Doğal kefir danesi, kendine özgü olağanüstü bir mikrobiotaya sahiptir. Bu yapı polisakkarit bir matriks içinde laktik asit bakterilerini (LAB), asetik asit bakterilerini ve mayaları belli oranlarda bulundurmaktadır. Gerçek kefir, kefir daneleri kullanılarak geleneksel yöntemlerle üretilmekte endüstride ise çoğunlukla kefir daneleri yerine starter kültürler kullanılmaktadır. Kullanılan bu starter kültürler kefir danesi mikrobiotasından çok farklı olup sadece bazı probiyotik mikroorganizmaları içeren fermente bir içecektir. Projenin amacı; doğal kefir danesinden ve starter kefir kültürü kullanılarak üretilen kefirlerin immün sistemi üzerine etkilerinin in vivo çalışmayla belirlenmesidir. Projede farklı üretilen kefirler Balb/c farelerine (6-8 haftalık, 20-25 gram, erkek) iki hafta gavaj yoluyla 300 µl/gün uygulanmıştır. Sakrifikasyon işleminden sonra doğal kefir danesi kullanılarak üretilen kefir ile beslenen farelerin (KGI grubu), starter kültür kullanılarak üretilen kefir ile beslenen farelerin (STI grubu) ve fosfat tamponlu salin (PBS) ile beslenen kontrol grubu farelerinin (CNI) bağırsak dokuları alınmıştır. Daha sonra intestinal sıvı örneklerinde İmmünoglobulin (Ig) A, İmmüoglubulin G, İnterlökin(IL)-4, İnterlökin-10, İnterlökin-12, Toll Like Reseptör (TLR) -4 analizleri yapılarak immünolojik değerlendirilmiştir. Sakrifikasyon sonrası fare bağırsak doku örnekleri homojenize edilerek -80 °C'de depolanmıştır. CNI, KGI ve STI grubunun IgA değerleri sırasıyla 60,87; 72,78 ve 55,31 ng/mL; IgG değerleri 26,59; 38,90 ve 29,44 ng/mL; IL-4 değerleri sırasıyla 84; 40,28 ve 53,28 pg/mL; IL-10 değerleri sırasıyla 110,98; 175,91 ve 134,77 pg/mL; IL-12 değerleri 53,90; 22,93 ve 24,75 pg/mL; TLR-4 değerleri 0,53; 0,43 ve 1,37 ng/mL olarak tespit edilmiştir. Probiyotik özelliği yüksek olan dane kefirinin birçok immünolojik mekanizmayı modüle etme kapasitesine sahip olduğu belirlenmiştir. Natural kefir grain has an unique microbiota. This structure contains lactic acid bacteria (LAB), acetic acid bacteria and yeast in certain ratios in a polysaccharide matrix. Original kefir is produced using traditional method by using kefir grains, whereas starter cultures are used instead of kefir grains in the industry. These starter cultures used are very different from kefir grain microbiota which is fermented drink containing some probiotic microorganisms. The aim of this project; determination and comparison of the effects on the immune system of kefir produced by using natural kefir grain and starter kefir culture. In the project, kefir produced with different cultures were applied to Balb/c mice (6-8 weeks, 20-25 grams, male) by gavage for two weeks at 300 μl /day. Intestinal tissues of mice fed with phosphate buffered saline (PBS) control group (CNI), mice fed with kefir produced using starter culture (STI group) and mice fed with kefir produced using kefir grains (KGI group) were taken after sacrification treatment. Immunoglobulin (Ig) A, Immuoglubulin G, Interleukin (IL)-4, Interleukin-10, Interleukin-12, Toll Like Receptor (TLR)-4 were analyzed immunologically in intestinal fluid samples. After sacrification, mice intestinal tissue samples were homogenized and stored at -80°C. The IgA values of the CNI, KGI and STI groups were respectively 60,87; 72,78 and 55,31 ng/mL; IgG values 26,59; 38,90 and 29,44 ng/mL; IL-4 values of 84; 40,28 and 53,28 pg/mL; IL-10 values 110,98; 175,91 and 134,77 pg/mL; IL-12 values 53,90; 22,93 ve 24,75 pg/mL; TLR-4 values 0,53; 0,43 and 1,37 ng/mL were determined.
