Bütirik asit fermentasyonu gerçekleştiren Clostridium spp. depolama süresince peynirde gelişim göstererek istenmeyen tat-koku ile beraber çatlak ve yarıklar meydana getirmektedir. Bu durum geç şişme kusuru olarak bilinmekte ve süt endüstrisinde önemli kayıplara sebep olmaktadır. Bu tez çalışmasında geç şişme kusuruna sebep olan ve önemli maddi kayıplarla sonuç Clostridium spp. gelişiminin in vitro şartlarda ve peynir matriksinde engellenmesi araştırılmıştır. Bu amaçla in vitro şartlarda gerçekleştirilen çalışmalarda nisin, lizozim ve ticari olarak satılan 12 farklı peynirden elde edilen suda çözünür peptitlerin (SÇP) Clostridium türleri (Clostridium sporogenes 73, C. sporogenes 97 ve Clostridium butyricum 99) üzerindeki antimikrobiyal etkileri incelenmiştir. Nisinin C. sporogenes 73, C. sporogenes 97 ve C. butyricum 99 vejetatif hücreleri üzerine minimum inhibisyon konsantrasyonu (MİK) sırasıyla 6.25 mg/L, 12.5 mg/L ve 6.25 mg/L olarak tespit edilmiştir. Bu çalışmada kullanılan tüm Clostridium türlerinin/suşlarının sporlarına karşı nisinin MİK değeri 1.56 mg/L olrak belirlenmiştir. Lizozim ise çalışmamızda kullandığımız 5 mg/mL maksimum konsantrasyonda MİK göstermemiştir. Fakat bu konsantrasyonda C. sporogenes 73, C. sporogenes 97 ve C. butyricum 99 vejetatif hücreleri üzerine sırasıyla %10.1, %11.4 ve %7.65, sporları üzerine ise sırasıyla %4.6, %7.9 ve %3.4 oranında inhibisyon etki göstermiştir. SÇP'ler ise 50 mg/mL konsantrasyonda Clostridium spp. vejetatif hücreleri üzerine %24.4-99.6 arasında inhibisyon etki gösterirken sporlar üzerine ise herhangi bir inhibisyon etki göstermemiştir. Peynir matriksinde Clostridium spp. gelişimini engellemek amacıyla nisin, lizozim ve yardımcı kültürlerin (Lactoplantibacillus plantarum AB6-25 ve L. plantarum Lb9) etkileri incelenmiştir. Bu amaçla yüzey yanıt yöntemi kullanılarak Clostridium inhibisyonunun optimizasyonu araştırılmıştır. L. plantarum oranı (peynir sütüne inokülasyon aşamasında L. plantarum kültürlerinin karışım oranları), nisin ve lizozim bağımsız değişkenleri sırasıyla 25:75%–50:50%–75:25%, 0 mg/L–6 mg/L–12 mg/L ve 0 mg/L–0.1 mg/L – 0.2 mg/L olarak seçilmiştir. Box-Behnken tasarımına göre gerçekleştirilen optimizasyonda 17 farklı peynir üretimi gerçekleştirilmiş ve cevap olarak Clostridium sayısı, pH ve %asitlik değerleri belirlenmiştir. Peynir üretiminde süte C. sporogenes kültürlerinden yaklaşık 4 log EMS/mL oranında inoküle edilmiş ve 45 günlük depolama sonucunda alınan sonuçlara göre optimum koşullar L. plantarum kültür oranı %49.54 : 50.46, nisin 1.762 mg/L ve lizozim 0.2 mg/L olarak hesaplanmıştır. Optimum koşullarda doğrulama üretimi yapılmıştır. Peynirlerin 60 günlük depolama süresi boyunca fizikokimyasal ve mikrobiyolojik özellikleri 15 gün aralıkla takip edilmiştir. Depolama sonunda kontrol örneğine göre geç şişme sorumlusu C. sporogenes sayısı 1.3 log EMS/g daha düşük olduğu belirlenmiş ve ambalajda geç şişme semptomu gözlemlenmemiştir. Butyric acid fermentation performed by Clostridium spp. during storage period leads to the development of unwanted taste and odor along with cracks and crevices in cheese. This is known as late blowing defect (LBD) and causes significant losses in the dairy industry. In this thesis, the inhibition of Clostridium spp., which causes LBD, was investigated in vitro and within the cheese matrix. In studies conducted for this purpose under in vitro conditions the anticlostridial effects of nisin, lysozyme and water-soluble peptides (WSPs) obtained from 12 different commercially available cheeses have been investigated. The Minimum inhibition concentration (MIC) values of nisin of Clostridium sporogenes 73, C sporogenes 97 and Clostridium butyricum 99 vegetative cells have been determined as 6.25 mg/L, 12.5 mg/L and 6.25 mg/L, respectively. The MIC value of nisin aganist spores of all Clostridium species/strains used in our study was determined to be 1.56 mg/L. On the other hand, lysozyme did not show MIC value at the maximum concentration of 5 mg/mL that we used in our study. However, at this concentration, inhibition effect on C. sporogenes 73, C. sporogenes 97 and C. butyricum 99 vegetative cells was 10.1%, 11.4% and 7.65%, respectively, and on spores 4.6%, 7.9% and 3.4%, respectively. While WSPs showed an inhibition effect between 24.4-99.6% on Clostridium spp vegetative cells at 50 mg/mL concentration, but did not show any inhibition effects on the spores. The effects of nisin, lysozyme and co-cultures (Lactoplantibacillus plantarum AB6-25 and L. plantarum Lb9) were investigated in order to prevent Clostridium spp growth in cheese matrix. For this purpose, the surface response method was used to optimization of Clostridium inhibition. The ratios of L. plantarum (mixture ratios of L. plantarum cultures during cheese milk inoculation), as well as the independent variables of nisin and lysozyme were determined as follows: L. plantarum ratios 25:75%- 50:50%- 75:25%, nisin concentrations 0 mg/L-6mg/L-12 mg/L and lysozyme concentrastions 0 mg/L-0.1mg/L-0.2mg/L. In the optimization conducted using the Box-Behnken design, 17 different production cheeses were carried out and response variables the count of Clostridium, pH and % acidity values were determined. In cheese production, C. sporogenes cultures were inoculated into milk at a rate of approximately 4 log MPN/mL and according to the results obtained after 45 days of storage, optimum conditions were calculated as L plantarum culture ratio 49.54 : 50.46%, nisin 1.762 and lysozyme 0.2 mg/L. A verification production was carried out under optimum conditions. The physicochemical and microbiological properties of the cheeses were monitored at intervals of 15-day during the 60-day storage period. At the end of storage, the number of C. sporogenes responsible for LBD was determined to be 1.3 log MPN/g lower than the control sample, and no late blowing symptoms were in the packaging.
