daha yüksek değerlere sahip olduğu bulunmuştur. Muz örneklerinde yapılan analizlerde ise hem antioksidan kapasite hem de toplam fenolik madde analiz sonuçlarına göre en yüksek değer SKD-K örneğinde bulunmuştur. Duyusal değerlendirme sonuçlarına göre çalışmamızla geliştirmiş olduğumuz su kefiri emdirilmiş elma ve muz örneklerinin panelistler tarafından yüksek kabule sahip olduğu değerlendirilmiştir. Su kefiri emdirilen kuru elma örneklerinde yapılan SEM (Taramalı Elektron Mikroskopisi) analizinden elde edilen elektron mikrografları, su kefirinde bulunan laktik asit bakterileri ve mayaların elma dokusunda immobilize edildiğini göstermiştir. Elma dokularında hücreler arası boşlukların boyutunun, 8.277-9,040 µm aralığındaki hücrelerin geçmesine izin verecek kadar büyük olduğu görülmüştür. L. casei emdirilen kuru elma örneklerinde yapılan SEM (Taramalı Elektron Mikroskopisi) analizinden elde edilen görüntülerde 1,229-1,466 µm arasında boyutlara sahip çubuk şeklinde olan çok sayıda LC bakterilerinin elma dokusuna emildiği görülmektedir. Su kefiri emdirilmiş ve konvektif kurutulmuş elma örneklerinde yapılan raf ömrü analizlerinde 6 hafta +4 °C'de depolama sonunda vakumlu emdirme yöntemiyle 4,896 log kob/g, normal daldırma yöntemiyle 3,5 log kob/g gibi yüksek bir canlı bakteri sayısı gözlenmiştir. L. casei emdirilmiş ve konvektif kurutulmuş elma kurularında ise 1 aylık depolama sonucu vakumlu emdirmede 6,15 log kob/g, normal daldırma yöntemiyle elde edilen örneklerde ise 4,30 log kob/g canlı bakteri sayısı korunmuştur. Ayrıca 6 haftalık depolama süresince örneklerin kuru madde, pH, asitlik, nem, su aktivitesi gibi fizikokimyasal özelliklerindeki değişimler 2 haftalık aralıklarla analiz edilerek değerlendirilmiştir. Sonuç olarak bu çalışmada vakumlu emdirme yöntemiyle bakteri içeriği yüksek yeni bir probiyotik ürün başarıyla geliştirilmiştir. Raf ömrü boyunca canlılığın daha uzun süre korunması vakumlu emdirme yönteminin avantajı olarak karşımıza çıkmaktadır. Bu yöntem süt temelli olmayan probiyotik gıdaların geliştirilmesi, gıdaların biyoaktif bileşenlerle zenginleştirilmesi için iyi bir alternatif oluşturmaktadır. Vacuum impregnation is one of the newer techniques that allows controlled amounts of solution to be impregnated into the porous structure of fruits and vegetables. This technique is used in the development of many functional foods containing different types of substances, such as fortifying fruit and vegetable products with probiotics. The aim of this thesis is to obtain a functional new product by impregnating water kefir as a probiotic culture and a probiotic strain (Lacticaseibacillus casei ATCC 393) into a food matrix (apple (Golden delicious) and banana (Musa spp.)) by vacuum impregnation technique and drying. An experimental vacuum impregnation setup was prepared for the vacuum process. Cube apple pieces and banana slices were used as impregnation matrix, L. casei strain and water kefir were used as probiotic culture. The lyophilized culture of L. casei was activated in the appropriate medium and used as the impregnation liquid with buffer solution. The total live LAB (Lactic Acid Bacteria) count in the impregnation fluid used for impregnation of apple samples with L. casei by normal immersion and vacuum impregnation methods was determined as 9.48±0.97 log cfu/g. After LC impregnation, the number of total live LAB was 7.89±0.16 log cfu/g in fresh apple samples applied with ND method, and 9.32±0.44 log cfu/g in fresh apple samples applied VE method. According to the total live LAB count performed on the fresh apple samples that were treated with LC impregnation with the VE method, more than 1 log (1.43 log) of live bacteria was detected in the apples according to the ND method. Total lactic acid bacteria count in apple samples dried in a convective dryer at 40 °C for 10 hours to a moisture content of 7-11%, ıt was found as 7.08 ± 0.7 log cfu/g in the LCD-K sample and 9.05 ± 0.65 log cfu/g in the LCV-K sample. After lyophilized drying, the total number of lactic acid bacteria in LCD-L and LCV-L samples, respectively; 7.75 ± 0.69 log cfu/g and 7.96 ± 0.89 log cfu/g were determined. After both drying processes, a higher number of viable lactic acid bacteria were detected in the VE-treated samples compared to the ND samples. A higher number of bacteria was observed with convective drying than with lyophilized drying. Total lactic acid bacteria count of water kefir sample used as impregnation liquid in apple samples was 7.82±1.82 log cfu/g; yeast count was determined as 7.04±0.38 log cfu/g. Total lactic acid bacteria count was 6.51±2.13 and 6.87±2.29 log cfu/g in SKD-Y and SKV-Y samples (fresh apple samples) impregnated with water kefir by ND and VE methods, respectively; yeast count was determined as 6.11±0.41 and 6.49±0.18 log cfu/g. In banana samples, which were studied as an alternative product to apple samples, water kefir impregnation was applied with ND and VE methods, and after impregnation, the samples were stored in a convective dryer at 40 °C for 10 hours, in the range of 8.55±0.78% -11.62±1.3% dried to moisture content. In wet banana samples impregnated with water kefir with the ND method, there were 2.53 log cfu/g less than the number of bacteria in the impregnation liquid, and 0.97 log cfu/g less viable bacteria in VE applied wet banana samples. The number of viable bacteria after convective drying, in the SKD-K sample; 6.37±0.86, in SKV-C sample; 6.57±0.27 log cfu/g was found. Yeast numbers in wet samples, respectively; 5.21±0.37log cfu/g, 5.78±0.13 log cfu/g, in dry samples; 5.93±0.86 log cfu/g, 6.57±0.27 log cfu/g was determined. According to the results of color analysis in apple samples, higher L and b values and lower a values were measured in lyophyse samples compared to convective samples. In the samples impregnated with both water kefir and L casei, the lightest colored sample was the K-L sample, and the darkest one was the vacuum impregnated-convective drying sample. Similarly, the results in the color measurement of the banana samples were the lightest colored control sample, and the darkest sample was the vacuum impregnated sample. According to the results of antioxidant capacity and total phenolic content analysis of apple samples, it was found that convective drying samples had higher values than lyophilized samples, and samples impregnated with water kefir had higher values than samples impregnated with L. casei. In the analyzes performed on banana samples, the highest value was found in the SKD-K sample, according to both antioxidant capacity and total phenolic content analysis results. According to the sensory evaluation results, the apple and banana samples impregnated with water kefir, which we developed with our study, were considered to have high acceptance by the panelists. Electron micrographs obtained from SEM (Scanning Electron Microscopy) analysis of dried apple samples impregnated with water kefir showed that lactic acid bacteria and yeasts in water kefir were immobilized in apple tissue. The size of the intercellular spaces in apple tissues was found to be large enough to allow cells in the 8.277-9,040 µm range to pass through. In the images obtained from the SEM (Scanning Electron Microscopy) analysis of dried apple samples impregnated with L. casei, it is seen that a large number of rod-shaped LC bacteria with dimensions between 1,229 and 1,466 µm were absorbed into the apple tissue. In the shelf life analyzes of water kefir impregnated and convective dried apple samples, a high viable bacterial count of 4,896 log cfu/g with vacuum impregnation method and 3.5 log cfu/g with normal immersion method was observed after 6 weeks of storage at +4 °C. In L. casei impregnated and convective dried apples, after 1 month of storage, 6.15 log cfu/g viable bacteria were preserved in vacuum impregnation and 4.30 log cfu/g in samples obtained by normal immersion method. In addition, the changes in the physicochemical properties of the samples such as dry matter, pH, acidity, moisture, water activity during the 6-week storage were analyzed and evaluated at 2-week intervals. As a result, in this study, a new probiotic product with high bacterial content was successfully developed by vacuum impregnation method. Longer preservation of vitality during the shelf life is the advantage of the vacuum impregnation method. This method is a good alternative for the development of non-dairy probiotic foods and for the enrichment of foods with bioactive components. In banana samples, which were studied as an alternative product to apple samples, water kefir impregnation was applied with ND and VE methods, and after impregnation, the samples were stored in a convective dryer at 40 °C for 10 hours, in the range of 8.55±0.78% -11.62±1.3% dried to moisture content. In wet banana samples impregnated with water kefir with the ND method, there were 2.53 log cfu/g less than the number of bacteria in the impregnation liquid, and 0.97 log cfu/g less viable bacteria in VE applied wet banana samples. The number of viable bacteria after convective drying, in the SKD-K sample; 6.37±0.86, in SKV-C sample; 6.57±0.27 log cfu/g was found. Yeast numbers in wet samples, respectively; 5.21±0.37log cfu/g, 5.78±0.13 log cfu/g, in dry samples; 5.93±0.86 log cfu/g, 6.57±0.27 log cfu/g was determined. According to the results of color analysis in apple samples, higher L and b values and lower a values were measured in lyophyse samples compared to convective samples. In the samples impregnated with both water kefir and L casei, the lightest colored sample was the K-L sample, and the darkest one was the vacuum impregnated-convective drying sample. Similarly, the results in the color measurement of the banana samples were the lightest colored control sample, and the darkest sample was the vacuum impregnated sample. According to the results of antioxidant capacity and total phenolic content analysis of apple samples, it was found that convective drying samples had higher values than lyophilized samples, and samples impregnated with water kefir had higher values than samples impregnated with L. casei. In the analyzes performed on banana samples, the highest value was found in the SKD-K sample, according to both antioxidant capacity and total phenolic content analysis results. According to the sensory evaluation results, the apple and banana samples impregnated with water kefir, which we developed with our study, were considered to have high acceptance by the panelists. Electron micrographs obtained from SEM (Scanning Electron Microscopy) analysis of dried apple samples impregnated with water kefir showed that lactic acid bacteria and yeasts in water kefir were immobilized in apple tissue. The size of the intercellular spaces in apple tissues was found to be large enough to allow cells in the 8.277-9,040 µm range to pass through. In the images obtained from the SEM (Scanning Electron Microscopy) analysis of dried apple samples impregnated with L. casei, it is seen that a large number of rod-shaped LC bacteria with dimensions between 1,229 and 1,466 µm were absorbed into the apple tissue. In the shelf life analyzes of water kefir impregnated and convective dried apple samples, a high viable bacterial count of 4,896 log cfu/g with vacuum impregnation method and 3.5 log cfu/g with normal immersion method was observed after 6 weeks of storage at +4 °C. In L. casei impregnated and convective dried apples, after 1 month of storage, 6.15 log cfu/g viable bacteria were preserved in vacuum impregnation and 4.30 log cfu/g in samples obtained by normal immersion method. In addition, the changes in the physicochemical properties of the samples such as dry matter, pH, acidity, moisture, water activity during the 6-week storage were analyzed and evaluated at 2-week intervals. As a result, in this study, a new probiotic product with high bacterial content was successfully developed by vacuum impregnation method. Longer preservation of vitality during the shelf life is the advantage of the vacuum impregnation method. This method is a good alternative for the development of non-dairy probiotic foods and for the enrichment of foods with bioactive components.
