<p>Objectives: The aim of the present study was to examine the effects of N-acetylcys-teine and/or folic acid on fatty liver in obese rats fed on a high-fat diet (HFD).</p><p>Background:The aim of the present study was to demonstrate the potential protective effect of folic acid on the liver due to its involvement in cellular metabolism. A secondary aim was to develop a new approach by comparing folic acid to a frequently used agent with varying results.</p><p>Methods: We used thirty-five Wistar Albino rats divided into four groups in the present study. The groups were as follows: FAG: HFD + folic acid, NASG: HFD + N-acetyl-</p><p>cysteine, NFG: HFD + N-acetylcysteine + folic acid, PCG: only HFD. We fed the rats for twelve weeks to obesity, followed by four weeks of experimentation.</p><p>Results: The present study has demonstrated that mechanisms related to folic acid increasing phospholipid/lipid permeability in membranes may lead to an increase in total cholesterol values. We have also found that folic acid repairs DNA fractures caused by oxidative stress and that N-acetylcysteine administration partially improves DNA fractures. Folic acid protects the liver from oxidative stress by decreasing malondialdehyde while increasing superoxide dismutase values and corrected SOD values of NASG to statistical significance. NFG rats recovered from fatty liver according to histochemical staining in the iNOS results. </p><p>Conclusion: The results suggest that folic acid may be an effective agent in preventing damage formation in fatty liver.&nbsp;</p><p>Keywords: Obesity, fatty liver, N-Acetylcysteine, folic acid</p>
